    High School Competitive Diving Injuries: National Athletic Treatment Injury and Outcomes Network (NATION)

    Purpose: Elite diving coaches and USA diving officials have become increasingly concerned about injury prevention among adolescent divers. However, little is known about such injuries. The purpose of this study was to describe the injuries among high school students who participated on high school diving teams. Subjects: High school students who participated on the diving teams of high schools that were included in the National Athletic Treatment, Injury and Outcomes Network (NATION) for 2011–2014. Methods: Descriptive epidemiology using injury exposure data on 56 boys\u27 Swimming and Diving teams and 55 girls\u27 Swimming and Diving teams from the National Athletic Treatment, Injury and Outcomes Network (NATION) for 2011–2014. Results: Only 12 injuries were reported, and 8 (67%) were concussions. The incidence of concussions was the same between boys and girls. Conclusion: Concussions are the highest reported injury among high school divers in the NATION data. Student athletes who had minor injuries may not have been evaluated by an athletic trainer. Researchers need better injury surveillance data for high school divers

    Video Meteor Fluxes

    The flux of meteoroids, or number of meteoroids per unit area per unit time, is critical for calibrating models of meteoroid stream formation and for estimating the hazard to spacecraft from shower and sporadic meteors. Although observations of meteors in the millimetre to centimetre size range are common, flux measurements (particularly for sporadic meteors, which make up the majority of meteoroid flux) are less so. It is necessary to know the collecting area and collection time for a given set of observations, and to correct for observing biases and the sensitivity of the system. Previous measurements of sporadic fluxes are summarized in Figure 1; the values are given as a total number of meteoroids striking the earth in one year to a given limiting mass. The Gr n et al. (1985) flux model is included in the figure for reference. Fluxes for sporadic meteoroids impacting the Earth have been calculated for objects in the centimeter size range using Super-Schmidt observations (Hawkins & Upton, 1958); this study used about 300 meteors, and used only the physical area of overlap of the cameras at 90 km to calculate the flux, corrected for angular speed of meteors, since a large angular speed reduces the maximum brightness of the meteor on the film, and radiant elevation, which takes into account the geometric reduction in flux when the meteors are not perpendicular to the horizontal. They bring up corrections for both partial trails (which tends to increase the collecting area) and incomplete overlap at heights other than 90 km (which tends to decrease it) as effects that will affect the flux, but estimated that the two effects cancelled one another. Halliday et al. (1984) calculated the flux of meteorite-dropping fireballs with fragment masses greater than 50 g, over the physical area of sky accessible to the MORP fireball cameras, counting only observations in clear weather. In the micron size range, LDEF measurements of small craters on spacecraft have been used to estimate the flux (Love & Brownlee, 1993); here the physical area of the detector is well known, but the masses depend strongly on the unknown velocity distribution. In the same size range, Thomas & Netherway (1989) used the narrow-beam radar at Jindalee to calculate the flux of sporadics. In between these very large and very small sizes, a number of video and photographic observations were reduced by Ceplecha (2001). These fluxes were calculated (details are given in Ceplecha, 1988) taking the Halliday et al. (1984) MORP fireball fluxes, slightly corrected in mass, as a calibration, and adjusting the flux of small cameras to overlap with the number/mass relation from that work

    DNA Methylation Patterns in Cord Blood of Neonates Across Gestational Age Association With Cell-Type Proportions

    Background: A statistical methodology is available to estimate the proportion of cell types (cellular heterogeneity) in adult whole blood specimens used in epigenome-wide association studies (EWAS). However, there is no methodology to estimate the proportion of cell types in umbilical cord blood (also a heterogeneous tissue) used in EWAS. Objectives: The objectives of this study were to determine whether differences in DNA methylation (DNAm) patterns in umbilical cord blood are the result of blood cell type proportion changes that typically occur across gestational age and to demonstrate the effect of cell type proportion confounding by comparing preterm infants exposed and not exposed to antenatal steroids. Methods: We obtained DNAm profiles of cord blood using the Illumina HumanMethylation27k BeadChip array for 385 neonates from the Boston Birth Cohort. We estimated cell type proportions for six cell types using the deconvolution method developed by Houseman et al. (2012). Results: The cell type proportion estimates segregated into two groups that were significantly different by gestational age, indicating that gestational age was associated with cell type proportion. Among infants exposed to antenatal steroids, the number of differentially methylated CpGs dropped from 127 to 1 after controlling for cell type proportion. Discussion: EWAS utilizing cord blood are confounded by cell type proportion. Careful study design including correction for cell type proportion and interpretation of results of EWAS using cord blood are critical

    Nemo phosphorylates Eyes absent and enhances output from the Eya-Sine oculis transcriptional complex during Drosophila retinal determination

    AbstractThe retinal determination gene network comprises a collection of transcription factors that respond to multiple signaling inputs to direct Drosophila eye development. Previous genetic studies have shown that nemo (nmo), a gene encoding a proline-directed serine/threonine kinase, can promote retinal specification through interactions with the retinal determination gene network, although the molecular point of cross-talk was not defined. Here, we report that the Nemo kinase positively and directly regulates Eyes absent (Eya). Genetic assays show that Nmo catalytic activity enhances Eya-mediated ectopic eye formation and potentiates induction of the Eya-Sine oculis (So) transcriptional targets dachshund and lozenge. Biochemical analyses demonstrate that Nmo forms a complex with and phosphorylates Eya at two consensus mitogen-activated protein kinase (MAPK) phosphorylation sites. These same sites appear crucial for Nmo-mediated activation of Eya function in vivo. Thus, we propose that Nmo phosphorylation of Eya potentiates its transactivation function to enhance transcription of Eya-So target genes during eye specification and development

    The Phylogeography of MERS-CoV in Hospital Outbreak-Associated Cases Compared to Sporadic Cases in Saudi Arabia.

    This study compared the phylogeography of MERS-CoV between hospital outbreak-associated cases and sporadic cases in Saudi Arabia. We collected complete genome sequences from human samples in Saudi Arabia and data on the multiple risk factors of human MERS-CoV in Saudi Arabia reported from 2012 to 2018. By matching each sequence to human cases, we identified isolates as hospital outbreak-associated cases or sporadic cases. We used Bayesian phylogenetic methods including temporal, discrete trait analysis and phylogeography to uncover transmission routes of MERS-CoV isolates between hospital outbreaks and sporadic cases. Of the 120 sequences collected between 19 June 2012 and 23 January 2017, there were 64 isolates from hospital outbreak-associated cases and 56 from sporadic cases. Overall, MERS-CoV is fast evolving at 7.43 Ă— 10-4 substitutions per site per year. Isolates from hospital outbreaks showed unusually fast evolutionary speed in a shorter time-frame than sporadic cases. Multiple introductions of different MERS-CoV strains occurred in three separate hospital outbreaks. MERS-CoV appears to be mutating in humans. The impact of mutations on viruses transmissibility in humans is unknown

    Complete Genomic Sequence of Bacteriophage B3, a Mu-Like Phage of \u3ci\u3ePseudomonas aeruginosa\u3c/i\u3e

    Bacteriophage B3 is a transposable phage of Pseudomonas aeruginosa. In this report, we present the complete DNA sequence and annotation of the B3 genome. DNA sequence analysis revealed that the B3 genome is 38,439 bp long with a G+C content of 63.3%. The genome contains 59 proposed open reading frames (ORFs) organized into at least three operons. Of these ORFs, the predicted proteins from 41 ORFs (68%) display significant similarity to other phage or bacterial proteins. Many of the predicted B3 proteins are homologous to those encoded by the early genes and head genes of Mu and Mu-like prophages found in sequenced bacterial genomes. Only two of the predicted B3 tail proteins are homologous to other well-characterized phage tail proteins; however, several Mu-like prophages and transposable phage D3112 encode approximately 10 highly similar proteins in their predicted tail gene regions. Comparison of the B3 genomic organization with that of Mu revealed evidence of multiple genetic rearrangements, the most notable being the inversion of the proposed B3 immunity/early gene region, the loss of Mu-like tail genes, and an extreme leftward shift of the B3 DNA modification gene cluster. These differences illustrate and support the widely held view that tailed phages are genetic mosaics arising by the exchange of functional modules within a diverse genetic pool

    Dominant genetic effects on corneal astigmatism: The genes in myopia (GEM) twin study

    Purpose: This study was conducted to assess the relative influence of genetics and environment on corneal astigmatism and corneal curvature in a large sample of twins. Methods: A total of 612 twin pairs (345 monozygotic [MZ] and 267 dizygotic [DZ]) aged between 18 and 86 years (mean age, 52.11 &#0177; 15.85 years) were recruited from the Australian Twin Registry (ATR). Each subject completed a general questionnaire, undertook a dilated eye examination, including ocular biometric measurements, and contributed a blood sample. Corneal astigmatism was defined as the absolute difference between the K1 and K2 meridians and corneal curvature as the average of K1 and K2. Results: Intrapair correlations were significantly higher (P < 0.001) in MZ twin pairs compared with those in DZ twin pairs for both corneal astigmatism (CA; r mz = 0.48 vs. r dz = 0.13) and corneal curvature (CC; r mz = 0.84 vs. r dz = 0.41). A sex-limited model with parameters estimating additive genetic, nonadditive genetic, and unique environmental influences (denoted ADE) was the most parsimonious model explaining both measures. Heritability estimates were as high as 60% and 71% for CA and CC, respectively. Conclusions: This study provides evidence that genetic factors explain interindividual variation in CA and CC, with nonadditive genetic factors explaining most of the variance due to those genetic factors. Heritability estimates were sex specific and indicate the need for future linkage studies for the identification of genes involved in the etiology of CA and CC

    Prediction of Core Body Temperature Based on Skin Temperature, Heat Flux, and Heart Rate Under Different Exercise and Clothing Conditions in the Heat in Young Adult Males

    Non-invasive, multi-parameter methods to estimate core body temperature offer several advantages for monitoring thermal strain, although further work is required to identify the most relevant predictor measures. This study aimed to compare the validity of an existing and two novel multi-parameter rectal temperature prediction models. Thirteen healthy male participants (age 30.9 ± 5.4 years) performed two experimental sessions. The experimental procedure comprised 15 min baseline seated rest (23.2 ± 0.3°C, 24.5 ± 1.6% relative humidity), followed by 15 min seated rest and cycling in a climatic chamber (35.4 ± 0.2°C, 56.5 ± 3.9% relative humidity; to +1.5°C or maximally 38.5°C rectal temperature, duration 20–60 min), with a final 30 min seated rest outside the chamber. In session 1, participants exercised at 75% of their heart rate maximum (HR max) and wore light athletic clothing (t-shirt and shorts), while in session 2, participants exercised at 50% HR max, wearing protective firefighter clothing (jacket and trousers). The first new prediction model, comprising the input of 18 non-invasive measures, i.e., insulated and non-insulated skin temperature, heat flux, and heart rate (“Max-Input Model”, standard error of the estimate [SEE] = 0.28°C, R2 = 0.70), did not exceed the predictive power of a previously reported model which included six measures and no insulated skin temperatures (SEE = 0.28°C, R2 = 0.71). Moreover, a second new prediction model that contained only the two most relevant parameters (heart rate and insulated skin temperature at the scapula) performed similarly (“Min-Input Model”, SEE = 0.29, R2 = 0.68). In conclusion, the “Min-Input Model” provided comparable validity and superior practicality (only two measurement parameters) for estimating rectal temperature versus two other models requiring six or more input measures

    The intestinal virome in children with cystic fibrosis differs from healthy controls.

    Intestinal bacterial dysbiosis is evident in children with cystic fibrosis (CF) and intestinal viruses may be contributory, given their influence on bacterial species diversity and biochemical cycles. We performed a prospective, case-control study on children with CF and age and gender matched healthy controls (HC), to investigate the composition and function of intestinal viral communities. Stool samples were enriched for viral DNA and RNA by viral extraction, random amplification and purification before sequencing (Illumina MiSeq). Taxonomic assignment of viruses was performed using Vipie. Functional annotation was performed using Virsorter. Inflammation was measured by calprotectin and M2-pyruvate kinase (M2-PK). Eight CF and eight HC subjects were included (50% male, mean age 6.9 ± 3.0 and 6.4 ± 5.3 years, respectively, p = 0.8). All CF subjects were pancreatic insufficient. Regarding the intestinal virome, no difference in Shannon index between CF and HC was identified. Taxonomy-based beta-diversity (presence-absence Bray-Curtis dissimilarity) was significantly different between CF and HC (R2 = 0.12, p = 0.001). Myoviridae, Faecalibacterium phage FP Taranis and unclassified Gokushovirinae were significantly decreased in CF compared with HC (q<0.05). In children with CF (compared to HC), the relative abundance of genes annotated to (i) a peptidoglycan-binding domain of the peptidoglycan hydrolases (COG3409) was significantly increased (q<0.05) and (ii) capsid protein (F protein) (PF02305.16) was significantly decreased (q<0.05). Picornavirales, Picornaviridae, and Enterovirus were found to positively correlate with weight and BMI (r = 0.84, q = 0.01). Single-stranded DNA viruses negatively correlated with M2-PK (r = -0.86, q = 0.048). Children with CF have an altered intestinal virome compared to well-matched HC, with both taxonomic and predicted functional changes. Further exploration of Faecalibacterium phages, Gokushovirinae and phage lysins are warranted. Intestinal viruses and their functions may have important clinical implications for intestinal inflammation and growth in children with CF, potentially providing novel therapeutic targets
