A critical role of the transcription factor Miz-1 in T cell activation and maturation
A functional immune system is important to protect the organism against various pathogens and to induce an adequate response to vaccinations. B and T lymphocytes are the main drivers of adaptive immunity. An important regulator of their development is the MYC-interacting zinc finger protein 1 (Miz-1). The transcription factor Miz-1 is highly necessary for adequate T cell development by restricting apoptosis during V(D)J recombination. However, the importance of Miz-1 in mature T cells is yet not fully understood. To provide new insights into the role of Miz-1 for T lymphocytes, mice possessing a conditional knockout of Miz-1 either in all hematopoietic cells (Vav-Cre x Miz-1fl/fl) or specifically in T cells (CD4 Cre x Miz-1fl/fl) were used. As the POZ domain of Miz-1 is necessary for its transcriptional activity, this domain was targeted by Cre-mediated deletion in both mouse models. Phenotypical analysis of these mice showed severe perturbations of the peripheral T cells, resulting in a reduction of mature T cells in several secondary lymphoid organs. Likewise, this decrease was observed in CD4+ as well as in CD8+ T cells. T cell activation experiments showed that Miz-1 controls proliferation as well as apoptosis of peripheral T cells. Miz-1 deficiency resulted in a faster proliferation but also an excessive induction of apoptosis. Furthermore, the process of apoptosis seems to be p53-independent and induced via the extrinsic apoptosis pathway. Flow cytometry experiments revealed that Miz-1 is a novel regulator of T cell aging, since young mice possessing a functional Miz-1 deletion exhibited similar distributions of T cell populations as old control mice. Due to Miz-1 deficiency, the numbers of naĂŻve T cells dropped while frequencies of memory T cells were increased. Thus, Miz-1 is necessary to maintain the homeostasis of naĂŻve and memory T cells in the course of aging
Der Zusammenhang von kultureller Distanz, kultureller Anpassung und beruflichem Belastungserleben: Eine Studie zum Gelingen der Entsendung von Lehrkräften im deutschen Auslandsschulwesen
Vor dem Hintergrund theoretischer Annahmen zu kultureller Anpassung bei Migrationserfahrungen in kulturell distanten und proximalen Kulturen werden die Befunde einer empirischen Studie bei N = 426 Auslandslehrkräften berichtet. Die Untersuchung versuchte die Frage zu beantworten, ob kulturelle Anpassung eher in proximalen Kulturen gelingt und ob sich berufliche Belastung und berufliche und allgemeine Zufriedenheitswerte bei Lehrkräften in Abhängigkeit von der Gastkultur zeigen lassen. Der Zusammenhang zwischen kultureller Anpassung und Zufriedenheit sowie zwischen kultureller Anpassung und Belastungserleben ließ sich nachweisen. Media competency and subject specific didactics
Die Frage, auf welche Weise medienerzieherische Inhalte am sinnvollsten in den Unterricht zu integrieren sind, wird sehr unterschiedlich beantwortet. Im Gegensatz zum fächerverbindenden Unterricht oder projektorientierten Konzept wird hier eine enge Verknüpfung mit den Didaktiken der Unterrichtsfächer favorisiert. Sie könnte eine Chance sein, der Medienerziehung bei der Unterrichtsplanung der Lehrkräfte einen höheren Rang einzuräumen, um Kindern medienbezogenes Handlungswissen und übertragbares Strukturwissen zu vermitteln. Building urban resilience through spatial planning following disasters
It is expected that the changing temperatures and rising sea levels caused through global climate change will result in an aggravation of disaster risks (e.g. from heat stress, storms, flooding, landslides, air pollution, drought and water scarcity), particularly in urban areas (IPCC, 2014). This trend is exaggerated by the continuing trend of urbanization, which is projected to result in 66% of the people worldwide to live in cities in 2050 (United Nations, Department of Economic and Social Affairs, Population Division, 2014, p. 1). It is an important obligation of spatial planners to facilitate that this urban development follows certain standards, including safe housing and the provision of basic utilities and infrastructure as well as adequate health services (PreventionWeb, 2015; United Nations, Department of Economic and Social Affairs, Population Division, 2014).
Resilience is one of the key concepts to address these challenges. If a city is resilient, it is able to “resist, absorb, accommodate to and recover from the effects of a hazard in a timely and efficient manner, including through the preservation and restoration of its essential basic structures and functions” (UNISDR, 2009, p. 24). The adjustment of existing urban structures is time-consuming and cost-intensive; therefore, a resilient city can most efficiently be achieved when it is first developed (UN-Habitat, 2015) or after a disaster has erased the previously existing city structures (Olshansky, Hopkins, & Johnson, 2012). The time frame after the disaster can be considered as a window of opportunity for planners to build the city back better or – in other words – to build a resilient city. Although the relevance of spatial planners for the construction of resilient cities is obvious, there is little knowledge of spatial planning’s capabilities to achieve this goal so far. Drawing from experiences on the reconstruction process after the Great East Japan Earthquake and Tsunami 2011 in Japan’s Miyako City and Ishinomaki City, this dissertation addresses this topic. It explains which of the local spatial planning options can be used to build resilience and how the toolkit of spatial planners can be improved in order to be more efficient to build urban resilience. Even through these spatial planning options differ from country to country and the focus of this work on Japan only enables a limited transferability of the research results, the experiences from Tohoku Region are able to contribute to the ongoing discussion about spatial planning and urban resilience after disasters

Standards und Indikatoren fĂĽr die Entwicklung von Kompetenzen in der Lehrerbildung
In diesem Papier werden für die Schulpraktika im Lehramtsstudium, für das Referendariat und schließlich für die professionelle Lehrkraft Handlungsstandards in sechs zentralen Dimensionen dargestellt: 1) Handeln als Lehrperson 2) die Schule als Lern- und Lebensraum 3) Unterrichtsbeobachtung und –evaluation 4) Unterrichtsplanung, -durchführung und –analyse 5) Klassenführung 6) Leistungsmessung In einer spiralcurricularen Darstellung wird der Weg der professionellen Entwicklung, beginnend bei den ersten Handlungserfahrungen als Lehrperson, anhand von Regelstandards entwickelt. Dazu werden jeweils Indikatoren aufgeführt, an denen man die Erreichung des jeweiligen Standards beobachten kann. Ein theoretischer Teil führt in die erziehungswissenschaftliche Diskussion rund um das Thema Lehrerbildungsstandards ein und verankert die vorliegenden sowie die dazugehörigen Indikatoren in der aktuellen Unterrichtsqualitätsforschung
Gesundheit und Wohlbefinden von Kindern und Jugendlichen in St. Leonhard und Schweinau: Befunde aus einem Lehrforschungsprojekt in NĂĽrnberg
"Der vorliegende Bericht enthält Ergebnisse aus einem im Studienjahr 2008/09 durchgeführten Lehrforschungsprojekt über die Lebensqualität von Kindern und Jugendlichen in zwei sozialstrukturell und -ökologisch benachteiligten Stadtteilen Nürnbergs. Im Vordergrund stehen dabei schriftliche Befragungen von SchülerInnen und Eltern über Gesundheit und Wohlbefinden. Die damit erhobenen Daten für n=924 Grund- und Hauptschulkinder werden vorwiegend nach Schulform und Geschlechtszugehörigkeit analysiert. Darüber hinaus steht die Berichterstattung der in Nürnberg erscheinenden Presse über die beiden Stadtteile St. Leonhard und Schweinau im Fokus.
