    A Gel Probe Equilibrium Sampler for Measuring Arsenic Porewater Profiles and Sorption Gradients in Sediments: I. Laboratory Development

    A gel probe equilibrium sampler has been developed to study arsenic (As) geochemistry and sorption behavior in sediment porewater. The gels consist of a hydrated polyacrylamide polymer, which has a 92% water content. Two types of gels were used in this study. Undoped (clear) gels were used to measure concentrations of As and other elements in sediment porewater. The polyacrylamide gel was also doped with hydrous ferric oxide (HFO), an amorphous iron (Fe) oxyhydroxide. When deployed in the field, HFO-doped gels introduce a fresh sorbent into the subsurface thus allowing assessment of in situ sorption. In this study, clear and HFO-doped gels were tested under laboratory conditions to constrain the gel behavior prior to field deployment. Both types of gels were allowed to equilibrate with solutions of varying composition and re-equilibrated in acid for analysis. Clear gels accurately measured solution concentrations (±1%), and As was completely recovered from HFO-doped gels (±4%). Arsenic speciation was determined in clear gels through chromatographic separation of the re-equilibrated solution. For comparison to speciation in solution, mixtures of As(III) and As(V) adsorbed on HFO embedded in gel were measured in situ using X-ray absorption spectroscopy (XAS). Sorption densities for As(III) and As(V) on HFO embedded in gel were obtained from sorption isotherms at pH 7.1. When As and phosphate were simultaneously equilibrated (in up to 50-fold excess of As) with HFO-doped gels, phosphate inhibited As sorption by up to 85% and had a stronger inhibitory effect on As(V) than As(III). Natural organic matter (>200 ppm) decreased As adsorption by up to 50%, and had similar effects on As(V) and As(III). The laboratory results provide a basis for interpreting results obtained by deploying the gel probe in the field and elucidating the mechanisms controlling As partitioning between solid and dissolved phases in the environment

    Both equilibrium and nonequilibrium factors influence the efficacy of pharmaceutical agents that target intermediate states of biochemical reactions. We explored the intermediate state inhibition of gp41, part of the HIV-1 envelope glycoprotein complex (Env) that promotes viral entry through membrane fusion. This process involves a series of gp41 conformational changes coordinated by Env interactions with cellular CD4 and a chemokine receptor. In a kinetic window between CD4 binding and membrane fusion, the N- and C-terminal regions of the gp41 ectodomain become transiently susceptible to inhibitors that disrupt Env structural transitions. In this study, we sought to identify kinetic parameters that influence the antiviral potency of two such gp41 inhibitors, C37 and 5-Helix. Employing a series of C37 and 5-Helix variants, we investigated the physical properties of gp41 inhibition, including the ability of inhibitor-bound gp41 to recover its fusion activity once inhibitor was removed from solution. Our results indicated that antiviral activity critically depended upon irreversible deactivation of inhibitor-bound gp41. For C37, which targets the N-terminal region of the gp41 ectodomain, deactivation was a slow process that depended on chemokine receptor binding to Env. For 5-Helix, which targets the C-terminal region of the gp41 ectodomain, deactivation occurred rapidly following inhibitor binding and was independent of chemokine receptor levels. Due to this kinetic disparity, C37 inhibition was largely reversible, while 5-Helix inhibition was functionally irreversible. The fundamental difference in deactivation mechanism points to an unappreciated asymmetry in gp41 following inhibitor binding and impacts the development of improved fusion inhibitors and HIV-1 vaccines. The results also demonstrate how the activities of intermediate state inhibitors critically depend upon the final disposition of inhibitor-bound states

    We revisit Kellerer's Theorem, that is, we show that for a family of real probability distributions (μt)t[0,1](\mu_t)_{t\in [0,1]} which increases in convex order there exists a Markov martingale (St)t[0,1](S_t)_{t\in[0,1]} s.t.\ StμtS_t\sim \mu_t. To establish the result, we observe that the set of martingale measures with given marginals carries a natural compact Polish topology. Based on a particular property of the martingale coupling associated to Root's embedding this allows for a relatively concise proof of Kellerer's theorem. We emphasize that many of our arguments are borrowed from Kellerer \cite{Ke72}, Lowther \cite{Lo07}, and Hirsch-Roynette-Profeta-Yor \cite{HiPr11,HiRo12}.Comment: 8 pages, 1 figur

    International audiencePotential Aerosol Mass (PAM) can be defined as the maximum aerosol mass that the oxidation of precursor gases produces. In the measurement, all precursor gases are rapidly oxidized with extreme amounts of oxidants to low volatility compounds, resulting in the aerosol formation. Oxidation occurs in a small, simple, flow-through chamber that has a short residence time and is irradiated with ultraviolet light. The amount of the oxidants ozone (O3), hydroxyl (OH), and hydroperoxyl (HO2) were measured directly and can be controlled by varying the UV light and the relative humidity. Maximum values were 40 ppmv for O3, 500 pptv for OH, and 4 ppbv for HO2. The oxidant amounts are 100 to 1000 times troposphere values, but the ratios OH/O3 and HO2/OH are similar to troposphere values. The aerosol production mechanism and the aerosol mass yield were studied for several controlling variables, such as temperature, relative humidity, oxidant concentration, presence of nitrogen oxides (NOx), precursor gas composition and amount, and the presence of acidic seed aerosol. The measured secondary organic aerosol (SOA) yield of several natural and anthropogenic volatile organic compounds and a mixture of hydrocarbons in the PAM chamber were similar to those obtained in large, batch-style environmental chambers. This PAM method is being developed for measuring potential aerosol mass in the atmosphere, but is also useful for examining SOA processes in the laboratory and in environmental chambers

    Clergy Interest in Innovative Collaboration with Psychologists

    What forms of innovative collaboration are possible between clergy and psychologists? A total of 117 clergypersons (63% response rate) rated 6 scenarios of collaboration, indicating their level of interest and the extent to which they would like to remain involved with the psychologist. The scenarios were derived from two categories of collaboration articulated by in previous research: mental health services and enhancing parish life. Overall, clergy expressed relatively modest levels of interest in innovative collaboration, though they were somewhat interested in mental health consultation services. Many clergy refer troubled parishioners to clinical or counseling psychologists for treatment, but appear less interested in more innovative forms of collaboration

    Delivering a “Dose of Hope”: A Faith-Based Program to Increase Older African Americans’ Participation in Clinical Trials

    Background: Underrepresentation of older-age racial and ethnic minorities in clinical research is a significant barrier to health in the United States, as it impedes medical research advancement of effective preventive and therapeutic strategies. Objective: The objective of the study was to develop and test the feasibility of a community-developed faith-based intervention and evaluate its potential to increase the number of older African Americans in clinical research. Methods: Using a cluster-randomized design, we worked with six matched churches to enroll at least 210 persons. We provided those in the intervention group churches with three educational sessions on the role of clinical trials in addressing health disparity topics, and those in the comparison group completed surveys at the same timepoints. All persons enrolled in the study received ongoing information via newsletters and direct outreach on an array of clinical studies seeking participants. We evaluated the short-, mid-, and longer-term effects of the interventional program on clinical trial-related outcomes (ie, screening and enrollment)

    The Kramers equation simulation algorithm II. An application to the Gross-Neveu model

    We continue the investigation on the applications of the Kramers equation to the numerical simulation of field theoretic models. In a previous paper we have described the theory and proposed various algorithms. Here, we compare the simplest of them with the Hybrid Monte Carlo algorithm studying the two-dimensional lattice Gross-Neveu model. We used a Symanzik improved action with dynamical Wilson fermions. Both the algorithms allow for the determination of the critical mass. Their performances in the definite phase simulations are comparable with the Hybrid Monte Carlo. For the two methods, the numerical values of the measured quantities agree within the errors and are compatible with the theoretical predictions; moreover, the Kramers algorithm is safer from the point of view of the numerical precision.Comment: 20 pages + 1 PostScript figure not included, REVTeX 3.0, IFUP-TH-2
