The differences in thermal profiles between normal and leukemic cells exposed to anticancer drug evaluated by differential scanning calorimetry
Chronic lymphocytic leukemia (CLL) is a heterogenous disease with an imbalance between apoptosis and cell proliferation. Therefore, the main goal in CLL therapy is to induce apoptosis and effectively support this process in transformed B lymphocytes. In the current study, we have compared differential scanning calorimetry (DSC) profiles of nuclei isolated from CLL cells and normal mononuclear cells exposed to cladribine or fludarabine combined with mafosfamide (CM; FM), and additionally to CM combined with monoclonal antibody鈥攔ituximab (RCM) for 48 h, as well as in culture medium only (controls). Under current study, the mononuclear cells from peripheral blood (PBMCs) of healthy individuals have been included. The obtained results have shown the presence of thermal transition at 95 卤 5 掳C in most of nuclear preparations (92.2 %) isolated from blood of CLL patients. This thermal characteristic parameter was changed after drug exposure, however, to a different extent. These thermal changes were accompanied by the decrease of cell viability, an elevation of apoptosis rate and the changes in expression/proteolysis of poly(ADP-ribose)polymerase-1鈥攎ain marker of apoptosis. Importantly, in DSC profiles of nuclear preparations of PBMCs from blood of healthy donors exposed to investigated drug combinations and control CLL cells, the lack of such changes was observed. The use of ceramics fused to metal has gained wide popularity when choosing a material suitable for making dental crowns and bridges not only for aesthetic reasons, but more importantly due to the strength properties of the metallic substructure when placed on the previously grinded dental stump. The process of fusing dental ceramic to the metallic substructure does not always prove to be a success. Metal-ceramic dental restorations do not always exhibit flaws visible to the eye on the outer surface; often the metal-ceramic fusing process creates microscopic flaws. This thesis presents the different kinds of flaws which can occur when bonding two different kinds of prosthetic materials together and the outcome we can expect as a result

This thesis will allow its author to define the features of a metal structure cooled by means of various methods. All performed tests revealed some flaws in the metal structure. They made it possible to define the metal-ceramics connection and draw some preliminary conclusions showing at the same time that what has a great influence on the metal structure and the metal-ceramics connection is the way of cooling the casing material shortly after it has been cast

The thesis determines features of metal structure which directly after the casting in screnning substance cooled in running water. The research shown failures in metal structure, the change in elements construction, composition in particular places of the sample ( in ceramic and metal layer and on the boundry between them) shown the image of its structure and identification of structural ingredients and allowed to determine the combination between two phases creating the prosthetic restoration
