    ORDINARY LEAST SQUARES REGRESSION OF ORDERED CATEGORICAL DATA: INFERENTIAL IMPLICATIONS FOR PRACTICE

    Ordered categorical responses (OCRs) are frequently encountered in many disciplines. Examples of interest in agriculture include quality assessments, such as for soil or food products, and evaluation of lesion severity, such as teat ends status in dairy cattle. OCRs are characterized by multiple categories recorded on a ranked scale that, while apprising relative order, is not informative of absolute magnitude of or proportionality between the categories. A number of statistically sound models for OCRs are available in the statistical literature, such as logistic regression and probit models, but these are commonly underutilized in practice. Instead, the ordinary least squares linear regression (OLSLR) model is often employed despite violation of basic model assumptions. In this study, the inferential implications of OLSLR-based inference on OCRs were investigated using a simulation study that evaluated realized Type I error rate and empirical statistical power. The design of the simulation study was motivated by applications reported in the subject-matter literature. A variety of plausible scenarios were considered for simulation, including various shapes of the frequency distribution and number of categories of the OCR. Using survey data on frequency of antimicrobial use in cattle feedlots, we illustrated the inferential performance of OLSLR on OCRs relative to a probit model

    Regional agriculture surveys using ERTS-1 data

    The Center for Remote Sensing Research has conducted studies designed to evaluate the potential application of ERTS data in performing agricultural inventories, and to develop efficient methods of data handling and analysis useful in the operational context for performing large area surveys. This work has resulted in the development of an integrated system utilizing both human and computer analysis of ground, aerial, and space imagery, which has been shown to be very efficient for regional crop acreage inventories. The technique involves: (1) the delineation of ERTS images into relatively homogeneous strata by human interpreters, (2) the point-by-point classification of the area within each strata on the basis of crop type using a human/machine interactive digital image processing system; and (3) a multistage sampling procedure for the collection of supporting aerial and ground data used in the adjustment and verification of the classification results

    Facilitators and Barriers to Prescribing PreExposure Prophylaxis (PrEP) for the Prevention of HIV

    Background: What is PrEP and who gets it? PrEP is the use of medication by individuals to prevent HIV contraction, approved in 2012 after demonstrating safety and efficacy in the iPrEx study and Partners PrEP2 trials. HIV infection risk is 92% lower in patients using PrEP. Truvada®, a combination of tenofovir and emtricitabine taken orally daily, is the only approved PrEP regimen and is intended to compliment other prevention strategies such as condoms. HIV negative-individuals at risk for exposure to HIV have been identified as men who have sex with men (MSM), IV drug users, heterosexuals who have unprotected sex with partners of unknown HIV status, and those in serodiscordant relationships. Barriers to PrEP Implementation PrEP is effective when patients adhere; however, both the medical community and some high-risk populations have been slow to adopt it as an HIV prevention strategy. Surveys have shown clinicians perceived barriers to PrEP such as adverse side effects, viral drug resistance, increased high-risk behavior, cost, and training. HIV in Vermont New diagnoses of HIV among Vermont residents has remained relatively stable over the last twenty years. Vermont CARES, a non-profit, offers free and anonymous HIV tests and in-person risk-reduction counseling.     E-QED: Electrical Bug Localization During Post-Silicon Validation Enabled by Quick Error Detection and Formal Methods

    During post-silicon validation, manufactured integrated circuits are extensively tested in actual system environments to detect design bugs. Bug localization involves identification of a bug trace (a sequence of inputs that activates and detects the bug) and a hardware design block where the bug is located. Existing bug localization practices during post-silicon validation are mostly manual and ad hoc, and, hence, extremely expensive and time consuming. This is particularly true for subtle electrical bugs caused by unexpected interactions between a design and its electrical state. We present E-QED, a new approach that automatically localizes electrical bugs during post-silicon validation. Our results on the OpenSPARC T2, an open-source 500-million-transistor multicore chip design, demonstrate the effectiveness and practicality of E-QED: starting with a failed post-silicon test, in a few hours (9 hours on average) we can automatically narrow the location of the bug to (the fan-in logic cone of) a handful of candidate flip-flops (18 flip-flops on average for a design with ~ 1 Million flip-flops) and also obtain the corresponding bug trace. The area impact of E-QED is ~2.5%. In contrast, deter-mining this same information might take weeks (or even months) of mostly manual work using traditional approaches

    Inhibition of the \u3cem\u3edapE\u3c/em\u3e-Encoded \u3cem\u3eN\u3c/em\u3e-Succinyl- ʟ, ʟ-diaminopimelic Acid Desuccinylase from \u3cem\u3eNeisseria meningitidis\u3c/em\u3e by ʟ-Captopril

    Binding of the competitive inhibitor ʟ-captopril to the dapE-encoded N-succinyl-ʟ, ʟ-diaminopimelic acid desuccinylase from Neisseria meningitidis (NmDapE) was examined by kinetic, spectroscopic, and crystallographic methods. ʟ-Captopril, an angiotensin-converting enzyme (ACE) inhibitor, was previously shown to be a potent inhibitor of the DapE from Haemophilus influenzae (HiDapE) with an IC50 of 3.3 μM and a measured Ki of 1.8 μM and displayed a dose-responsive antibiotic activity toward Escherichia coli. ʟ-Captopril is also a competitive inhibitor of NmDapE with a Ki of 2.8 μM. To examine the nature of the interaction of ʟ-captopril with the dinuclear active site of DapE, we have obtained electron paramagnetic resonance (EPR) and magnetic circular dichroism (MCD) data for the enzymatically hyperactive Co(II)-substituted forms of both HiDapE and NmDapE. EPR and MCD data indicate that the two Co(II) ions in DapE are antiferromagnetically coupled, yielding an S = 0 ground state, and suggest a thiolate bridge between the two metal ions. Verification of a thiolate-bridged dinuclear complex was obtained by determining the three-dimensional X-ray crystal structure of NmDapE in complex with ʟ-captopril at 1.8 Å resolution. Combination of these data provides new insights into binding of ʟ-captopril to the active site of DapE enzymes as well as important inhibitor–active site residue interaction’s. Such information is critical for the design of new, potent inhibitors of DapE enzymes

    Ultraspectral Sounding Retrieval Error Budget and Estimation

    The ultraspectral infrared radiances obtained from satellite observations provide atmospheric, surface, and/or cloud information. The intent of the measurement of the thermodynamic state is the initialization of weather and climate models. Great effort has been given to retrieving and validating these atmospheric, surface, and/or cloud properties. Error Consistency Analysis Scheme (ECAS), through fast radiative transfer model (RTM) forward and inverse calculations, has been developed to estimate the error budget in terms of absolute and standard deviation of differences in both spectral radiance and retrieved geophysical parameter domains. The retrieval error is assessed through ECAS without assistance of other independent measurements such as radiosonde data. ECAS re-evaluates instrument random noise, and establishes the link between radiometric accuracy and retrieved geophysical parameter accuracy. ECAS can be applied to measurements of any ultraspectral instrument and any retrieval scheme with associated RTM. In this paper, ECAS is described and demonstration is made with the measurements of the METOP-A satellite Infrared Atmospheric Sounding Interferometer (IASI).
