    Chromosome segregation drives division site selection in Streptococcus pneumoniae.

    Accurate spatial and temporal positioning of the tubulin-like protein FtsZ is key for proper bacterial cell division. <i>Streptococcus pneumoniae</i> (pneumococcus) is an oval-shaped, symmetrically dividing opportunistic human pathogen lacking the canonical systems for division site control (nucleoid occlusion and the Min-system). Recently, the early division protein MapZ was identified and implicated in pneumococcal division site selection. We show that MapZ is important for proper division plane selection; thus, the question remains as to what drives pneumococcal division site selection. By mapping the cell cycle in detail, we show that directly after replication both chromosomal origin regions localize to the future cell division sites, before FtsZ. Interestingly, Z-ring formation occurs coincidently with initiation of DNA replication. Perturbing the longitudinal chromosomal organization by mutating the condensin SMC, by CRISPR/Cas9-mediated chromosome cutting, or by poisoning DNA decatenation resulted in mistiming of MapZ and FtsZ positioning and subsequent cell elongation. Together, we demonstrate an intimate relationship between DNA replication, chromosome segregation, and division site selection in the pneumococcus, providing a simple way to ensure equally sized daughter cells

    Arithmetic complexity via effective names for random sequences

    We investigate enumerability properties for classes of sets which permit recursive, lexicographically increasing approximations, or left-r.e. sets. In addition to pinpointing the complexity of left-r.e. Martin-L\"{o}f, computably, Schnorr, and Kurtz random sets, weakly 1-generics and their complementary classes, we find that there exist characterizations of the third and fourth levels of the arithmetic hierarchy purely in terms of these notions. More generally, there exists an equivalence between arithmetic complexity and existence of numberings for classes of left-r.e. sets with shift-persistent elements. While some classes (such as Martin-L\"{o}f randoms and Kurtz non-randoms) have left-r.e. numberings, there is no canonical, or acceptable, left-r.e. numbering for any class of left-r.e. randoms. Finally, we note some fundamental differences between left-r.e. numberings for sets and reals

    Medication Information Seeking Behavior in a Social Context: The Role of Lay and Professional Social Network Contacts

    This study provided a view of the social context of medication information seeking from a patient’s perspective.This was an exploratory qualitative study with 40 adults to determine how patients communicate within social networks to seek medication information. Semi-structured interviews were used to determine the structure (who), the content provided (what), and the function of social sources of information (how/why). Data underwent ethnographic content analysis using theory and prior research driven themes. Coding matrices were created to identify emerging patterns for who supplied what information and how the information was used. Participants described seeking medication information from health professional or lay social network sources. Health professional sources’ strongest role was to provide factual information. In contrast, lay sources provided factual information and affective information such as personal experiences and beliefs or attitudes. Information sought from social sources displayed similar functioning roles in terms of how the information was used by the participants seeking the information. The study concluded that medication information is sought from social sources both inside and outside of healthcare. Emerging patterns found that lay sources may provide patients more than affective information about medications. Further, patients may be receiving factually based information other than from health professionals. By coming to a more complete understanding of the social nature of the information environment, health professionals can better understand information needs from a patient’s perspective

    Conservation of the dark bee (Apis mellifera mellifera): estimating C-lineage introgression in Nordic breeding stocks

    Displacement and admixture are threatening the survival and genetic integrity of the European dark bee, Apis mellifera mellifera. Studies on the phenotype-genotype map and genotype by environment interactions in honey bees are demonstrating that variation at subspecies level exists and is worth conserving. SNP-based tools for monitoring genetic integrity in bees have been developed, but are not yet widely used by European dark bee breeders. We used a panel of ancestry informative SNP markers to assess the level of admixture in Nordic dark bee breeding stocks. We found that bee breeders falsely classified admixed stocks based on morphometry as purebred and vice versa. Even though most Nordic A. m. mellifera breeding stocks have low proportions of C-lineage ancestry, we recommend to incorporate genotyping in Nordic dark bee breeding programmes to ensure that minimal genetic diversity is lost, while the genetic integrity of the subspecies is maintained.We are greatly indebted to the participating bee breeders for allowing us to include their samples in this study and for providing the samples.     Eavesdropping and crosstalk between secreted quorum sensing peptide signals that regulate bacteriocin production in Streptococcus pneumoniae.

    Quorum sensing (QS), where bacteria secrete and respond to chemical signals to coordinate population-wide behaviors, has revealed that bacteria are highly social. Here, we investigate how diversity in QS signals and receptors can modify social interactions controlled by the QS system regulating bacteriocin secretion in Streptococcus pneumoniae, encoded by the blp operon (bacteriocin-like peptide). Analysis of 4096 pneumococcal genomes detected nine blp QS signals (BlpC) and five QS receptor groups (BlpH). Imperfect concordance between signals and receptors suggested widespread social interactions between cells, specifically eavesdropping (where cells respond to signals that they do not produce) and crosstalk (where cells produce signals that non-clones detect). This was confirmed in vitro by measuring the response of reporter strains containing six different blp QS receptors to cognate and non-cognate peptides. Assays between pneumococcal colonies grown adjacent to one another provided further evidence that crosstalk and eavesdropping occur at endogenous levels of signal secretion. Finally, simulations of QS strains producing bacteriocins revealed that eavesdropping can be evolutionarily beneficial even when the affinity for non-cognate signals is very weak. Our results highlight that social interactions can mediate intraspecific competition among bacteria and reveal that competitive interactions can be modified by polymorphic QS systems
