    Electrocardiography and echocardiography in athletic heart imagining

    The aim of the study was evaluation of electrocardiographic and echocardiographic parameters in athletes. „Athletic heart” characteristics were compared with fit persons’ heart. 96 athletes participated in the study. Sportsmen were divided into: static (S), dynamic (D) and composite (SD) exercise groups and Polish (I), European (II), World (III) champions. 30 students from Sport Academy formed the control group (K). Electrocardiographic and echocardiographic examination were performed in everyone. As regards the type of exercise, the end-systolic left ventricular (LV) dimension was smaller in S in comparison with D and SD (28.9 vs 32.2 and 32.68 mm; P<0.05). LV mass was bigger in D in comparison with K (273.2 vs 218.6 g; P<0.05). Medium Pulmonary Artery Pressure (MPAP) in S and SD was lower in respect to D and K (11.75; 11.08 vs 15.52; 17.43 mm Hg; P<0.05). We observed lower heart rate in D, SD in comparison with K (58.64; 60.54 vs 68.8; P<0.05), bigger R wave amplitude in V5 (RV5) (21.65; 23.5 vs 15.03 mm; P<0.05) and V6 (RV6) (23.5 vs 15.3 mm; P<0.05) in group S in respect to K. LV mass was bigger in III than in K (261.3 vs 218.6 g; P<0.05). MPAP was lower in I and II in comparison with K (11.42; 13.13 vs 17.43 mmHg; P<0.05). HR was lower in categories I, II than in K (61.32; 60.13 vs 68.8; P<0.05), RV5 was bigger in I in comparison with K (19.5 vs 15.03 mm; P<0.05). The electrocardiography and echocardiography proves to find some significant differences between athletic and fit persons’ heart especially as concerns MPAP, RV5, RV6 values

    The Role of Toll-Like Receptor 2 and 4 Innate Immunity Pathways in Intracortical Microelectrode-Induced Neuroinflammation

    We have recently demonstrated that partial inhibition of the cluster of differentiation 14 (CD14) innate immunity co-receptor pathway improves the long-term performance of intracortical microelectrodes better than complete inhibition. We hypothesized that partial activation of the CD14 pathway was critical to a neuroprotective response to the injury associated with initial and sustained device implantation. Therefore, here we investigated the role of two innate immunity receptors that closely interact with CD14 in inflammatory activation. We implanted silicon planar non-recording neural probes into knockout mice lacking Toll-like receptor 2 (Tlr2−/−), knockout mice lacking Toll-like receptor 4 (Tlr4−/−), and wildtype (WT) control mice, and evaluated endpoint histology at 2 and 16 weeks after implantation. Tlr4−/− mice exhibited significantly lower BBB permeability at acute and chronic time points, but also demonstrated significantly lower neuronal survival at the chronic time point. Inhibition of the Toll-like receptor 2 (TLR2) pathway had no significant effect compared to control animals. Additionally, when investigating the maturation of the neuroinflammatory response from 2 to 16 weeks, transgenic knockout mice exhibited similar histological trends to WT controls, except that knockout mice did not exhibit changes in microglia and macrophage activation over time. Together, our results indicate that complete genetic removal of Toll-like receptor 4 (TLR4) was detrimental to the integration of intracortical neural probes, while inhibition of TLR2 had no impact within the tests performed in this study. Therefore, approaches focusing on incomplete or acute inhibition of TLR4 may still improve intracortical microelectrode integration and long term recording performance

    Effectiveness of ChĂȘneau brace treatment for idiopathic scoliosis: prospective study in 79 patients followed to skeletal maturity

    <p>Abstract</p> <p>Background</p> <p>Progressive idiopathic scoliosis can negatively influence the development and functioning of 2-3% of adolescents, with health consequences and economic costs, placing the disease in the centre of interest of the developmental medicine. The aim of this study was to evaluate the effectiveness of ChĂȘneau brace in the management of idiopathic scoliosis.</p> <p>Methods</p> <p>A prospective observational study according to SOSORT and SRS recommendations comprised 79 patients (58 girls and 21 boys) with progressive idiopathic scoliosis, treated with ChĂȘneau brace and physiotherapy, with initial Cobb angle between 20 and 45 degrees, no previous brace treatment, Risser 4 or more at the final evaluation and minimum one year follow-up after weaning the brace. Achieving 50° of Cobb angle was considered surgical recommendation.</p> <p>Results</p> <p>At follow-up 20 patients (25.3%) improved, 18 patients (22.8%) were stable, 31 patients (39.2%) progressed below 50 degrees and 10 patients (12.7%) progressed beyond 50 degrees (2 of these 10 patients progressed beyond 60 degrees). Progression concerned the younger and less skeletally mature patients.</p> <p>Conclusion</p> <p>Conservative treatment with ChĂȘneau orthosis and physiotherapy was effective in halting scoliosis progression in 48.1% of patients. The results of this study suggest that bracing is effective in reducing the incidence of surgery in comparison with natural history.</p

    Lipid rafts are essential for release of phosphatidylserine-exposing extracellular vesicles from platelets.

    Platelets protect the vascular system during damage or inflammation, but platelet activation can result in pathological thrombosis. Activated platelets release a variety of extracellular vesicles (EVs). EVs shed from the plasma membrane often expose phosphatidylserine (PS). These EVs are pro-thrombotic and increased in number in many cardiovascular and metabolic diseases. The mechanisms by which PS-exposing EVs are shed from activated platelets are not well characterised. Cholesterol-rich lipid rafts provide a platform for coordinating signalling through receptors and Ca2+ channels in platelets. We show that cholesterol depletion with methyl-ÎČ-cyclodextrin or sequestration with filipin prevented the Ca2+-triggered release of PS-exposing EVs. Although calpain activity was required for release of PS-exposing, calpain-dependent cleavage of talin was not affected by cholesterol depletion. P2Y12 and TPα, receptors for ADP and thromboxane A2, respectively, have been reported to be in platelet lipid rafts. However, the P2Y12 antagonist, AR-C69931MX, or the cyclooxygenase inhibitor, aspirin, had no effect on A23187-induced release of PS-exposing EVs.
