    A Research of nasal methicillin resistant/sensitive Staphylococcus aureus and pharyngeal beta-haemolytic Streptococcus carriage in midwifery students in Kahramanmaras, Eastern Mediterranean Region of Turkey

    Background: In the hospitals where hygienic conditions are not provided, nasal methicillin resistant/sensitive Staphylococcus aureus (MRSA/MSSA) colonization is seen in the hospital personnel and patients. Both the individuals’ themselves being MRSA/MSSA carriers and also other people around them are under risk and this bacterium leads to nasal colonization and then can propagate through contaminated hands and hospital materials.Objective: In this study, it was aimed to research MRSA/MSSA and beta-haemolytic streptococcus (BHS) carriage in the nose and throat flora of students of, School of Health.Methods: In the study, total of 160 midwifery students taking midwifery education was included in the research but 135 (84.4 %) out of 160 students were able to be reached. The nose and throat swab samples taken from students were examined through microbiological methods.Results: 14.1 % of students were nasal MSSA carrier. While there wasn't the nasal MRSA carrier among students attending the study in the 1st and 2nd class, 3.3 % (1 person) of the 3rd class students and 10.0 % (3 people) of the 4th class students were the nasal MRSA carrier. In addition, BHS carriage was determined in 1.5 % of students and there wasn't any difference between classes.Conclusions: These results indicate that midwifery students might have been contaminated with MSSA/MRSA during clinical practice. That MSSA/MRSA being an important pathogen is seen in midwifery students taking education in the birth units threatens the mother-child health. [Ethiop. J. Health Dev. 2010;24(1):57-60

    New markers: Urine xanthine oxidase and myeloperoxidase in the early detection of urinary tract infection

The aim of this study was to evaluate if xanthine oxidase and myeloperoxidase levels quantitation method may alternate routine culture method, which takes more time in the diagnosis of urinary tract infections. Material and Methods. Five hundred and forty-nine outpatients who had admitted to Clinic Microbiology Laboratory were included in the study. The microorganisms were identified by using VITEK System. The urine specimens that were negative from the quantitative urine culture were used as controls. The activities of MPO and XO in spot urine were measured by spectrophotometric method. Results. Through the urine cultures, 167 bacteria were isolated from 163 urine specimens; 386 cultures yielded no bacterial growth. E. coli was the most frequent pathogen. In infection with E. coli both XO and MPO levels were increased the most. The sensitivity, specificity, positive predictive value, and negative predictive value for XO were 100%, 100%, 100%, and 100%, respectively. These values for MPO were 87%, 100%, 100%, and 94%, respectively. Conclusion. These data obtained suggest that urine XO and MPO levels may be new markers in the early detection of UTI. © 2014 Pinar Ciragil et al

    Evaluation of local trace element status and 8-Iso-prostaglandin F2? concentrations in patients with Tinea pedis

    Background: Tinea pedis (TP) is an infection of the feet caused by fungi. The infectious diseases caused by dermatophytes are mainly related to the enzymes produced by these fungi. Up to the now, the local 8-iso-prostaglandin F2? (8-iso-PGF2?), concentration as oxidative stress biomarker and trace elements status have not been published in patients with TP. The aim of this study is to evaluate the relationship between oxidative stress and trace elements (Cu, Zn, Se), and to evaluate the ratios of Cu/Zn and Cu/Se in this disorder. Methods: Forty-three consecutive patients with a diagnosis of unilateral interdigital TP were enrolled in this study. The samples were obtained by scraping the skin surface. 8-iso-PGF2? concentrations in scraping samples were determined by ELISA. In addition, the levels of Se, Zn and Cu in scraping samples were determined on flame and furnace atomic absorption spectrophotometer using Zeeman background correction. Results: Oxidative stress was confirmed by the significant elevation in 8-iso-PGF2? concentrations (p < 0.05). When compared to non-lesional area, Zn and Se levels were significantly lower on lesional area, whereas Cu levels was higher on the lesional area than the non-lesional area (p < 0.05). In addition, the correlation results of this study were firstly shown that there were significant and positive correlations between Cu and 8-iso-PGF2? parameters, but negative correlations between Se-Cu; Se-8-iso-PGF2? parameters in lesional area. Furthermore, the ratios of Cu/Zn and Cu/Se were significantly higher on the lesional area than the non-lesional area (p < 0.05). According to sex and fungal subtypes, there was no significant difference in the concentrations of 8-iso-PGF2? and trace elements in patients with TP (p > 0.05). Conclusions: Our results showed that there is a possible link between oxidative stress (increased 8-iso-PGF2? concentrations) and imbalanced of trace elements status in lesional area of TP patients. The use of antifungal agents together with both Zn and Se drugs could be helpful in the both regression of disease and in shortening the duration of disease. © 2016 Miraloglu et al

    Antiviral activity of hatay propolis against replication of herpes simplex virus type 1 and type 2

    PubMedID: 26856414Background: Propolis is a bee product widely used in folk medicine and possessing many pharmacological properties. In this study we aimed to investigate: i) the antiviral activities of Hatay propolis samples against HSV-1 and HSV-2 in HEp-2 cell line, and ii) the presence of the synergistic effects of propolis with acyclovir against these viruses. Material/Methods: All experiments were carried out in HEp-2 cell cultures. Proliferation assays were performed in 24-well flat bottom microplates. We inoculated 1×105 cells per ml and RPMI 1640 medium with 10% fetal calf serum into each well. Studies to determine cytotoxic effect were performed. To investigate the presence of antiviral activity of propolis samples, different concentrations of propolis (3200, 1600, 800, 400, 200, 100, 75, 50, and 25 µg/mL) were added into the culture medium. The amplifications of HSV-1 and HSV-2 DNA were performed by realtime PCR method. Acyclovir (Sigma, USA) was chosen as a positive control. Cell morphology was evaluated by scanning electron microscopy (SEM). Results: The replication of HSV-1 and HSV-2 was significantly suppressed in the presence of 25, 50, and 100 µg/mL of Hatay propolis. We found that propolis began to inhibit HSV-1 replication after 24 h of incubation and propolis activity against HSV-2 was found to start at 48 h following incubation. The activity of propolis against both HSV-1 and HSV-2 was confirmed by a significant decrease in the number of viral copies. Conclusions: We determined that Hatay propolis samples have important antiviral effects compared with acyclovir. In particular, the synergy produced by antiviral activity of propolis and acyclovir combined had a stronger effect against HSV-1 and HSV-2 than acyclovir alone. © Med Sci Monit, 2016

    The use of platensimycin and platencin to fight antibiotic resistance

    Infectious diseases are known as one of the most life-threatening disabilities worldwide. Approximately 13 million deaths related to infectious diseases are reported each year. The only way to combat infectious diseases is by chemotherapy using antimicrobial agents and antibiotics. However, due to uncontrolled and unnecessary use of antibiotics in particular, surviving bacteria have evolved resistance against several antibiotics. Emergence of multidrug resistance in bacteria over the past several decades has resulted in one of the most important clinical health problems in modern medicine. For instance, approximately 440,000 new cases of multidrug-resistant tuberculosis are reported every year leading to the deaths of 150,000 people worldwide. Management of multidrug resistance requires understanding its molecular basis and the evolution and dissemination of resistance; development of new antibiotic compounds in place of traditional antibiotics; and innovative strategies for extending the life of antibiotic molecules. Researchers have begun to develop new antimicrobials for overcoming this important problem. Recently, platensimycin - isolated from extracts of Streptomyces platensis - and its analog platencin have been defined as promising agents for fighting multidrug resistance. In vitro and in vivo studies have shown that these new antimicrobials have great potential to inhibit methicillin-resistant Staphylococcus aureus, vancomycin-resistant enterococci, and penicillin-resistant Streptococcus pneumoniae by targeting type II fatty acid synthesis in bacteria. Showing strong efficacy without any observed in vivo toxicity increases the significance of these antimicrobial agents for their use in humans. However, at the present time, clinical trials are insufficient and require more research. The strong antibacterial efficacies of platensimycin and platencin may be established in clinical trials and their use in humans for coping with multidrug resistance may be allowed in the foreseeable future. © 2013 Allahverdiyev et al, publisher and licensee Dove Medical Press Ltd

    The relationship between acute coronary artery diseases with c-reactive protein +1059 G/C and angiotensin-converting enzyme I/D gene polymorphisms

    Objective: The purpose of this study was to evaluate the presence of an association between the CRP +1059 G/C and ACE I/D gene polymorphisms and patients who were diagnosed to have acute coronary syndrome and underwent coronary angiography. Methods: A total of 126 patients (mean age: 60.0±12.9) and 144 healthy individuals (mean age: 52.1±13.0) were included to this study. The presence of CRP +1059 G/C and ACE I/D gene polymorphisms were analyzed using the RFLP method. Results: When the patient and control groups were evaluated in terms of ACE I/D gene polymorphism, no statistically significant difference was found in the frequency of ACE DD and ACE ID between the two groups (P>0.05), while the percentage of ACE II genotype was statistically significantly higher in the patient group compared with the control group (P0.05). When the presence of the ACE I/D genotype and CRP G/C genotype was compared in patients with vessel disease (one vessel, two vessels and three vessels) among the patients with coronary artery diseases with the control group, statistically significant differences were found between the two groups (P<0.05). In addition, the frequency of the ACE I/D genotype in hypertensive patients with coronary artery disease was statistically significantly higher (P<0.033). Also, the frequency of the CRP +1059 G/C genotype was found to be statistically significantly higher in the patient group (P<0.026). Conclusion: This study demonstrated that CRP +1059 G/C and ACE I/D gene polymorphisms may be a genetic marker associated with coronary artery disease in patients diagnosed with ACS. © 2016, E-Century Publishing Corporation.
