    The RNA-binding protein LARP1 is a post-transcriptional regulator of survival and tumorigenesis in ovarian cancer

    RNA-binding proteins (RBPs) are increasingly identified as post-transcriptional drivers of cancer progression. The RBP LARP1 is an mRNA stability regulator, and elevated expression of the protein in hepatocellular and lung cancers is correlated with adverse prognosis. LARP1 associates with an mRNA interactome that is enriched for oncogenic transcripts. Here we explore the role of LARP1 in epithelial ovarian cancer, a disease characterized by the rapid acquisition of resistance to chemotherapy through the induction of pro-survival signalling. We show, using ovarian cell lines and xenografts, that LARP1 is required for cancer cell survival and chemotherapy resistance. LARP1 promotes tumour formation in vivo and maintains cancer stem cell-like populations. Using transcriptomic analysis following LARP1 knockdown, cross-referenced against the LARP1 interactome, we identify BCL2 and BIK as LARP1 mRNA targets. We demonstrate that, through an interaction with the 3 untranslated regions (3 UTRs) of BCL2 and BIK, LARP1 stabilizes BCL2 but destabilizes BIK with the net effect of resisting apoptosis. Together, our data indicate that by differentially regulating the stability of a selection of mRNAs, LARP1 promotes ovarian cancer progression and chemotherapy resistance

    Integrated Controls-Structures Design Methodology for Flexible Spacecraft

    This paper proposes an approach for the design of flexible spacecraft, wherein the structural design and the control system design are performed simultaneously. The integrated design problem is posed as an optimization problem in which both the structural parameters and the control system parameters constitute the design variables, which are used to optimize a common objective function, thereby resulting in an optimal overall design. The approach is demonstrated by application to the integrated design of a geostationary platform, and to a ground-based flexible structure experiment. The numerical results obtained indicate that the integrated design approach generally yields spacecraft designs that are substantially superior to the conventional approach, wherein the structural design and control design are performed sequentially

    A class of stabilizing controllers for flexible multibody systems

    The problem of controlling a class of nonlinear multibody flexible space systems consisting of a flexible central body to which a number of articulated appendages are attached is considered. Collocated actuators and sensors are assumed, and global asymptotic stability of such systems is established under a nonlinear dissipative control law. The stability is shown to be robust to unmodeled dynamics and parametric uncertainties. For a special case in which the attitude motion of the central body is small, the system, although still nonlinear, is shown to be stabilized by linear dissipative control laws. Two types of linear controllers are considered: static dissipative (constant gain) and dynamic dissipative. The static dissipative control law is also shown to provide robust stability in the presence of certain classes of actuator and sensor nonlinearities and actuator dynamics. The results obtained for this special case can also be readily applied for controlling single-body linear flexible space structures. For this case, a synthesis technique for the design of a suboptimal dynamic dissipative controller is also presented. The results obtained in this paper are applicable to a broad class of multibody and single-body systems such as flexible multilink manipulators, multipayload space platforms, and space antennas. The stability proofs use the Lyapunov approach and exploit the inherent passivity of such systems

    Experimental validation of an integrated controls-structures design methodology for a class of flexible space structures

    This paper describes the first experimental validation of an optimization-based integrated controls-structures design methodology for a class of flexible space structures. The Controls-Structures-Interaction (CSI) Evolutionary Model, a laboratory test bed at Langley, is redesigned based on the integrated design methodology with two different dissipative control strategies. The redesigned structure is fabricated, assembled in the laboratory, and experimentally compared with the original test structure. Design guides are proposed and used in the integrated design process to ensure that the resulting structure can be fabricated. Experimental results indicate that the integrated design requires greater than 60 percent less average control power (by thruster actuators) than the conventional control-optimized design while maintaining the required line-of-sight performance, thereby confirming the analytical findings about the superiority of the integrated design methodology. Amenability of the integrated design structure to other control strategies is considered and evaluated analytically and experimentally. This work also demonstrates the capabilities of the Langley-developed design tool CSI DESIGN which provides a unified environment for structural and control design

    Solar Dynamics Observatory On-Orbit Jitter Testing, Analysis, and Mitigation Plans

    The Solar Dynamics Observatory (SDO) was designed to understand the Sun and the Sun s influence on Earth. SDO was launched on February 11, 2010 carrying three scientific instruments: the Atmospheric Imaging Assembly (AIA), the Helioseismic and Magnetic Imager (HMI), and the Extreme Ultraviolet Variability Experiment (EVE). Both AIA and HMI are sensitive to high frequency pointing perturbations and have sub-arcsecond level line-of-sight (LOS) jitter requirements. Extensive modeling and analysis efforts were directed in estimating the amount of jitter disturbing the science instruments. To verify the disturbance models and to validate the jitter performance prior to launch, many jitter-critical components and subassemblies were tested either by the mechanism vendors or at the NASA Goddard Space Flight Center (GSFC). Although detailed analysis and assembly level tests were performed to obtain good jitter predictions, there were still several sources of uncertainties in the system. The structural finite element model did not have all the modes correlated to test data at high frequencies (greater than 50 Hz). The performance of the instrument stabilization system was not known exactly but was expected to be close to the analytical model. A true disturbance-to-LOS observatory level test was not available due to the tight schedule of the flight spacecraft, the cost in time and manpower, difficulties in creating gravity negation systems, and risks of damaging flight hardware. To protect the observatory jitter performance against model uncertainties, the SDO jitter team devised several on-orbit jitter reduction plans in addition to reserve margins on analysis results. Since some of these plans severely restricted the capabilities of several spacecraft components (e.g. wheels and High Gain Antennas), the SDO team performed on-orbit jitter tests to determine which jitter reduction plans, if any, were necessary to satisfy science LOS jitter requirements. The SDO on-orbit jitter tests were designed to satisfy the following four objectives: 1. Determine the acceptable reaction wheel operational speed range during Science Mode. 2. Determine HGA algorithm jitter parameters (number of stagger steps and enable/disable no-steprequests). 3. Determine acceptable EVE instrument filter wheels spin rates. 4. Determine if AIA instrument filter wheels excite the first AIA telescope structural mode. This paper provides detailed information on the SDO wheel jitter test plan, shows on-orbit jitter measurements and how ground predictions compare to those measurements, and describes the final jitter mitigation plan executed on SDO

The design of the LISA spacecraft is also tightly coupled to the design and requirements of the scientific payload; for this reason it is often referred to as a "sciencecraft." Here we describe some of the unique features of the LISA spacecraft design that help create the quiet environment necessary for gravitational wave observations

    Multispectral Imaging of Organ Viability during Uterine Transplantation Surgery

    Uterine transplantation surgery has been proposed as a treatment for permanent absolute uterine factor infertility (AUFI) in the case of loss of the uterus. Due to the complexity of the vasculature correct reanastomosis of the blood supply during transplantation surgery is a crucial step to ensure reperfusion and viability of the organ. While techniques such as fluorescent dye imaging have been proposed to visualise perfusion there is no gold standard for intraoperative visualisation of tissue oxygenation. In this paper results from a liquid crystal tuneable filter (LCTF)-based multispectral imaging (MSI) laparoscope are described. The system was used to monitor uterine oxygen saturation (SaO) before and after transplantation. Results from surgeries on two animal models (rabbits and sheep) are presented. A feature-based registration algorithm was used to correct for misalignment induced by breathing or peristalsis in the tissues of interest prior to analysis. An absorption spectrum was calculated at each spatial pixel location using reflectance data from a reference standard, and the relative contributions from oxy- and deoxyhaemoglobin were calculated using a least squares regression algorithm with non-negativity constraints. Results acquired during animal surgeries show that cornual oxygenation changes are consistent with those observed in point measurements taken using a pulse oximeter, showing reduced SaO following reanastomosis. Values obtained using the MSI laparoscope were lower than those taken with the pulse oximeter, which may be due to the latter’s use of the pulsatile arterial blood signal. Future work incorporating immunological test results will help to correlate SaO levels with surgical outcomes

    Chemotherapy-induced apoptosis, autophagy and cell cycle arrest are key drivers of synergy in chemo-immunotherapy of epithelial ovarian cancer

    Epithelial ovarian cancer (EOC) is the most lethal of all gynecological malignancies in the UK. Recent evidence has shown that there is potential for immunotherapies to be successful in treating this cancer. We have previously shown the effective application of combinations of traditional chemotherapy and CAR (chimeric antigen receptor) T cell immunotherapy in in vitro and in vivo models of EOC. Platinum-based chemotherapy synergizes with ErbB-targeted CAR T cells (named T4), significantly reducing tumor burden in mice. Here, we show that paclitaxel synergizes with T4 as well, and look into the mechanisms behind the effectiveness of chemo-immunotherapy in our system. Impairment of caspase activity using pan-caspase inhibitor Z-VAD reveals this chemotherapy-induced apoptotic pathway as an essential factor in driving synergy. Mannose-6-phosphate receptor-mediated autophagy and the arrest of cell cycle in G2/M are also shown to be induced by chemotherapy and significantly contributing to the synergy. Increased expression of PD-1 on T4 CAR T cells occurred when these were in culture with ovarian tumor cells; on the other hand, EOC cell lines showed increased PD-L1 expression following chemotherapy treatment. These findings provided a rationale to look into testing PD-1 blockade in combination with paclitaxel and T4 immunotherapy. Combination of these three agents in mice resulted in significant reduction of tumor burden, compared to each treatment alone. In conclusion, the mechanism driving synergy in chemo-immunotherapy of EOC is multifactorial. A deeper understanding of such process is needed to better design combination therapies and carefully stratify patients
