GC- and UHPLC-MS Profiles as a Tool to Valorizate the Red Alga Asparagopsis armata
Conference Report XVI International Symposium on Marine Natural Products | XI European Conference on Marine Natural Products, 1-5 September 2019, Peniche, Portugal.Asparagopsis armata is considered a biological invader and this red alga is in the last few years one of the worst nightmares for Azores coast biodiversity. So efforts to find an economically valuable application are welcome. In this context biological evaluations of its extracts, such as anti-aging, antioxidant and anticholinesterasic activities, were recently presented [1,2]. Naturally, the knowledge of this species chemical composition is utmost importance not only to find some valuable utilization but also to discovery its mechanisms of defence that can explain its invasive behaviour. In our effort to contribute to this problem solution we establish the GC-MS and UHPLC-MS profiles of both the non-polar and polar extracts. The main compounds in the lipophilic extract were palmitic acid and 1-monopalmitin and brominated compounds dominate both extracts. GC- and UHPLC-MS Profiles as a Tool to Valorize the Red Alga Asparagopsis armata
Asparagopsis armata Harvey is a red alga native from the southern hemisphere and then introduced in the Mediterranean Sea and the Atlantic Ocean, including the Azores Archipelago, where it is considered an invasive alga. Some studies show that the extracts exhibit antimicrobial and antifouling activities, and it is incorporated in some commercialized cosmetic products. (e.g., Ysaline®). However, knowledge of this species chemical composition is scarce. The GC-MS and UHPLC-MS profiles of both the nonpolar and polar extracts were established to contribute to this problem solution. According to the results, A. armata is rich in a great structural variety of halogenated lipophilic and aromatic compounds, some of them identified here for the first time. In the lipophilic extract, 25 compounds are identified, being the halogenated compounds and fatty acids, the two major compound families, corresponding to 54.8% and 35.7% of identified compounds (224 and 147 mg/100 g of dry algae, respectively). The 1,4-dibromobuten-1-ol and the palmitic acid are the two most abundant identified compounds (155 and 83.4 mg/100 g of dry algae, respectively). The polar extract demonstrated the richness of this species in brominated phenolics, from which the cinnamic acid derivatives are predominant. The results obtained herein open new perspectives for valuing the A. armata as a source of halogenated compounds and fatty acids, consequently improving its biotechnological and economic potential. Expression of Calmodulin and Myosin Light Chain Kinase during Larval Settlement of the Barnacle Balanus amphitrite
Barnacles are one of the most common organisms in intertidal areas. Their life cycle includes seven free-swimming larval stages and sessile juvenile and adult stages. The transition from the swimming to the sessile stages, referred to as larval settlement, is crucial for their survivor success and subsequent population distribution. In this study, we focused on the involvement of calmodulin (CaM) and its binding proteins in the larval settlement of the barnacle, Balanus ( = Amphibalanus) amphitrite. The full length of CaM gene was cloned from stage II nauplii of B. amphitrite (referred to as Ba-CaM), encoding 149 amino acid residues that share a high similarity with published CaMs in other organisms. Quantitative real-time PCR showed that Ba-CaM was highly expressed in cyprids, the stage at which swimming larvae are competent to attach and undergo metamorphosis. In situ hybridization revealed that the expressed Ba-CaM gene was localized in compound eyes, posterior ganglion and cement glands, all of which may have essential functions during larval settlement. Larval settlement assays showed that both the CaM inhibitor compound 48/80 and the CaM-dependent myosin light chain kinase (MLCK) inhibitor ML-7 effectively blocked barnacle larval settlement, whereas Ca2+/CaM-dependent kinase II (CaMKII) inhibitors did not show any clear effects. The subsequent real-time PCR assay showed a higher expression level of Ba-MLCK gene in larval stages than in adults, suggesting an important role of Ba-MLCK gene in larval development and competency. Expert consultation on public health needs related to surveillance of SARS-CoV-2 in wastewater, Summary report of Virtual meeting, 30 November 2020, WHO
This report summarizes the findings of the Expert consultation on public health needs related to surveillance of SARS-CoV-2 in wastewater that was organized by the WHO European Centre for Environment and Health on 30 November 2020 in a virtual format. It aimed to support Member State authorities by facilitating exchange on the use, usefulness and limitations of SARS-CoV-2 surveillance in wastewater from a public health perspective, based on practices and experiences emerging in countries. Surveillance of SARS-CoV-2 RNA in wastewater can provide important complementary and independent information in public health decision-making in the context of the pandemic. The health sector is the end-user of the information and therefore needs to take the lead in designing surveillance programmes, merging and linking the data with other surveillance platforms, and coordinating interpretation and communication of the findings.</p
Surveillance environnementale des Poliovirus en Belgique - Etude de faisabilité
La surveillance environnementale est proposée comme l’une des solutions possibles à l’amélioration la qualité de la Surveillance Polio en Belgique.
Dans ce sens, une étude de faisabilité relative à la Surveillance environnementale Polio a été commanditée à Sciensano, par le cabinet de la ministre de la Santé publique. L’étude a été finalisée en janvier 2020.
La surveillance environnementale envisagée par l’étude a pour objectif de démontrer l’absence de poliovirus (PV) circulants, ainsi que de détecter rapidement, le cas échant, toute réintroduction du virus. La présence d’établissements « PEF », détenteurs de poliovirus en Belgique (GSK Rixensart et Fri-pharma), renforce l’importance de disposer d’un tel outil.
La plus-value d’une surveillance environnementale réside aussi dans sa complémentarité à la surveillance actuelle. Elle offre notamment l’avantage de jouer un rôle d’avertissement précoce, avec une détection plus rapide du PV après son introduction dans la population. Un autre avantage est que le système permet de couvrir préférentiellement les personnes et les zones les plus à risque de PV.
Le système de surveillance environnementale proposé dans l’étude de faisabilité offre une flexibilité de mise en œuvre. Concrètement, le système pourra être intensifié en cas d’incident relatif au PV (p.ex déversement accidentel dans l’environnement), ou étendu à la surveillance d’autres pathogènes (p. ex. norovirus, rotavirus…), et ce, selon les priorités de santé publique.
Le design proposé assure aussi une sensibilité de détection suffisante.
Le réseau de surveillance se composerait des sites suivants : le site GSK Rixensart, en tant que « would-be-PEF » le plus critique, le centre d’accueil pour demandeurs d’asile « Petit Château » et le quartier congolais « Matonge » de Bruxelles. Pour assurer une bonne représentativité de la population, deux stations d’épuration des eaux usées, une à Bruxelles, et la seconde à Anvers, seraient également incluses au réseau de surveillance.
L’étude réalisée confirme la faisabilité technique de contrôler ces différents endroits du réseau d’assainissement belge (égouts et stations d’épuration des eaux usées), propose un plan d’échantillonnage adapté, le tout décliné en un projet sur 4 ans, avec réévaluation annuelle.
Le coût annuel pour le projet de surveillance environnementale est estimé entre 143 000 € et 166 500 € pour la période de 4 ans, selon le scénario choisi. La part principale de ce budget (~70%) revient à l’analyse des échantillons (ressources humaines, matériel de laboratoire, et coûts des analyses).</p

De eerste analyses van de aanwezigheid van COVID-19 in afvalwater wijzen op trends die vergelijkbaar zijn met de resultaten van de opsporingstests die sinds midden september 2020 worden verricht. De nationale surveillance van het afvalwater, gecoördineerd door het Belgisch instituut voor gezondheid Sciensano, biedt een globaal en objectief beeld van de omloop van het virus in België. Het onderzoek is niet beïnvloed door de evolutie van de teststrategie of de selectie van de geteste personen (leeftijd en aan- of afwezigheid van symptomen). Dat maakt het mogelijk om vroegtijdig elke belangrijke evolutie in de omloop van het virus op te sporen.</p

Depuis le 15 septembre, sous l’impulsion de Sciensano, un nouvel outil de surveillance du coronavirus à l’échelle nationale a vu le jour. Cet outil vise à suivre la circulation du SARS-CoV-2 dans la population via l’analyse, non plus des tests réalisés directement sur les personnes, mais de prélèvements dans les eaux usées.</p

Implicaties voor de Volksgezondheid — Het risico op een epidemie in BelgiĂ« blijft verwaarloosbaar dankzij een hoge vaccinatiegraad in de 3 gewesten. Maar zolang het virus nog circuleert in de wereld, kan een geval van poliomyelitis worden geĂŻmporteerd in BelgiĂ« en kan het virus worden overgedragen op personen die niet of onvolledig gevaccineerd zijn. Het surveillancesysteem moet dus kunnen aantonen dat het poliovirus niet circuleert op het grondgebied en een herintroductie van het virus snel kunnen detecteren.</p
