Effects of Friedreich's ataxia (GAA)(n)·(TTC)(n) repeats on RNA synthesis and stability

Expansions of (GAA)(n) repeats within the first intron of the frataxin gene reduce its expression, resulting in a hereditary neurodegenerative disorder, Friedreich's ataxia. While it is generally believed that expanded (GAA)(n) repeats block transcription elongation, fine mechanisms responsible for gene repression are not fully understood. To follow the effects of (GAA)(n)·(TTC)(n) repeats on gene expression, we have chosen E. coli as a convenient model system. (GAA)(n)·(TTC)(n) repeats were cloned into bacterial plasmids in both orientations relative to a promoter, and their effects on transcription and RNA stability were evaluated both in vitro and in vivo. Expanded (GAA)(n) repeats in the sense strand for transcription caused a significant decrease in the mRNA levels in vitro and in vivo. This decrease was likely due to the tardiness of the RNA polymerase within expanded (GAA)(n) runs but was not accompanied by the enzyme's dissociation and premature transcription termination. Unexpectedly, positioning of normal- and carrier-size (TTC)(n) repeats into the sense strand for transcription led to the appearance of RNA transcripts that were truncated within those repetitive runs in vivo. We have determined that these RNA truncations are consistent with cleavage of the full-sized mRNAs at (UUC)(n) runs by the E. coli degradosome

Issues of financial assurance of economy greening in the regions

The relevance of the analyzed issue is caused by the need to internalize environmental externalities in the modern world. The purpose of the article is to examine the issue of financial support of the green economy using the example of the regions included in the Volga Federal District (VFD). The leading methods to the study of this issue is a comparative analysis of environmental taxes and charges in Russia and countries of the European Union (EU) and the analysis of environmental problems in the regions of the Volga Federal District, which allow identifying the shortcomings of the existing environmental payment system and proposing measures to modernize the system of financial support of the green economy. The key shortcomings of the existing environmental payment system: low rates of environmental payments; the inappropriate use of funds received from environmental payments; concentration of 97% of revenues from environmental payments in the federal budget. The contents of the article may be helpful for public authorities of general and special competence to develop the principal directions of environmental policy and plan activities aimed at improving the green economy. © 2016 Shekhova et al

On the impact of nanometric γ’ precipitates on the tensile deformation of superelastic Co49Ni21Ga30

Results are presented reporting on the martensite domain variant selection and stress-induced martensite morphology in [001]-oriented superelastic Co49Ni21Ga30 shape memory alloy (SMA) single crystals under tensile load. In situ neutron diffraction, as well as in situ optical- and confocal laser scanning microscopy were conducted focusing on three differently treated samples, i.e. in the as-grown, solution-annealed and aged condition. An aging treatment performed at 350 °C promotes the precipitation of nanoprecipitates. These second phase precipitates contribute to an increase of the number of habit plane interfaces, while reducing lamellar martensite plate thickness compared to the as-grown and solution-annealed (precipitate free) samples. During tensile loading, all samples show a stress-induced formation of martensite, characterized by one single domain variant (“detwinned”) and one set of parallel habit planes in a shear band. The results clearly show that γ’ nanoprecipitates do not necessarily promote multi-variant interaction during tensile loading. Thus, reduced recoverability in Co-Ni-Ga SMAs upon aging cannot be solely attributed to this kind of interaction as has been proposed in literature so far
