    CHARACTERIZATION OF ARSENIC-TOLERANT BACTERIAL CULTURES FROM THE LOWER LAGUNA MADRE OF SOUTH TEXAS

    Two forms of arsenic are found in the environment: As(V) and As(III), the latter being more toxic, water-soluble, and mobile. Microorganisms may increase the mobility of arsenic by reducing As(V) to As(III); however, detoxification and immobilization can occur via the oxidation of As(III) to As(V). The US EPA has set a minimum contaminant level of 10 parts per billion (ppb) for arsenic in drinking water. The research objective was to confirm the presence of arsenic-tolerant bacteria in the Lower Laguna Madre of south Texas. Sediment samples were collected and inoculated into growth media which contained either 2 mM As(III) or 2 mM As(V) to enrich for As(III)-tolerant and As(V)-tolerant bacteria, respectively. Twenty six (26) As (III)-tolerant and 12 As(V)-tolerant cultures were obtained. Most isolates were small white colonies of Gram-positive rods. Biochemical tests using commercially-made test strips showed that As(V)-tolerant isolates displayed greater resource usage compared to As(III)-tolerant isolates but overall, few cultures demonstrated a wide-range of biochemical capabilities. Isolates with distinct morphological and biochemical phenotypes were subjected to Polymerase Chain Reaction (PCR) amplification and sequencing of the 16S rRNA genes to identify the bacteria. Closest sequence matches were to the eubacterial genera Mycoplasma, Salinispora, Frankia, and Pelodictyon. These results suggest that the Lower Laguna Madre is inhabited by a diverse group of microorganisms able to tolerate toxic concentrations of different arsenic species

    Postgraduate Spotlights:Using a Community of Inquiry approach to enhance student engagement in geographical higher education

    While the majority of pedagogical practice has been affected by the COVID-19 pandemic, the teaching of geographical research skills has been especially difficult with the loss of fieldwork and practical applications. Furthermore, the move to online teaching has diminished the learning communities in face-to-face classrooms. In an attempt to counteract these issues, this paper reflects on a learning activity in an undergraduate geographical research methods course, ‘Postgraduate Spotlights’ where two postgraduate researchers presented their specialist research methods followed by an interactive question-and-answer session with the undergraduates. We (as postgraduates, undergraduates and teaching staff) found that the open and critical discussion in the workshop fostered a Community of Inquiry that encouraged engagement from students stimulating their curiosity about geographical research methods. Through our discussion, we demonstrate the importance of having postgraduate researchers involved in teaching, as Graduate Teaching Assistants (GTAs) given their liminal role of researcher-learner. We also emphasise the importance of letting the students lead their own learning, building a Community of Inquiry across academic stages, and creating a constructive dialogue around geographical research methods. While the reproducibility of the workshop face-to-face remains to be seen, this article emphasises the potential for applying such an approach to stimulate free-flowing discussion and ultimately promote a Community of Inquiry

    Subconjunctivally implantable hydrogels with degradable and thermoresponsive properties for sustained release of insulin to the retina.

    The objective of this work is to develop subconjunctivally implantable, biodegradable hydrogels for sustained release of intact insulin to the retina to prevent and treat retinal neurovascular degeneration such as diabetic retinopathy. The hydrogels are synthesized by UV photopolymerization of N-isopropylacrylamide (NIPAAm) monomer and a dextran macromer containing multiple hydrolytically degradable oligolactate-(2-hydroxyetheyl methacrylate) units (Dex-lactateHEMA) in 25:75 (v:v) ethanol:water mixture solvent. Insulin is loaded into the hydrogels during the synthesis process with loading efficiency up to 98%. The hydrogels can release biologically active insulin in vitro for at least one week and the release kinetics can be modulated by varying the ratio between NIPAAm and Dex-lactateHEMA and altering the physical size of the hydrogels. The hydrogels are not toxic to R28 retinal neuron cells in culture medium with 100% cell viability. The hydrogels can be implanted under the conjunctiva without causing adverse effects to the retina based on hematoxylin and eosin stain, immunostaining for microglial cell activation, and electroretinography. These subconjunctivally implantable hydrogels have potential for long-term periocular delivery of insulin or other drugs to treat diabetic retinopathy and other retinal diseases
