    Associations between epigenetic aging and childhood peer victimization, depression, and suicidal ideation in adolescence and adulthood: A study of two population-based samples

    Background: Prior studies indicate that peer victimization (including bullying) is associated with higher risk for depression and suicidal ideation across the life course. However, molecular mechanisms underlying these associations remain unclear. This two-cohort study proposes to test whether epigenetic aging and pace of aging, as well as a DNA methylation marker of responsive to glucocorticoids, are associated to childhood peer victimization and later depressive symptoms, or suicidal ideation. Methods: Cohort 1: Epigenome-wide DNA methylation (EPIC array) was measured in saliva collected when participants were 10.47 years (standard deviation = 0.35) in a subsample of the Quebec Longitudinal Study of Child Development (QLSCD, n = 149 participants), with self-reported peer victimization at 6-8 years, depressive symptoms (mean symptoms, and dichotomized top 30% symptoms) and suicidal ideation at 15-17 years. Cohort 2: Epigenome-wide DNA methylation (EPIC array) was measured in blood collected from participants aged 45.13 years (standard deviation = 0.37) in a subsample of the 1958 British Birth cohort (1958BBC, n = 238 participants) with information on mother-reported peer victimization at 7-11 years, self-reported depressive symptoms at 50 years, and suicidal ideation at 45 years. Five epigenetic indices were derived: three indicators of epigenetic aging [Horvath's pan-tissue (Horvath1), Horvath's Skin-and-Blood (Horvath2), Pediatric-Buccal-Epigenetic age (PedBE)], pace of aging (DunedinPACE), and stress response reactivity (Epistress). Results: Peer victimization was not associated with the epigenetic indices in either cohort. In the QLSCD, higher PedBE epigenetic aging and a slower pace of aging as measured by DunedinPACE predicted higher depressive symptoms scores. In contrast, neither the Horvath1, or Horvath2 epigenetic age estimates, nor the Epistress score were associated with depressive symptoms in either cohort, and none of the epigenetic indices predicted suicidal ideation. Conclusion: The findings are consistent with epigenome-wide and candidate gene studies suggesting that these epigenetic indices did not relate to peer victimization, challenging the hypothesis that cumulative epigenetic aging indices could translate vulnerability to depressive symptoms and suicidal ideation following peer victimization. Since some indices of epigenetic aging and pace of aging signaled higher risk for depressive symptoms, future studies should pursue this investigation to further evaluate the robustness and generalization of these preliminary findings

    The CUORE Cryostat: A 1-Ton Scale Setup for Bolometric Detectors

    The cryogenic underground observatory for rare events (CUORE) is a 1-ton scale bolometric experiment whose detector consists of an array of 988 TeO2 crystals arranged in a cylindrical compact structure of 19 towers. This will be the largest bolometric mass ever operated. The experiment will work at a temperature around or below 10 mK. CUORE cryostat consists of a cryogen-free system based on pulse tubes and a custom high power dilution refrigerator, designed to match these specifications. The cryostat has been commissioned in 2014 at the Gran Sasso National Laboratories and reached a record temperature of 6 mK on a cubic meter scale. In this paper, we present results of CUORE commissioning runs.     Depuis 2014, le Musée des beaux-arts de Montréal développe une plateforme nommée ÉducArt, qui suggère des activités éducatives réalisées à partir de sa collection encyclopédique pour soutenir les écoles secondaires québécoises dans leur mission culturelle. Comme l’élaboration de ressources destinées au milieu scolaire demande de tenir compte des besoins et des réalités de la classe (Van der Maren, 1996, 2003) et que le gouvernement du Québec entend poursuivre le financement de plateformes muséales (Rocheleau, 2016), il apparaît important d’étudier les enjeux associés à l’appropriation de ressources numériques muséales, telles que celles diffusées sur la plateforme ÉducArt, par des enseignants du secondaire du Québec. Dans cette perspective, un cadre d’analyse visant à mieux comprendre ces enjeux semble essentiel. Cet article se veut une contribution en ce sens, car à notre connaissance, aucun modèle théorique n’existe pour comprendre les enjeux particuliers que rencontrent des enseignants du secondaire lorsqu’ils tentent de s’approprier des ressources numériques muséales.Since 2014, the Montreal Museum of Fine Arts develops a platform called ÉducArt, which offers educational activities carried out from its encyclopaedic collection to support Quebec high schools in their cultural mission. As the development of such resources for schools requires consideration of the class needs and realities (Van der Maren, 1996; 2003) and Quebec’s government intends to continue financing museum platforms (Rocheleau, 2016), it appears important to study the issues associated with the appropriation of digital museum resources, such as those published on the ÉducArt platform, by high school teachers in Quebec. In this context, an analytical framework to better understand these issues seems essential. This article is intended to be a contribution in this direction because, to our knowledge, no theoretical model exists in order to specifically understand the issues encountered by high school teachers when trying to appropriate digital museum resources

    Exploring the psychological rewards of a familiar semirural landscape: connecting to local nature through a mindful approach

    This study analyses a 53,000 word diary of a year engaging with nature through over 200 trips to a semi-rural landscape. Thematic analysis revealed two themes; the transition from observer to nature connectedness and the ways in which the natural environment was experienced once a connection was made. These themes are discussed in relation to theories that seek to explain the positive effect of nature and nature connectedness. The findings are important as they suggest that repeated engagement with local semi-rural countryside can lead to a mindful approach and psychological rewards that do not require travel into the wilderness. The work informs further research into outcomes and processes of nature based interventions such as: trip frequency, duration and diary keeping

    The CUORE cryostat: an infrastructure for rare event searches at millikelvin temperatures

    The CUORE experiment is the world's largest bolometric experiment. The detector consists of an array of 988 TeO2 crystals, for a total mass of 742 kg. CUORE is presently taking data at the Laboratori Nazionali del Gran Sasso, Italy, searching for the neutrinoless double beta decay of 130Te. A large custom cryogen-free cryostat allows reaching and maintaining a base temperature of about 10 mK, required for the optimal operation of the detector. This apparatus has been designed in order to achieve a low noise environment, with minimal contribution to the radioactive background for the experiment. In this paper, we present an overview of the CUORE cryostat, together with a description of all its sub-systems, focusing on the solutions identified to satisfy the stringent requirements. We briefly illustrate the various phases of the cryostat commissioning and highlight the relevant steps and milestones achieved each time. Finally, we describe the successful cooldown of CUORE

    Cystamine/cysteamine rescues the dopaminergic system and shows neurorestorative properties in an animal model of Parkinson's disease.

    The neuroprotective properties of cystamine identified in pre-clinical studies have fast-tracked this compound to clinical trials in Huntington's disease, showing tolerability and benefits on motor symptoms. We tested whether cystamine could have such properties in a Parkinson's disease murine model and now provide evidence that it can not only prevent the neurodegenerative process but also can reverse motor impairments created by a 6-hydroxydopamine lesion 3weeks post-surgery. Importantly, we report that cystamine has neurorestorative properties 5weeks post-lesion as seen on the number of nigral dopaminergic neurons which is comparable with treatments of cysteamine, the reduced form of cystamine used in the clinic, as well as rasagiline, increasingly prescribed in early parkinsonism. All three compounds induced neurite arborization of the remaining dopaminergic cells which was further confirmed in ex vivo dopaminergic explants derived from Pitx3-GFP mice. The disease-modifying effects displayed by cystamine/cysteamine would encourage clinical testing

    Effect of graphene additive on flexural and interlaminar shear strength properties of carbon fiber-reinforced polymer composite

    Composite materials are widely used in various manufacturing fields from aeronautic and aerospace industries to the automotive industry. This is due to their outstanding mechanical properties with respect to their light weight. However, some studies showed that the major flaws of these materials are located at the fiber/matrix interface. Therefore, enhancing matrix adhesion properties could significantly improve the overall material characteristics. This study aims to analyze the effect of graphene particles on the adhesion properties of carbon fiber-reinforced polymer (CFRP) through interlaminar shear strength (ILSS) and flexural testing. Seven modified epoxy resins were prepared with different graphene contents. The CFRP laminates were next manufactured using a method that guarantees a repeatable and consistent fiber volume fraction with a low porosity level. Short beam shear and flexural tests were performed to compare the effect of graphene on the mechanical properties of the different laminates. It was found that 0.25 wt.% of graphene filler enhanced the flexural strength by 5%, whilst the higher concentrations (2 and 3 wt.%) decreased the flexural strength by about 7%. Regarding the ILSS, samples with low concentrations (0.25 and 0.5 wt.%) demonstrated a decent increase. Meanwhile, 3 wt.% slightly decreases the ILSS

    Radon backgrounds in the DEAP-1 liquid-argon-based Dark Matter detector

    The DEAP-1 \SI{7}{kg} single phase liquid argon scintillation detector was operated underground at SNOLAB in order to test the techniques and measure the backgrounds inherent to single phase detection, in support of the \mbox{DEAP-3600} Dark Matter detector. Backgrounds in DEAP are controlled through material selection, construction techniques, pulse shape discrimination and event reconstruction. This report details the analysis of background events observed in three iterations of the DEAP-1 detector, and the measures taken to reduce them. The 222^{222}Rn decay rate in the liquid argon was measured to be between 16 and \SI{26}{\micro\becquerel\per\kilogram}. We found that the background spectrum near the region of interest for Dark Matter detection in the DEAP-1 detector can be described considering events from three sources: radon daughters decaying on the surface of the active volume, the expected rate of electromagnetic events misidentified as nuclear recoils due to inefficiencies in the pulse shape discrimination, and leakage of events from outside the fiducial volume due to imperfect position reconstruction. These backgrounds statistically account for all observed events, and they will be strongly reduced in the DEAP-3600 detector due to its higher light yield and simpler geometry
