    Increased circulating obestatin in patients with chronic obstructive pulmonary disease

    BACKGROUND: Some peptides, which regulate the metabolic balance, are thought to play important roles in nutritional disorders and systemic inflammation in COPD. Treatment of rats with obestatin decreased body-weight gain. Obestatin was also found to be correlated with inflammation in rheumatoid arthritis. The aims of this study were to investigate the level of circulating obestatin in COPD and to analyze the relationship among obestatin and nutritional status, and systemic inflammation. METHODS: 32 COPD patients with BMI less than 20 kg/m(2) and 22 normal controls were included. Body composition was estimated using “foot-to-foot” BIA technology. Circulating obestatin was determined with enzyme-linked immunosorbent assay. Pulmonary function, TNF-α and C reactive protein were also measured. RESULTS: The level of circulating obestatin was higher in COPD with underweight than that in normal control (5562.75 ± 3435.43 pg/ml in COPD, 3663.90 ± 2313.95 pg/ml in controls, p = 0.028). BMI, Waist circumference, hip circumference, bodyFAT and FAT% in COPD group were lower than those in normal control. Positive correlation was found among circulating C reactive protein, TNF-α and obestatin. There was no significant correlation among BMI, pulmonary function and obestatin. CONCLUSIONS: This study shows that circulating obestatin is higher in underweight COPD patients, and positively correlated to systemic inflammation, but not to nutritional status

    High Viral Load of Human Bocavirus Correlates with Duration of Wheezing in Children with Severe Lower Respiratory Tract Infection

    Background: Human bocavirus (HBoV) is a newly discovered parvovirus and increasing evidences are available to support its role as an etiologic agent in lower respiratory tract infection (LRTI). The objective of this study is to assess the impact of HBoV viral load on clinical characteristics in children who were HBoV positive and suffered severe LRTI. Methods: Lower respiratory tract aspirates from 186 hospitalized children with severe LRTI were obtained by bronchoscopy. HBoVs were detected by real-time PCR and other 10 infectious agents were examined using PCR and/or direct fluorescent assay. Results: Thirty-one patients (24.6%) were tested positive for HBoV in the respiratory tract aspirates. Fifteen samples had a high viral load (.10 4 copies/mL) and the other sixteen samples had a low viral load (,10 4 copies/mL). The duration of presented wheezing and hospitalization was longer in children with high viral load of HBoV than that in children with low viral load. The days of wheezing showed a correlation with viral load of HBoV. Conclusion: We confirmed that HBoV was frequently detected in patients with severe LRTI. Wheezing was one of the most common symptoms presented by patients with positive HBoV. A high HBoV viral load could be an etiologic agent for LRTI

    Increased IL-10 mRNA expression in tumor-associated macrophage correlated with late stage of lung cancer

    <p>Abstract</p> <p>Background</p> <p>Monocyte recruited into the tumor and maturation to tumor-associated macrophage (TAM). <it>Interleukin-10(IL-10) </it>is a potent immunosuppressive cytokine, which can be secreted from both primary tumor and stromal cells. However, there are controversies regarding its role in the progression of cancer. So it is important to isolate TAM from tumor cells to study the role of <it>IL-10 </it>in the progress of cancer. The aim of our study was to determine whether <it>IL-10 </it>expressed by TAM correlated with clinicopathological factors in NSCLC.</p> <p>Methods</p> <p>TAM in NSCLC was isolated by short-term culture in serum free medium with the modification to literature reports. The mRNA expression levels of <it>IL-10</it>, <it>cathepsin B</it>, <it>cathepsin S</it>, which were closely related with TAM according to the literatures, were evaluated by Quantitative real-time RT-PCR in 63 NSCLC. The relationships between their expression levels and clinicopathological features were investigated.</p> <p>Results</p> <p>We successfully achieved up to 95% purity of TAM, derived from 63 primary lung cancer tissues. TAM expressed high levels of <it>IL-10</it>, <it>cathepsin B </it>in NSCLC. High levels of <it>IL-10 </it>in TAM significantly correlated with stage, tumor size, lymph node metastasis, lymphovascular invasion or histologic poor differentiation.</p> <p>Conclusions</p> <p>Our results revealed that TAM with high levels of <it>IL-10 </it>expression may play an important role in the progression of non-small cell lung cancer. The data also suggested that TAMs may involve in tumor immunosuppression through overexpressed <it>IL-10</it>. Additionally, the phenotype of isolated TAM can be potentially used to predict clinicopathological features as well.</p

    Experimental demonstration of picometer level signal extraction with time-delay interferometry technique

    In this work, we have built an experimental setup to simulate the clock noise transmission with two spacecrafts and two optical links, and further demonstrated the extraction of picometer level signal drowned by the large laser frequency noise and clock noise with the data post-processing method. Laser frequency noise is almost eliminated by using the idea of time-delay interferometry (TDI) to construct an equal arm interferometer. Clock asynchronism and clock jitter noise are significantly suppressed by laser sideband transmitting the clock noise using an electro-optic modulator (EOM). Experimental results show a reduction in laser frequency noise by approximately 10^5 and clock noise by 10^2, recovering a weak displacement signal with an average amplitude about 60 picometer and period 1 second. This work has achieved the principle verification of the noise reduction function of TDI technique to some extent, serving the data processing research of space-borne gravitational wave detection

    Fermi Surface and Band Renormalization in (Sr,K)Fe2_2As2_2 Superconductor from Angle-Resolved Photoemission Spectroscopy

    High resolution angle-resolved photoemission measurements have been carried out on (Sr,K)Fe2_2As2_2 superconductor (Tc=21 K). Three hole-like Fermi surface sheets are clearly resolved for the first time around the Gamma point. The overall electronic structure shows significant difference from the band structure calculations. Qualitative agreement between the measured and calculated band structure is realized by assuming a chemical potential shift of -0.2 eV. The obvious band renormalization suggests the importance of electron correlation in understanding the electronic structure of the Fe-based compounds.Comment: 4 pages, 4 figure

    Combination with Methotrexate and Cyclophosphamide Attenuated Maturation of Dendritic Cells: Inducing Treg Skewing and Th17 Suppression In Vivo

    Immune disorder is considered the main pathogenesis of autoimmune diseases, such as rheumatoid arthritis (RA). The balance of the two special subsets of CD4+T cells, T helper cell 17 (Th17), and Regulator T cell (Treg) is the key factor of maintaining a normal immune response. Dendritic cells (DCs), which are the most powerful antigen-presenting cells, play an important role in regulating the balance of Th17 and Treg. The combination of disease modifying antirheumatic drugs (DMARDs) is an important strategy of RA therapy. In this study, we investigated the effect of MTX and CTX on DC maturation in ovalbumin (OVA) immunized mice. Th17 inflammatory response is stronger, while the level of DCs maturity is higher. In contrast, the immunosuppression of Treg is stronger. We found that MTX combined with CTX significantly inhibited the DCs maturity and downregulated the antigen presenting capacity of DCs. As a result, it reestablished a balance of Th17 and Treg. Our study adds a novel mechanism and therapeutic target of MTX combined with CTX for autoimmune disease treatment

    “We are in the forgotten corner!” a qualitative study of experiences and challenges among Chinese older women at the onset of acute myocardial infarction

    BackgroundAcute myocardial infarction (AMI) is a common and serious cardiovascular disease (CVD) that is one of the leading causes of death among women globally and in China. However, there are sex-associated differences and inequalities in the detection and management of AMI, especially in older people. There is little research demonstrating how challenges and barriers affect older women’s help-seeking behavior and health-related procedures in China.PurposeThe objective of this study was to explore the experiences of older women with AMI, focusing on their perception, challenges, and coping strategies at the onset of AMI in Wuhan, China.MethodsThis study utilized a qualitative research design approach and conducted semi-structured, in-depth, and audio-recorded interviews with 18 women aged 65–84 years, purposively selected from two tertiary hospitals in Wuhan City from November 2021 to April 2022.ResultsInterpretative Phenomenological Analysis (IPA) was used in this study to analyze the data on 18 participants and three major themes were generated: disease perception disorder, negative coping strategies, and barriers due to social-environmental contexts.ConclusionTo reduce older women’s delay in seeking help, healthcare professionals should provide public health education that emphasizes sex-related disparities, and age-specific knowledge-attitude aspects to high-risk groups. Policy-based and health administration recommendations, including e-health information support, access to care, and social-environmental factors, should be highlighted to promote women’s health behavior

    Functional Ecological Gene Networks to Reveal the Changes Among Microbial Interactions Under Elevated Carbon Dioxide Conditions

    Biodiversity and its responses to environmental changes is a central issue in ecology, and for society. Almost all microbial biodiversity researches focus on species richness and abundance but ignore the interactions among different microbial species/populations. However, determining the interactions and their relationships to environmental changes in microbial communities is a grand challenge, primarily due to the lack of information on the network structure among different microbial species/populations. Here, a novel random matrix theory (RMT)-based conceptual framework for identifying functional ecological gene networks (fEGNs) is developed with the high throughput functional gene array hybridization data from the grassland microbial communities in a long-term FACE (Free Air CO2 Enrichment) experiment. Both fEGNs under elevated CO2 (eCO2) and ambient CO2 (aCO2) possessed general characteristics of many complex systems such as scale-free, small-world, modular and hierarchical. However, the topological structure of the fEGNs is distinctly different between eCO2 and aCO2, suggesting that eCO2 dramatically altered the interactions among different microbial functional groups/populations. In addition, the changes in network structure were significantly correlated with soil carbon and nitrogen dynamics, and plant productivity, indicating the potential importance of network interactions in ecosystem functioning.     The Influences of Drought and Land-Cover Conversion on Inter-Annual Variation of NPP in the Three-North Shelterbelt Program Zone of China Based on MODIS Data

    Terrestrial ecosystems greatly contribute to carbon (C) emission reduction targets through photosynthetic C uptake.Net primary production (NPP) represents the amount of atmospheric C fixed by plants and accumulated as biomass. The Three-North Shelterbelt Program (TNSP) zone accounts for more than 40% of China’s landmass. This zone has been the scene of several large-scale ecological restoration efforts since the late 1990s, and has witnessed significant changes in climate and human activities.Assessing the relative roles of different causal factors on NPP variability in TNSP zone is very important for establishing reasonable local policies to realize the emission reduction targets for central government. In this study, we examined the relative roles of drought and land cover conversion(LCC) on inter-annual changes of TNSP zone for 2001–2010. We applied integrated correlation and decomposition analyses to a Standardized Evapotranspiration Index (SPEI) and MODIS land cover dataset. Our results show that the 10-year average NPP within this region was about 420 Tg C. We found that about 60% of total annual NPP over the study area was significantly correlated with SPEI (p<0.05). The LCC-NPP relationship, which is especially evident for forests in the south-central area, indicates that ecological programs have a positive impact on C sequestration in the TNSP zone. Decomposition analysis generally indicated that the contributions of LCC, drought, and other Natural or Anthropogenic activities (ONA) to changes in NPP generally had a consistent distribution pattern for consecutive years. Drought and ONA contributed about 74% and 23% to the total changes in NPP, respectively, and the remaining 3% was attributed to LCC. Our results highlight the importance of rainfall supply on NPP variability in the TNSP zone
