Histopathological characteristics of metastasizing squamous cell carcinoma of the skin and lips

Clinical and histopathological criteria have been proposed to identify tumours that may have an increased risk of metastasis. The aim of this study was to define such high-risk tumours, especially since the incidence of SCC of the skin is increasing. METHODS AND RESULTS: Histopathological features of metastasized skin and lip tumours and a matched group of non-metastasizing tumours were reassessed. Characteristics studied were: tumour width, excision margins, histological subtype, Clark level, Breslow depth, tumour differentiation, inflammation, perineural and angio-invasive growth, ulceration and desmoplasia. Data were statistically analysed separately for skin and labial lesions. Desmoplasia, Clark level, Breslow depth, maximum diameter, angio-invasion, grading, perineural invasion, plasma cells and eosinophilic inflammatory response proved to be statistically significantly related to metastasis of skin tumours. Breslow depth, plasma cells and grading appeared to be statistically significantly related to metastasis of SCC of the lips. CONCLUSIONS: A typical metastatic SCC showed: a tumour width of at least 15 mm, a vertical tumour thickness (= Breslow) of at least 2 mm, less differentiation, presence of desmoplasia and an inflammatory response with eosinophils and plasma cells

Laser diagnostics of pulverized coal combustion in O<inf>2</inf>/N<inf>2</inf> and O<inf>2</inf>/CO<inf>2</inf> conditions: velocity and scalar field measurements
Optical diagnostic techniques are applied to a 21 kW laboratory-scale pulverized coal–methane burner to map the reaction zone during combustion, in mixtures with varying fractions of O_2, N_2 and CO_2. Simultaneous Mie scatter and OH planar laser-induced fluorescence (PLIF) measurements have been carried out to study the effect of the oxidizer/diluent concentrations as well as the coal-loading rate. The spatial distribution of soot is captured using laser-induced incandescence (LII). Additionally, velocity profiles at selected axial locations are measured using the pairwise two-dimensional laser Doppler velocimetry technique. The OH PLIF images capture the reaction zones of pilot methane–air flames and the variation of the coal flame structure under various O_2/CO_2 compositions. Coal particles devolatilize immediately upon crossing the flame interface, so that the Mie scatter signal almost vanishes. Increasing coal-loading rates leads to higher reaction rates and shorter flames. LII measurements show that soot is formed primarily in the wake of remaining coal particles in the product regions. Finally, differences in the mean and RMS velocity field are explained by the combined action of thermal expansion and the changes in particle diameter between reacting and non-reacting flows.This work is supported by EPSRC research Grant (#EP/G062153/1). Financial toxicity: a potential side effect of prostate cancer treatment among Australian men.
The purpose of this study was to understand the extent, nature and variability of the current economic burden of prostate cancer among Australian men. An online cross-sectional survey was developed that combined pre-existing economic measures and new questions. With few exceptions, the online survey was viable and acceptable to participants. The main outcomes were self-reported out-of-pocket costs of prostate cancer diagnosis and treatment, changes in employment status and household finances. Men were recruited from prostate cancer support groups throughout Australia. Descriptive statistical analyses were undertaken. A total of 289 men responded to the survey during April and June 2013. Our study found that men recently diagnosed (within 16 months of the survey) (n = 65) reported spending a median AU14 000) for their cancer treatment while 75% of men spent up to AU250 or less in out-of-pocket expenses and some men facing very high costs. On average, respondents in paid employment at diagnosis stated that they had retired 4-5 years earlier than planned
High Frequency Measurement of Temperature and Composition Spots with LITGS
Temperature and composition spots in a turbulent flow are detected and time-resolved using laser-induced thermal grating spectroscopy (LITGS). A 355 nm wavelength particle image velocimetry laser is operated at 0.5–1 kHz to generate the thermal grating using biacetyl as an absorber in trace amounts. In an open laminar jet, a feasibility study shows that small (≃ 3%) fluctuations in the mean flow properties are well captured with LITGS. However, corrections of the mean flow properties by the presence of the trace biacetyl are necessary to properly capture the fluctuations. The actual density and temperature variation in the flow are determined using a calibration procedure validated using a laminar jet flow. Finally, traveling entropy and composition spots are directly measured at different locations along a quartz tube, obtaining good agreement with expected values. This study demonstrates that LITGS can be used as a technique to obtain instantaneous, unsteady temperature and density variations in a combustion chamber, requiring only limited optical access.EPSRC, Qualcom
Modularity of gene-regulatory networks revealed in sea-star development
Evidence that conserved developmental gene-regulatory networks can change as a unit during deutersostome evolution emerges from a study published in BMC Biology. This shows that genes consistently expressed in anterior brain patterning in hemichordates and chordates are expressed in a similar spatial pattern in another deuterostome, an asteroid echinoderm (sea star), but in a completely different developmental context (the animal-vegetal axis). This observation has implications for hypotheses on the type of development present in the deuterostome common ancestor
Zinc intake, status and indices of cognitive function in adults and children: a systematic review and meta-analysis
In developing countries, deficiencies of micronutrients are thought to have a major impact on child development; however, a consensus on the specific relationship between dietary zinc intake and cognitive function remains elusive. The aim of this systematic review was to examine the relationship between zinc intake, status and indices of cognitive function in children and adults. A systematic literature search was conducted using EMBASE, MEDLINE and Cochrane Library databases from inception to March 2014. Included studies were those that supplied zinc as supplements or measured dietary zinc intake. A meta-analysis of the extracted data was performed where sufficient data were available. Of all of the potentially relevant papers, 18 studies met the inclusion criteria, 12 of which were randomised controlled trials (RCTs; 11 in children and 1 in adults) and 6 were observational studies (2 in children and 4 in adults). Nine of the 18 studies reported a positive association between zinc intake or status with one or more measure of cognitive function. Meta-analysis of data from the adult’s studies was not possible because of limited number of studies. A meta-analysis of data from the six RCTs conducted in children revealed that there was no significant overall effect of zinc intake on any indices of cognitive function: intelligence, standard mean difference of <0.001 (95% confidence interval (CI) –0.12, 0.13) P=0.95; executive function, standard mean difference of 0.08 (95% CI, –0.06, 022) P=0.26; and motor skills standard mean difference of 0.11 (95% CI –0.17, 0.39) P=0.43. Heterogeneity in the study designs was a major limitation, hence only a small number (n=6) of studies could be included in the meta-analyses. Meta-analysis failed to show a significant effect of zinc supplementation on cognitive functioning in children though, taken as a whole, there were some small indicators of improvement on aspects of executive function and motor development following supplementation but high-quality RCTs are necessary to investigate this further
A G-quadruplex-binding compound showing anti-tumour activity in an in vivo model for pancreatic cancer
We report here that a tetra-substituted naphthalene-diimide derivative (MM41) has significant in vivo anti-tumour activity against the MIA PaCa-2 pancreatic cancer xenograft model. IV administration with a twice-weekly 15 mg/kg dose produces ca 80% tumour growth decrease in a group of tumour-bearing animals. Two animals survived tumour-free after 279 days. High levels of MM41 are rapidly transported into cell nuclei and were found to accumulate in the tumour. MM41 is a quadruplex-interactive compound which binds strongly to the quadruplexes encoded in the promoter sequences of the BCL-2 and k-RAS genes, both of which are dis-regulated in many human pancreatic cancers. Levels of BCL-2 were reduced by ca 40% in tumours from MM41-treated animals relative to controls, consistent with BCL-2 being a target for MM41. Molecular modelling suggests that MM41 binds to a BCL-2 quadruplex in a manner resembling that previously observed in co-crystal structures with human telomeric quadruplexes. This supports the concept that MM41 (and by implication other quadruplex-targeting small molecules) can bind to quadruplex-forming promoter regions in a number of genes and down-regulate their transcription. We suggest that quadruplexes within those master genes that are up-regulated drivers for particular cancers, may be selective targets for compounds such as MM41
Constraining noncommutative field theories with holography
An important window to quantum gravity phenomena in low energy noncommutative
(NC) quantum field theories (QFTs) gets represented by a specific form of UV/IR
mixing. Yet another important window to quantum gravity, a holography,
manifests itself in effective QFTs as a distinct UV/IR connection. In matching
these two principles, a useful relationship connecting the UV cutoff
, the IR cutoff and the scale of
noncommutativity , can be obtained. We show that an effective
QFT endowed with both principles may not be capable to fit disparate
experimental bounds simultaneously, like the muon and the masslessness of
the photon. Also, the constraints from the muon preclude any possibility
to observe the birefringence of the vacuum coming from objects at cosmological
distances. On the other hand, in NC theories without the UV completion, where
the perturbative aspect of the theory (obtained by truncating a power series in
) becomes important, a heuristic estimate of the region
where the perturbative expansion is well-defined , gets affected when holography is applied by providing the energy of the
system a -dependent lower limit. This may affect models
which try to infer the scale by using data from low-energy

Clinical trials update of the European Organization for Research and Treatment of Cancer Breast Cancer Group
The present clinical trial update consists of a review of two of eight current studies (the 10981-22023 AMAROS trial and the 10994 p53 trial) of the European Organization for Research and Treatment of Cancer Breast Cancer Group, as well as a preview of the MIND-ACT trial. The AMAROS trial is designed to prove equivalent local/regional control for patients with proven axillary lymph node metastasis by sentinel node biopsy if treated with axillary radiotherapy instead of axillary lymph node dissection, with reduced morbidity. The p53 trial started to assess the potential predictive value of p53 using a functional assay in yeast in patients with locally advanced/inflammatory or large operable breast cancer prospectively randomised to a taxane regimen versus a nontaxane regimen
