    Blood flow changes in pelvic vessels associated with the application of an abdominal compression belt in healthy postpartum women

    Introduction: Postpartum haemorrhage (PPH) accounts for a high proportion of maternal mortality and morbidity throughout the world. A uterine compression belt which has been developed recently represents a very low tech, low cost solution in managing postpartum haemorrhage. Objectives: To evaluate the blood flow changes in pelvic vessels following application of the postpartum haemorrhage compression belt (Laerdal Global Health, Stavanger, Norway). Methods: The sample included healthy postpartum women within 6 hours of vaginal delivery. The study was performed at Teaching Hospital, Ragama, Sri Lanka. PPH compression belt was applied on the lower abdomen in a supine position with a slight lateral tilt. Patient’s pulse, blood pressure and Doppler indices (RI, PI and PFV) of the uterine, internal iliac and femoral arteries were measured using transabdominal Doppler ultrasonography. Lower limb oxygen saturation was also measured. Measurements were obtained by connecting the subjects to a multimonitor throughout the study period of 20 minutes. Median RI, PI and PFV was calculated and comparisons were made between the baseline and after belt application at 10 and 20 minutes. Results: A total of 20 healthy women were included and the mean time from delivery to study inclusion was 2.5 (range 0.5–5.0) hours. There were no adverse outcomes or altered vital signs noted among participants. Overall there were no significant changes in the internal iliac, uterine and femoral artery blood flow after application of the compression belt. Conclusions: There were no significant changes in the internal iliac, uterine and femoral artery blood flow after application of the compression belt. This preliminary study only shows that the application of the PPH compression belt has no apparent adverse changes in the iliac, uterine and femoral artery blood flow in postpartum mothers

    Spectroscopic Methods For Lubricant Quality Control In Engines And Gear Boxes

    Lubricants play a vital role in reducing the wear and tear of engine/gear box metal parts. Number of analytical and spectroscopic methods have been used to analyze the quality of the lubricant oil. Moreover some parameters such as Total Acid Number (TAN), viscosity index also have been used to analyze the quality of the oil. Several used wind turbine gear oil samples were analyzed by various spectroscopic methods such as UV-Visible, Fourier-transform infrared (FTIR) and Fluorescence Spectroscopy. Fluorescence method gave promising results among those three spectroscopic methods. In order to study thermal degradation, motor oil samples were subjected to articial aging in the laboratory conditions by heating them up to different temperatures for different time periods and then subsequently analyzed with fluorescence spectroscopic method. Subsequently two used engine oil samples from a same diesel engine vehicle were analyzed using fluorescence spectroscopic method.  Notable variation in fluorescence emission intensities was observed with oil aging. Intensity of the fluorescence emission signal decrease with oil degradation.  Therefore fluorescence spectroscopic method can be used to predict the reusability of gear oils as well as to identify the oil degradation. This method can be further extended to develop a novel potential sensor to detect the quality of oil in various types of engines.     Sustainable Use of Biomass Boiler Ash as a Reinforcement Filler for Polyamide 6 Composite

    The use of biomass combustion ash as a reinforcing filler has taken the attention in recent years to reduce the overall production cost and increase the mechanical properties of plastics. This study evaluates the effect of biomass boilers ash (BBA) as a reinforcing filler on polyamide 6 composites (PA6). The chemical composition and thermal stability of BBA were analyzed by X-ray fluorescence (XRF), Thermogravimetric analysis (TGA), and Differential thermal analysis (DTA) thermographs. Test samples were prepared by varying the ash content (2.5%, 5%, 7.5%, and 10%), and also samples were prepared without adding ash as a control. Moreover, surface modification was carried out by using N-2 (Aminoethyl) 3-aminopropyl triethoxy diamino-silane as a possible coupling agent for BBA  and it was tested by varying the coupling agent concentration (0.5%, 1%, and 2%) by dry process and the samples were prepared by extrusion and injection molding processes. Comparative analysis of the degree of crystallinity, mechanical properties including tensile, flexural, and thermal properties were tested. The highest degree of crystallinity was obtained with 7.5% BBA-filled composites. The addition of 2% surface-modified BBA (SBBA) filled composites (PA6 / 10% Ash 2% CA) showed an increase of tensile strength and elongation at break than the 10% unmodified BBA (UBBA) (PA6/ 10% Ash) filled composites. Stress at peak and bending modulus values in the flexural test data were increased up to 10%. But it was significantly reduced with the increasing of modifying agent concentration due to the increment of stiffness of the composite

    ONC201 in combination with paxalisib for the treatment of H3K27-altered diffuse midline glioma

    Diffuse midline gliomas (DMG), including diffuse intrinsic pontine gliomas (DIPGs), are the most lethal of childhood cancers. Palliative radiotherapy is the only established treatment, with median patient survival of 9-11 months. ONC201 is a DRD2 antagonist and ClpP agonist that has shown preclinical and emerging clinical efficacy in DMG. However, further work is needed to identify the mechanisms of response of DIPGs to ONC201 treatment and to determine whether recurring genomic features influence response. Using a systems-biological approach, we showed that ONC201 elicits potent agonism of the mitochondrial protease ClpP to drive proteolysis of electron transport chain and tricarboxylic acid cycle proteins. DIPGs harboring PIK3CA-mutations showed increased sensitivity to ONC201, while those harboring TP53-mutations were more resistant. Metabolic adaptation and reduced sensitivity to ONC201 was promoted by redox-activated PI3K/Akt signaling, which could be counteracted using the brain penetrant PI3K/Akt inhibitor, paxalisib. Together, these discoveries coupled with the powerful anti-DIPG/DMG pharmacokinetic and pharmacodynamic properties of ONC201 and paxalisib have provided the rationale for the ongoing DIPG/DMG phase II combination clinical trial NCT05009992

    A novel dynamic multicellular co-culture system for studying individual blood-brain barrier cell types in brain diseases and cytotoxicity testing

    Blood brain barrier (BBB) cells play key roles in the physiology and pathology of the central nervous system (CNS). BBB dysfunction is implicated in many neurodegenerative diseases, including Alzheimer’s disease (AD). The BBB consists of capillary endothelial cells, pericytes encircling the endothelium and surrounding astrocytes extending their processes towards it. Although there have been many attempts to develop in vitro BBB models, the complex interaction between these celltypes makes it extremely difficult to determine their individual contribution to neurotoxicity in vivo. Thus, we developed and optimised an in vitro multicellular co-culture model within the Kirkstall Quasi Vivo System. The main aim was to determine the optimal environment to culture human brain primary endothelial cells, pericytes and astrocytes whilst maintaining cellular communication without formation of a barrier in order to assess the contribution of each cell type to the overall response. As a proof of concept for the present system, the effects of amyloid-beta 25-35 peptide (Aβ25-35), a hall mark of AD, were explored. This multicellular system will be a valuable tool for future studies on the specific roles of individual BBB cell type (while making connection with each other through medium) in CNS disorders as well as in cytotoxicity tests

    Two potentially lethal conditions of probable immune origin occurring in a pregnant woman: a case report

    Abstract Background Thrombotic thrombocytopenic purpura and peripartum cardiomyopathy are potentially lethal complications of pregnancy. We describe a case in which both of these developed in the same patient. The etiologies of both conditions remain uncertain, but they share immune hyperreactivity as a possible cause. Case presentation A 33-year-old Lankan primigravida gave birth at 38 weeks of gestation by cesarean section when she presented with right-sided abdominal pain and a provisional diagnosis of appendicitis. Her pain persisted postoperatively, and on the second postoperative day, she physicaly collapsed suddenly with abdominal distention. Immediate laparotomy revealed generalized oozing from the peritoneum resulting in hemoperitoneum and intestinal hemorrhage. Her laboratory reports showed microangiopathic hemolytic anemia and thrombocytopenia. She also had elevated liver enzyme, lactate dehydrogenase, and creatinine concentrations. A diagnosis of thrombotic thrombocytopenic purpura was made. After a steady recovery, she was discharged from the hospital on the 16th postoperative day, but 12 hours later, she was readmitted with acute-onset progressively worsening shortness of breath. Echocardiography confirmed peripartum cardiomyopathy. She was treated with a bromocriptine and heart failure regimen. At 6 weeks postpartum, her laboratory test results and cardiac function had improved. Conclusions A possible autoimmune association might have caused both conditions in our patient. This case report serves as a warning message that pregnant women with one possible condition with autoimmune association could go on to develop other similar conditions
