    The Interchangeability of CVLT-II and WMS-IV Verbal Paired Associates Scores: A Slightly Different Story

    We investigated the similarity of the Wechsler Memory Scale-Fourth Edition (WMS-IV) Auditory Memory Index (AMI) scores when California Verbal Learning Test-Second Edition (CVLT-II) scores are substituted for WMS-IV Verbal Paired Associates (VPA) subtest scores. College students (n = 103) were administered select WMS-IV subtests and the CVLT-II in a randomized order. Immediate and delayed VPA scaled scores were significantly greater than VPA substitute scaled scores derived from CVLT-II performance. At the Index level, AMI scores were significantly lower when CVLT-II scores were used in place of VPA scores. It is important that clinicians recognize the accepted substitution of CVLT-II scores can result in WMS-IV scores that are inconsistent with those derived from standard administration. Psychometric issues that plausibly contribute to these differences and clinical implications are discussed

    Antibodies to actin in autoimmune haemolytic anaemia

    BACKGROUND: In autoimmune haemolytic anaemia (AIHA) autoreactive antibodies directed against red blood cells are upregulated leading to erythrocyte cell death. M. suis infections in pigs (IAP) induce AIHA of both the warm and the cold type. Aim of this study was to identify target autoantigens of warm autoreactive IgG antibodies. For this, sera from experimentally M. suis infected pigs were screened for autoreactivity. RESULTS: In sera of 95 % of all tested animals actin-reactive antibodies were found. The reactivity was shown to be species specific, i.e. reactivity with porcine actin was significantly higher than with rabbit-actin. Sera of animals previously immunised with the adhesion protein MSG1 of M. suis showed reactivity with actin prior to infection with M. suis indicating molecular mimicry to be involved in specific autoreactive mechanism. A potentially cross reactive epitope could be detected. CONCLUSIONS: This is the first report of autoreactive anti-actin antibodies involved in pathogenesis of autoimmune haemolytic anaemia

    Inorganic pyrophosphatase in uncultivable hemotrophic mycoplasmas: identification and properties of the enzyme from Mycoplasma suis

    BACKGROUND: Mycoplasma suis belongs to a group of highly specialized hemotrophic bacteria that attach to the surface of host erythrocytes. Hemotrophic mycoplasmas are uncultivable and the genomes are not sequenced so far. Therefore, there is a need for the clarification of essential metabolic pathways which could be crucial barriers for the establishment of an in vitro cultivation system for these veterinary significant bacteria.Inorganic pyrophosphatases (PPase) are important enzymes that catalyze the hydrolysis of inorganic pyrophosphate PPi to inorganic phosphate Pi. PPases are essential and ubiquitous metal-dependent enzymes providing a thermodynamic pull for many biosynthetic reactions. Here, we describe the identification, recombinant production and characterization of the soluble (s)PPase of Mycoplasma suis. RESULTS: Screening of genomic M. suis libraries was used to identify a gene encoding the M. suis inorganic pyrophosphatase (sPPase). The M. suis sPPase consists of 164 amino acids with a molecular mass of 20 kDa. The highest identity of 63.7% was found to the M. penetrans sPPase. The typical 13 active site residues as well as the cation binding signature could be also identified in the M. suis sPPase. The activity of the M. suis enzyme was strongly dependent on Mg2+ and significantly lower in the presence of Mn2+ and Zn2+. Addition of Ca2+ and EDTA inhibited the M. suis sPPase activity. These characteristics confirmed the affiliation of the M. suis PPase to family I soluble PPases. The highest activity was determined at pH 9.0. In M. suis the sPPase builds tetramers of 80 kDa which were detected by convalescent sera from experimentally M. suis infected pigs. CONCLUSION: The identification and characterization of the sPPase of M. suis is an additional step towards the clarification of the metabolism of hemotrophic mycoplasmas and, thus, important for the establishment of an in vitro cultivation system. As an antigenic and conserved protein the M. suis sPPase could in future be further analyzed as a diagnostic antigen

    Rare human skin infection with Corynebacterium ulcerans: transmission by a domestic cat

    Corynebacterium ulcerans is mainly known for its ability to cause animal infections. Some strains of C. ulcerans produce diphtheria toxin, which can cause life-threatening cardiopathies and neuropathies in humans. Human cutaneous C. ulcerans infection is a very rare disease that mimics classical cutaneous diphtheria. We present a very rare case of a C. ulcerans skin infection caused by a non-diphtheria toxin-producing strain of C. ulcerans that resolved after 3weeks of therapy with amoxicillin-clavulanate. A pet cat was the probable source of infection. The presence of C. ulcerans in the mouth of the cat was confirmed by 16S rRNA gene analysis and the API Coryne system. In cases of human infection with potentially toxigenic corynebacteria, it is important to determine the species and examine the isolate for diphtheria toxin production. If toxigenicity is present, diphtheria antitoxin should be administered immediately. Carriers and potential infectious sources of C. ulcerans include not only domestic livestock but also pet animals.     Twenty years of European mountain permafrost dynamics-the PACE legacy

    This paper reviews and analyses the past 20 years of change and variability of European mountain permafrost in response to climate change based on time series of ground temperatures along a south-north transect of deep boreholes from Sierra Nevada in Spain (37°N) to Svalbard (78°N), established between 1998 and 2000 during the EU-funded PACE (Permafrost and Climate in Europe) project. In Sierra Nevada (at the Veleta Peak), no permafrost is encountered. All other boreholes are drilled in permafrost. Results show that permafrost warmed at all sites down to depths of 50 m or more. The warming at a 20 m depth varied between 1.5 °C on Svalbard and 0.4 °C in the Alps. Warming rates tend to be less pronounced in the warm permafrost boreholes, which is partly due to latent heat effects at more ice-rich sites with ground temperatures close to 0 °C. At most sites, the air temperature at 2 m height showed a smaller increase than the near-ground-surface temperature, leading to an increase of surface offsets (SOs). The active layer thickness (ALT) increased at all sites between c. 10% and 200% with respect to the start of the study period, with the largest changes observed in the European Alps. Multi-temporal electrical resistivity tomography (ERT) carried out at six sites showed a decrease in electrical resistivity, independently supporting our conclusion of ground ice degradation and higher unfrozen water content

    Neuropsychological Outcomes of U.S. Veterans with Report of Remote Blast-Related Concussion and Current Psychopathology

    This study explored whether remote blast-related MTBI and/or current Axis I psychopathology contribute to neuropsychological outcomes among OEF/OIF veterans with varied combat histories. OEF/OIF veterans underwent structured interviews to evaluate history of blast-related MTBI and psychopathology and were assigned to MTBI (n = 18), Axis I (n = 24), Co-morbid MTBI/Axis I (n = 34), or post-deployment control (n = 28) groups. A main effect for Axis I diagnosis on overall neuropsychological performance was identified (F(3,100) = 4.81; p = .004), with large effect sizes noted for the Axis I only (d = .98) and Co-morbid MTBI/Axis I (d = .95) groups relative to the control group. The latter groups demonstrated primary limitations on measures of learning/memory and processing speed. The MTBI only group demonstrated performances that were not significantly different from the remaining three groups. These findings suggest that a remote history of blast-related MTBI does not contribute to objective cognitive impairment in the late stage of injury. Impairments, when present, are subtle and most likely attributable to PTSD and other psychological conditions. Implications for clinical neuropsychologists and future research are discussed.     Direct Metal Laser-sintered Stainless Steel: Comparison Of Microstructure And Hardness Between Different Planes

    Microstructural analysis and micro-hardness measurements were performed on different planes of 316L stainless steel fabricated by direct metal laser sintering (DMLS) technique. A fine cellular network was observed within the steel microstructure, where morphology of most cells changed from columnar on XZ-plane (vertical section) to equiaxed on XY-plane (horizontal section). Correspondingly, morphology of most grains was found to alter from columnar for the XZ-plane to equiaxed in the case of the XY-plane. Moreover, X-ray diffraction (XRD) analysis revealed a fully austenitic structure for both the planes. The average micro-hardness value for the XZ-plane and XY-plane was insignificantly (≈ 3%) different, which was attributed to the random grain orientation observed on both the planes. However, the average micro-hardness of the DMLS-fabricated 316L stainless steel in this contribution was approximately 25% higher than that of the as-cast one
