    Eradication of Helicobacter pylori usually consists of a 7-day course of triple therapy including metronidazole or amoxicillin plus clarithromycin plus a proton pump inhibitor. We report about a rare adverse event of Hp eradication in a patient with moderate chronic and moderate active pangastritis. Shortly after the end of treatment cholestatic hepatitis occurred which was most likely related to clarithromycin, perhaps enhanced by amoxicillin. Since liver dysfunction was self-limited, no further treatment was required. In summary, clinicians should be aware about the presented rare adverse event of Helicobacter pylori eradication treatment for a close monitoring of those patients and rapid management of acute liver failure

    Regaining momentum for international climate policy beyond Copenhagen

    The 'Copenhagen Accord' fails to deliver the political framework for a fair, ambitious and legally-binding international climate agreement beyond 2012. The current climate policy regime dynamics are insufficient to reflect the realities of topical complexity, actor coalitions, as well as financial, legal and institutional challenges in the light of extreme time constraints to avoid 'dangerous' climate change of more than 2°C. In this paper we analyze these stumbling blocks for international climate policy and discuss alternatives in order to regain momentum for future negotiations

    An arrhythmogenic metabolite in atrial fibrillation

    Abstract Background Long-chain acyl-carnitines (ACs) are potential arrhythmogenic metabolites. Their role in atrial fibrillation (AF) remains incompletely understood. Using a systems medicine approach, we assessed the contribution of C18:1AC to AF by analysing its in vitro effects on cardiac electrophysiology and metabolism, and translated our findings into the human setting. Methods and results Human iPSC-derived engineered heart tissue was exposed to C18:1AC. A biphasic effect on contractile force was observed: short exposure enhanced contractile force, but elicited spontaneous contractions and impaired Ca2+ handling. Continuous exposure provoked an impairment of contractile force. In human atrial mitochondria from AF individuals, C18:1AC inhibited respiration. In a population-based cohort as well as a cohort of patients, high C18:1AC serum concentrations were associated with the incidence and prevalence of AF. Conclusion Our data provide evidence for an arrhythmogenic potential of the metabolite C18:1AC. The metabolite interferes with mitochondrial metabolism, thereby contributing to contractile dysfunction and shows predictive potential as novel circulating biomarker for risk of AF

    The Rabbitfish Siganus virgatus as Key Macroalgae Browser in Coral Reefs of the Gulf of Thailand

    Coral reef resilience is greatly influenced by herbivory. There is a need to identify key fish species fulfilling this critical function in biogeographically distinct regions. This experimental in situ study investigated fish herbivory in coral reefs of the lower Gulf of Thailand characterized by a considerably low herbivorous fish biomass and diversity, but high live coral and low macroalgal cover. This provided an intriguing situation for macroalgal browsing research. Visual census techniques assessed the abundance of local herbivorous fish species, and filmed single-choice assays using the macroalga Turbinaria evaluated mass-standardized bites (ms-bites) and biomass removal. Multiple-choice assays offering four locally abundant macroalgae identified specific biomass removal and ms-bites to uncover selection and avoidance patterns of observed fish species. The rabbitfish Siganusvirgatus constituted only 39% of herbivore biomass but accounted for 90% of ms-bites. In multiple-choice assays, fishes took most (61%) bites on Sargassum, followed by Padina (28%) and Turbinaria (11%), while Lobophora was avoided. S. virgatus exhibited the most generalized browsing pattern of all species observed. Coinciding with recent studies, our findings suggest that S. virgatus plays a key functional role in reefs characterized by low diversity of herbivores and low functional redundancy

    Farnesoid X receptor activation by bile acids suppresses lipid peroxidation and ferroptosis

    Abstract Ferroptosis is a regulated cell death modality that occurs upon iron-dependent lipid peroxidation. Recent research has identified many regulators that induce or inhibit ferroptosis; yet, many regulatory processes and networks remain to be elucidated. In this study, we performed a chemical genetics screen using small molecules with known mode of action and identified two agonists of the nuclear receptor Farnesoid X Receptor (FXR) that suppress ferroptosis, but not apoptosis or necroptosis. We demonstrate that in liver cells with high FXR levels, knockout or inhibition of FXR sensitized cells to ferroptotic cell death, whereas activation of FXR by bile acids inhibited ferroptosis. Furthermore, FXR inhibited ferroptosis in ex vivo mouse hepatocytes and human hepatocytes differentiated from induced pluripotent stem cells. Activation of FXR significantly reduced lipid peroxidation by upregulating the ferroptosis gatekeepers GPX4, FSP1, PPARα, SCD1, and ACSL3. Together, we report that FXR coordinates the expression of ferroptosis-inhibitory regulators to reduce lipid peroxidation, thereby acting as a guardian of ferroptosis
