    Comparison of temporary or continuous immersion systems for micropropagation in liquid medium of axillary shoots of Salix viminalis.

    In this study the response to culture in liquid medium of axillary shoots of two woody species, chestnut and willow, has been compared. Both species differ in their recalcitrance to micropropagation, especially in their rooting and acclimatization abilities. Parameters such as explant type, supporting material, bioreactor type, number of immersions and/or aerations were tested. Continuous and temporary immersion systems have been applied to both species and several differences were detected. Hyperhydricity was not observed in willow shoots cultured in liquid medium with aeration, but this disorder affected chestnut cultures unless supporting materials as rockwool cubes were used. Besides, willow shoots rooted spontaneously in liquid culture, whereas chestnut shoots required an auxin treatment. In spite of these differences, both species showed better performance in liquid culture than in semi-solid medium, and shoots from an adult willow genotype and from eight selected chestnut genotypes (hybrids of European and Asian chestnuts) were successfully rooted and acclimatized.Peer reviewe

    Norepinephrine to increase blood pressure in endotoxaemic pigs is associated with improved hepatic mitochondrial respiration

    ABSTRACT: INTRODUCTION: Low blood pressure, inadequate tissue oxygen delivery and mitochondrial dysfunction have all been implicated in the development of sepsis-induced organ failure. This study evaluated the effect on liver mitochondrial function of using norepinephrine to increase blood pressure in experimental sepsis. METHODS: Thirteen anaesthetized pigs received endotoxin (Escherichia coli lipopolysaccharide B0111:B4; 0.4 mug/kg per hour) and were subsequently randomly assigned to norepinephrine treatment or placebo for 10 hours. Norepinephrine dose was adjusted at 2-hour intervals to achieve 15 mmHg increases in mean arterial blood pressure up to 95 mmHg. Systemic (thermodilution) and hepatosplanchnic (ultrasound Doppler) blood flow were measured at each step. At the end of the experiment, hepatic mitochondrial oxygen consumption (high-resolution respirometry) and citrate synthase activity (spectrophotometry) were assessed. RESULTS: Mean arterial pressure (mmHg) increased only in norepinephrine-treated animals (from 73 [median; range 69 to 81] to 63 [60 to 68] in controls [P = 0.09] and from 83 [69 to 93] to 96 [86 to 108] in norepinephrine-treated animals [P = 0.019]). Cardiac index and systemic oxygen delivery (DO2) increased in both groups, but significantly more in the norepinephrine group (P < 0.03 for both). Cardiac index (ml/min per.kg) increased from 99 (range: 72 to 112) to 117 (110 to 232) in controls (P = 0.002), and from 107 (84 to 132) to 161 (147 to 340) in norepinephrine-treated animals (P = 0.001). DO2 (ml/min per.kg) increased from 13 (range: 11 to 15) to 16 (15 to 24) in controls (P = 0.028), and from 16 (12 to 19) to 29 (25 to 52) in norepinephrine-treated animals (P = 0.018). Systemic oxygen consumption (systemic VO2) increased in both groups (P < 0.05), whereas hepatosplanchnic flows, DO2 and VO2 remained stable. The hepatic lactate extraction ratio decreased in both groups (P = 0.05). Liver mitochondria complex I-dependent and II-dependent respiratory control ratios were increased in the norepinephrine group (complex I: 3.5 [range: 2.1 to 5.7] in controls versus 5.8 [4.8 to 6.4] in norepinephrine-treated animals [P = 0.015]; complex II: 3.1 [2.3 to 3.8] in controls versus 3.7 [3.3 to 4.6] in norepinephrine-treated animals [P = 0.09]). No differences were observed in citrate synthase activity. CONCLUSION: Norepinephrine treatment during endotoxaemia does not increase hepatosplanchnic flow, oxygen delivery or consumption, and does not improve the hepatic lactate extraction ratio. However, norepinephrine increases the liver mitochondria complex I-dependent and II-dependent respiratory control ratios. This effect was probably mediated by a direct effect of norepinephrine on liver cells

    Does Ionized Magnesium Offer a Different Perspective Exploring the Association between Magnesemia and Targeted Cardiovascular Risk Factors?

    Evidence of the association of magnesium (Mg) with arterial stiffness has so far been conflicting. The interplay between hypertension and elevated body mass index (BMI), with hypomagnesemia, instead, has been described in the literature in a more consistent way. Our study aims at revisiting the correlations between blood Mg levels and hemodynamic and body composition parameters in the general population, exploring the sensitivity profile of ionized Mg (Ion-Mg) compared to total Mg (Tot-Mg). We collected data from 755 subjects randomly chosen from a Swiss population previously described and stratified our sample into four equivalent classes according to ionized (whole blood) and total (serum) magnesium. After correcting for age, statistically significant differences emerged between: (i) Tot-Mg ≤ 0.70 and 0.81 ≤ Tot-Mg ≤ 0.90 for cf-PWV (p = 0.039); (ii) Tot-Mg ≤ 0.70 and Tot-Mg ≥ 0.91 for o-PWV (p = 0.046). We also found a statistically significant difference among groups of Ion-Mg values for the 24 h extremes of systolic blood pressure (p = 0.048) and among groups of Tot-Mg for BMI (p = 0.050). Females showed significantly lower levels of total magnesium (p = 0.035) and ionized magnesium (p &lt; 0.001) than males. The overall agreement between magnesium analysis methods was 64% (95%CI: 60.8−67.7%). Our results confirm that Ion-Mg compared with Tot-Mg offers a different profile in detecting both correlations with hemodynamic and body composition parameters and dysmagnesemias. Lower levels of magnesium were associated with worse arterial aging parameters, larger 24 h blood pressure excursions, and higher BMI. Ion-Mg was superior in detecting the correlation with blood pressure only. Considering Ion-Mg as a more specific marker of the magnesium status, and the partially contradictory results of our explorative cross-sectional study, to avoid confounding factors and misinterpretations, ionized magnesium should be used as reference in future studies

    The objective of this paper is to develop a twodimensional numerical model to simulate the response of Scots pine (Pinus sylvestris L.) defect free timber members in order to predict the behaviour of these members when subjected to external forces. For this purpose, data of the mechanical properties of Scots pine were obtained by performing experimental tests on specimens. We determined the stresses and deformations of timber beams in the elastic-plastic and plastic phases. In addition, we developed a finite element software that considered the orthotropic nature of timber, the non-linearity of the compression-reduction branch and the differing moduli of elasticity in tension and compression for Scots pine beams free from defects. The software developed simulates an experimental four point bending test according to UNE-EN 408 Standard.El objetivo de este trabajo es el desarrollo de un modelo numérico bidimensional de piezas de madera de Pinus sylvestris L. libre de defectos que prediga su comportamiento frente a solicitaciones externas. Para su desarrollo, fue necesario realizar ensayos experimentales sobre probetas de pequeño tamaño con el fin de obtener los datos de las propiedades mecánicas para el Pinus sylvestris L. de procedencia española. A partir de los datos experimentales obtenidos, se desarrolla un programa de elementos finitos que considera la ortotropía de la madera, la no linealidad de la rama compresión-acortamiento y los distintos módulos de elasticidad a tracción y a compresión para vigas libres de defectos. El programa simula el ensayo experimental de flexión en cuatro puntos según la Norma UNE-EN 408 y aborda la determinación de las tensiones y deformaciones de las vigas de madera en las tres fases de comportamiento: elástica, elastoplástica y plástica

    The use of liquid silicone for breast augmentation was widespread in the 1960s but was abandoned at the end of the decade due to numerous studies describing the development of a large number of local complications, as well as remote migration of small amounts of silicone. The use of liquid silicone also leads to enormous difficulty in the early diagnosis of breast cancer; these patients are precluded from routine screening programs and must undergo exhaustive periodic examinations. Magnetic resonance imaging has become the most effective test for the early detection of breast cancer in these patients. Indications for subcutaneous mastectomy are the presence of local complications, suspicion of a malignant lesion, or the patient’s desire to prevent both these potential problems
