Effects of letrozole, non-steroidal aromatase inhibitor, on serum sex steroid levels, oocyte diameter and gonadosomatic index (GSI) in rainbow trout (Oncorhynchus mykiss)
The objective of this study was to contribute to investigate the efficacy of letrozole, a potent non-steroidal aromatase inhibitor, on serum sex steroid levels and oocyte growth in rainbow trout females. Serum steroid levels were measured using an indirect enzyme-linked immunosorbent assay (ELISA) before and 22 days after an initial injection with 1, 2.5 mg kg^-1 and subsequent weekly injection with 2.5 mg kg^-1 letrozole. After the last blood collection, gonad tissues were removed and oocyte diameter and GSI were determined. In all groups treated with letrozole, serum 17β-esteradiol levels (E2) decreased significantly and inversely serum 17α-20ß dihydroxy-4 pregnen-3-one (17α-20ß-P), testosterone (T) levels increased significantly from 6 to 96 h after injection (p<0.05). The lowest E2 levels, oocyte diameter and GSI were measured in the group treated weekly with 2.5 mg kg^-1 letrozole (p<0.05). This study demonstrates that letrozole has the potential to interfere in the synthesis of endogenous estrogens from androgens and delay gonadal development
Growth performance and digestive enzymes activities of Pacific white leg shrimp (Litopenaeus vannamei) juveniles fed dietary mixtures of four medicinal plants
In present study the effect of dietary inclusion of equal amounts of four traditional medicines; Matricaria chamomilla L, Zataria multiflora L, Mentha piperita L and Terminalia chebulo L on growth performance and digestive enzymes activity in Litopenaeus vannamei was investigated. Two diets, including a control basal diet and an experimental diet with 30 g kg^-1 herbal mixture supplementation were prepared. Juveniles (with average weight of 2.63±0.11 g) were fed control and supplemented diet for 60 days. At the end of the experiment, shrimp fed the experimental diet showed significant increase in SGR, WGR, compared with those of the control group. Shrimp fed with herbal mixture supplementation revealed significantly higher lipase, protease and amylase activities as compared with the control group in 60 days. The results indicated that using equal mixtures of four medicinal plants in L. vannamei diets can improve the growth parameters and digestive enzyme activities
Mortality Risk Related to Heatwaves in Dezful City, Southwest of Iran
Background: We aimed to evaluate the impact of heatwaves on daily deaths due to non-accidental, cardiovascular and respiratory causes in the city of Dezful in Iran from 2013 to 2019. Method: We collected daily ambient temperature and mortality and defined 2 types of heatwaves by combining daily temperature ⩾90th in each month of the study period or since 30 years with duration ⩾2 and 3 days. We used a distributed lag non-linear model to study the association between each type of heatwave definition, and deaths due to non-accidental, cardiovascular and respiratory causes with lags up to 13 days. Results: There was no discernible correlation in this area, despite the fact that heatwaves raised the risk of death from cardiovascular causes and lowered the risk from respiratory causes. On the other hand, the risk of total non-accidental mortality on days with the heatwaves is significantly higher than normal days. In main effects, the heatwaves have a significant relationship with the risk of total non-accidental mortality (in the first heatwave definition, Cumulative Excess Risk (CER) in lag0-2 was 10.4 and in second heatwave definition, CER values in lag0, 0-2, and 0-6 were 12.4, 29.2, and 38.8 respectively). Also, in added effects, heatwaves have a significant relationship with the risk of total non-accidental mortality (in the first heatwave definition, CER in lag0 and 0-2 were 1.79 and 4.11 and in the second heatwave definition, CER values in lag0, 0-2, and 0-6 were 7.76, 18.35 and 24.87 respectively). In addition, heatwaves appeared to contribute to a cumulative excess risk of non-accidental death among the male group as well as the older adults. Conclusion: However, the results showed that heatwaves could have detrimental effects on health, even in populations accustomed to the extreme heat. Therefore, early warning systems which monitor heatwaves should provide the necessary warnings to the population, especially the most vulnerable groups
Effect of galacto oligosaccharide prebiotic on growth performance, survival and several of innate immunity parameters of rainbow trout (Oncorhynchus mykiss) larvae
Prebiotics are non-digestive food ingredients which beneficially affect the host by selectively stimulating the growth and activity of health-promoting bacteria in the intestinal tract. This experiment was conducted to evaluate the effect of dietary supplementation of galactooilgosaccharide on the growth performances, survival and several of innate immunity parameters of rainbow trout (Oncorhynchus mykiss) larvae for 60 days. In this experiment, 360 of larvae (with average weight of 0.13±0.01g) in 4 treatments and 3 replicates (n=30 in each replicate) in a completely randomized design was divided and fed with diets containing 0, 1, 2 and 3 percent galactooligosaccharide respectively. The present results showed that highest WG, SGR and PER were observed in the diet containing 2 and 3 percent of dietary galactoologosaccharide which had a significant difference compared with other treatments (p<0.05). The highest, survival, total protein, albumin and globulin levels were observed in treatments containing 2 percent of dietary galactooligosaccharide which showed a significant difference compared with other treatments (p<0.05). Finally, the present results suggest that diets containing 2 percent of dietary galactooligosaccharide could improve growth, survival and innate immunity responses of rainbow trout
Differentiation-specific action of orphan nuclear receptor NR5A1 (SF-1): transcriptional regulation in luteinizing bovine theca cells
BACKGROUND: The orphan nuclear receptor NR5A1 (steroidogenic factor-1, SF-1) is a master regulator of tissue-specific gene expression in reproductive and steroidogenic tissues. Two activating functions, AF-1 and AF-2, have been described to function in a cooperative manner to recruit transcriptional coactivators to the promoter regions of NR5A1-controlled genes. METHODS: The role of the NR5A1 activating functions AF-1 and AF-2 was studied in primary bovine theca cells. Bovine theca cells were infected with recombinant adenovirus vectors over-expressing wild-type NR5A1 or NR5A1 mutants, in which one of the activating functions of this orphan nuclear receptor had been impaired. Under different culture conditions, theca cell-specific transcript levels were measured by reverse transcription and real-time PCR. RESULTS: Under culture conditions optimized for cell growth, transcriptional up-regulation of CYP11A1 (P450 side chain-cleavage enzyme) and INSL3 (Insulin-like factor 3, Relaxin-like factor (RLF)) was found to be dependent on the presence of NR5A1 carrying an intact AF-2. Under conditions inducing luteal differentiation of theca cells, CYP11A1 and STAR (Steroidogenic acute regulatory protein) were up-regulated by the action of luteinizing hormone (LH), whereas the differentiation-specific up-regulation of INSL3 was suppressed by LH in luteinizing theca cells. Inhibition of insulin- or IGF1- (insulin-like growth factor I) dependent signal transduction by the RAF1 kinase inhibitor GW5074 and the mitogen-activated protein kinase kinase inhibitor PD98059 resulted in the finding that RAF1 kinase inhibition was able to counteract the LH-dependent regulation of NR5A1-controlled genes, whereas inhibition of the mitogen-activated protein kinase (MAP kinase) pathway did not have any significant effect. CONCLUSION: The regulation of the three NR5A1-controlled genes CYPA11, STAR, and INSL3 in luteinizing theca cells apparently is not dependent on NR5A1 activating functions AF-1 or AF-2. Activation of AF-1 here even appears to have an impairing effect on NR5A1 transcriptional activity, implying that up-regulation of NR5A1-controlled genes uses a different pathway. Our results might be explained by the possible existence of an interconnection between the RAF1 kinase and the cyclic AMP-protein kinase A pathway. Such a non-classical regulatory pathway might play an important role in the control of gene expression in reproductive and steroidogenic tissues.Norbert Walther, Martina Jansen, Wasima Akbary and Richard Ivel
Assessment of outer hair cell function and blood antioxidant status of rabbits exposed to noise and metal welding fumes
Objectives: To investigate the interaction between welding fumes and noise in causation of hearing impairment. Methods: Groups of rabbits (n = 6) were exposed to noise, welding fumes or combination of both prior to Distortion Product Otoacoustic-Emissions (DPOAEs) analysis. The function of outer hair cells (OHCs) was examined by DPOAE assessment over a broad range of frequencies. Variations in DPOAE amplitude were compared between control (n = 6) and exposed (n = 18) groups. Results: The DPOAEs levels measured at different frequencies (1379-6299 Hz) were found to decrease significantly (P < 0.05) in rabbits exposed to 110 dB sound pressure level (SPL) broadband noise (8 h/day, 12 days). In rabbits, exposed to carbon-steel welding fumes alone (157 mg/m3), the threshold shift was limited to the high frequencies (2759-6299 Hz), whereas, mixed exposure to noise and fumes resulted in reduction of DPOAEs at all the frequencies. Changes in DPOAEs were associated with increased susceptibility of erythrocytes to oxidation (P < 0.05). Exposure to noise or fumes alone or simultaneously, suppressed total antioxidant ability of plasma as measured by ferric reducing ability of plasma (FRAP). Noise alone or in combination with fumes resulted in depletion of blood glutathione (GSH). Despite suppression of FRAP in the exposed groups, GSH was found to remain unchanged due to welding fumes suggesting that antioxidants other than GSH are affected by toxicants present in metal welding fumes. Conclusion: Exposure to very high levels of welding fumes can increase noise-related effects on OHC function by extending hearing threshold shift to wide band frequencies. © 2006 Elsevier Ireland Ltd. A Population Pharmacokinetic Analysis of Milrinone in Pediatric Patients After Cardiac Surgery
The purpose of this study was to ascertain the optimal pharmacokinetic model for milrinone in pediatric patients after cardiac surgery when milrinone was administered as a slow loading dose followed by a constant-rate infusion. The data used for pharmacokinetic analysis were collected in a prospective, randomized, placebo-controlled multi-center trial of milrinone as prophylaxis for the development of low cardiac output syndrome after surgery for repair of complex congenital cardiac defects. Two blood samples were randomly collected from each patient for determination of plasma milrinone concentrations with subsequent population pharmacokinetic modeling. The pharmacokinetics of milrinone in pediatric patients under 6year's age were best described by a weight-normalized one compartment model after a slow loading dose followed by a constant-rate infusion. The volume of distribution was 482mlkg −1 , and was independent of age. Clearance was a linear function of age given by Cl=2.42mlkg −1 min −1 [1+0.0396*age].Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/45063/1/10928_2004_Article_485925.pd

Robust estimation of bacterial cell count from optical density
Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data
