On the Concept of Radicalism in Surgery for Cervical Cancer
The idea of a radical operation for carcinoma of the cervix originated with Ries and Wertheim. The purpose of this operation, which was applied by Wertheim in 1901, is to remove not only the affected organ, but also the parametrial tissues. However, the limits of the dissection of the parametrium have never been defined with anatomical precision. As a result the extent of such a dissection is left entirely to the choice of the individual surgeon. The results of such procedures, therefore, become difficult to compare statistically. Estudo da biodegradação anaeróbia de efluentes industriais
Mestrado em Engenharia do AmbienteNeste trabalho fez-se o estudo do tratamento anaeróbio de diferentes efluentes industriais em reatores UASB à escala laboratorial.
Numa primeira fase, fez-se o estudo do tratamento anaeróbio de efluentes de uma indústria de mobiliário de madeira (efluente de uma cabine de pintura e envernizamento). Fizeram-se ensaios para testar a influência da carga orgânica e da velocidade ascensional da alimentação. Neste estudo os reatores UASB foram operados de forma descontínua com recirculação total do efluente tratado. Foram testadas cargas de 2,300; 1,150 e 0,671 g CQO.L-1 e velocidades ascensionais de 0,036; 0,072; 0,108 e 0,179 m.h-1. As condições operacionais que favoreceram o tratamento deste efluente foram a carga 0,671 g CQO.L-1 e a velocidade ascensional 0,036 m.h-1, condições para as quais se obteve uma eficiência de remoção de CQO de 86,9%, uma eficiência de metanização da CQO removida de 72,1% e uma biodegradabilidade de 62,6%. Verificou-se que a carga e a velocidade ascensional mais baixas favoreceram a eficiência de metanização e a biodegradabilidade. Os resultados obtidos ao longo dos vários ensaios realizados sugerem a adaptação da biomassa ao substrato aplicado. Conclui-se que o tratamento anaeróbio é uma opção viável para a gestão dos efluentes de cabine de pintura e envernizamento de indústrias de mobiliário de madeira.
Numa segunda fase, foram estudadas diferentes estratégias de operação para o arranque de reatores UASB, inoculados com biomassa floculenta não adaptada ao substrato, utilizados no tratamento de efluentes de indústrias de lacticínios (efluente complexo contendo gorduras). As estratégias testadas foram: operação contínua, operação intermitente e operação intermitente com choques de gorduras. Foram realizados dois ensaios, o primeiro com uma carga orgânica de 5,0 g CQO.L-1. d-1 e o segundo ensaio com 8,5 g CQO.L-1.d-1. Em termos de remoção de CQO, as três estratégias de operação resultaram em valores semelhantes (de 51,3 a 60,4%). Nos dois ensaios verificou-se que a operação intermitente com choques de gorduras resultou nos valores mais elevados de metanização da CQO removida, com valores médios de 56,0% e 69,3% para o primeiro e segundo ensaios, respetivamente. Em comparação, para a operação intermitente, obtiveram-se eficiências de metanização médias de 53,4% e 60,7%, para o primeiro e segundo ensaios, respetivamente. Na operação contínua, as eficiências médias de metanização da CQO removida foram de 45,9% para o primeiro ensaio e 43,9% para o segundo ensaio. Verificou-se também que a operação intermitente com choque de gorduras teve um efeito benéfico em termos de sedimentabilidade da biomassa desenvolvida no reator, tendo-se observado a ausência de flutuação no topo do reator, ao contrário do que foi observado com a operação intermitente e com a operação contínua. Por outro lado, o modo de operação intermitente com choques de gordura apresentou a maior percentagem do grupo microbiano Syntrophomonadaceae, atingindo aproximadamente 20% da contagem total de microrganismos. Dos resultados obtidos conclui-se que a estratégia mais adequada para a adaptação da biomassa floculenta ao efluente de indústria de lacticínios é a operação intermitente com choque de gorduras.This work concerns the study of anaerobic treatment of different industrial wastewaters in laboratory-scale UASB reactors.
The first phase of the work foccused on the anaerobic treatment of a wastewater from a wood furniture industry (wastewater from a dying hood). Tests were made to assess the influence of the organic load and the upflow velocity upon the performance of the process. In this research the UASB reactors were operated in a batch mode with total recirculation of the treated effluent. Loads of 2.300; 1.150 and 0.671 g COD.L-1 and upflow velocities of 0.036, 0.072, 0.108 and 0.179 m.h-1 were tested. The operating conditions that favoured the anaerobic treatment of this wastewater were a load of 0.671 g COD.L-1 and an upflow velocity of 0,036 m.h-1, conditions that lead to a removal efficiency of 86.9%, a methanization efficiency of the removed COD of 72.1% and a biodegradability of 62.6%. It was also established that the lower loads and the lower uplflow velocities favoured the efficiency of methanization of the removed COD and the biodegradability of the wastewater. Finally the results obtained throughout the series of tests performed suggest that the biomass adapted to the applied substrate. It was concluded that the anaerobic process is a viable option for treating the wastewaters from the dying hoods of furniture industries.
In a second phase the work foccused on different startup strategies for UASB reactors, inocculated with flocculent biomass not adapted to the substrate, used for the treatment of dairy wastewaters (complex fat containing wastewaters). The tested strategies were: continuous operation, intermittent operation and intermittent operation with fat shocks. Two tests were performed: the first with an organic loading rate of 5.0 g COD.L-1.d-1 and the second with an organic loading rate of 8.5 g COD.L-1.d-1. In terms of COD removal the three tested strategies resulted in similar values (averages for all reactors from 51.3 to 60.4%). In both the tests it was verified that the intermittent operation with fat shocks resulted in the higher values of methanization of the removed COD with averages values of 56.0% and 69.3% for the first and second tests, respectively. In comparison for the intermittent operation the average methanization efficiencies of the removed COD were 53,4% and 60.7% for the first and the second tests, respectively. With the continuous operation the average methanization efficiencies of the removed COD were 45.9% for the first test and 43.9% for the second test. It was also verified that the intermittent operation with fat shocks had a beneficial effect in terms of biomass sedimentability. It was observed that no floating sludge layer was present in the top of the intermittent reactor with fat shocks in contrast with what was observed with the continuous and the intermittent reactors. On the other hand, the intermittent system with fat shock revealed that a high percentage of microbial group Syntrophomonadaceae, near to 20% of the total microbial count. From the results of this work it may be concluded that the most adequate strategy for the adaptation of flocculent biomass to dairy wastewater is the intermittent operation with fat shocks
Extraction of biomolecules with aqueous two phases systems
Mestrado em Materiais Derivados de Recursos RenováveisOs líquidos iónicos (LIs) representam uma nova classe de solventes iónicos
com pressões de vapor desprezáveis o que os torna potenciais substitutos
para compostos orgânicos voláteis que são actualmente utilizados. Para além
disso, a combinação selectiva do catião e do anião permite afinar as suas
propriedades termofísicas e a sua capacidade de solvatação/extracção.
Os sistemas aquosos bifásicos (SAB) são considerados uma alternativa
rentável e eficaz para a extracção, recuperação e purificação de diversas
biomoléculas.
Neste trabalho, traçaram-se os diagramas de fases de sistemas compostos por
líquido iónico + sal inorgânico + água e líquido iónico + hidratos de carbono +
água, à temperatura de 298 K e pressão atmosférica.
Para além do interesse em SAB baseados em LI e sais inorgânicos, a
aplicação de hidratos de carbono para substituir os sais inorgânicos em
sistemas de elevada carga iónica aparenta ser uma abordagem mais benéfica.
Os dois tipos de SAB estudados foram avaliados no que respeita à sua
capacidade para extracção de biomoléculas. As biomoléculas seleccionadas
comportam aminoácidos, corante alimentares e alcalóides. Os SAB compostos
por LIs demonstraram ser uma potencial abordagem para a extracção de
biomoléculas em processos biotecnológicos.
ABSTRACT: Ionic liquids (ILs) represent a new class of ionic solvents that exhibit negligible
vapour pressures and that further makes them potential substitutes for the
volatile organic compounds currently employed. Moreover, the selection of both
the cation and anion and, therefore, the possibility of fine-tunning their
thermophysical properties and solvation/extraction performance, is an
extremely important feature.
Aqueous biphasic systems (ATPS) are considered attractive alternatives for the
extraction, recovery and purification of several biomolecules.
In this work, phase diagrams composed by ionic liquid + inorganic salt + water,
at 298 K and atmospheric pressure, were determined.
Besides the interest on IL-based ATPS with inorganic salts, the application of
carbohydrates to substitute those highly charged systems seems to be a more
benign approach.
Both type of ATPS were evaluated trough their extraction capability for
biomolecules. The selected biomolecules include amino acids, food colourings
and alkaloids. ILs-based ATPS have shown to be a prospective extraction
media for biomolecules in biotechnological processes
The Role of Transmembrane AMPA Receptor Regulatory Proteins (TARPs) in Synapse Remodeling and Homeostatic Plasticity
Tese de doutoramento em Biologia, na especialidade de Biologia Molecular apresentada à Faculdade de Ciências e Tecnologia da
Universidade de Coimbra.Durante o desenvolvimento do sistema nervoso central o refinamento dos
circuitos neuronais é primariamente controlado por mecanismos de plasticidade de
Hebb. No entanto, esta plasticidade tende a destabilizar os circuitos neuronais,
levando a um descontrolo da excitabilidade neuronal. Existem evidências
convincentes na literatura do controlo homeostático da excitabilidade neuronal em
diversos sistemas. Conhecem-se algumas das vias de sinalização que controlam a
plasticidade homeostática, mas os mecanismos moleculares que regulam este
processo permanecem uma área de investigação activa.
Neste trabalho testámos o papel da stargazina, uma proteína da família das
proteínas transmembranares que interagem com os receptores do glutamato do tipo
AMPA (TARPs), na plasticidade sináptica homeostática. Para isso, investigámos de
que modo a perda da função da stargazina afecta o scaling sináptico induzido por
tetrodotoxina (TTX) em culturas de neurónios corticais. Descobrimos que o bloqueio
crónico da actividade com TTX aumenta os níveis de stargazina bem como a sua
fosforilação, causando acumulação sináptica da stargazina que coincide com
aumento da expressão superficial de receptores do glutamato do tipo AMPA
(AMPAR), uma característica do mecanismo de plasticidade homeostática em
resposta a inibição crónica da actividade neuronal. A fosforilação da stargazina está
aumentada após bloqueio da actividade e regula o scaling sináptico já que a
expressão de mutantes para a fosforilação da stargazina, S9A e S9D, impede a
acumulação sináptica da subunidade GluA1 dos receptores AMPA. Demonstrámos
também o envolvimento das proteínas cinase PKC e CaMKIIβ no scaling sináptico
sendo a stargazina um substrato destas cinases.
O estudo do papel da stargazina na plasticidade dependente de actividade
da sinapse retinogenicular envolveu a caracterização do desenvolvimento desta

sinapse nos ratinhos stargazer. Descobrimos que a stargazina é essencial para a
manutenção sináptica e plasticidade dependente de actividade, já que os ratinhos
stargazer apresentam um defeito na maturação da sinapse retinogenicular no
período sensível à visão. Analisámos ainda os níveis de TARPs e das subunidades
dos AMPAR no núcleo geniculado lateral (dLGN) após diferentes manipulações
visuais. É importante ressaltar que os níveis de stargazina são regulados pela
visão, e estão aumentados no tálamo visual quando os ratinhos são privados de
visão entre p20-27 (LDR-late dark rearing), sem se registarem alterações noutro
membro das TARPs, a TARPγ4. Finalmente, descobrimos que, de acordo com o
que foi descrito anteriormente (Gainey et al. 2009), o bloqueio crónico da actividade
de neurónios corticais aumenta a expressão da subunidade GluA2. Esta
subunidade está também aumentada no dLGN após LDR, demostrando que haverá
semelhantes efectores moleculares da plasticidade homeostático in vitro e in vivo.
De acordo com o nosso conhecimento, esta é a primeira evidência de
indução de plasticidade homeostática em regiões sub-corticais em resposta à
privação de visão binocular. Com base nos nossos resultados propomos um modelo
em que a fosforilação da stargazina é o interruptor principal para a acumulação
sináptica de AMPAR durante a plasticidade homeostática, uma forma de
plasticidade essencial para o desenvolvimento de circuitos e adaptação ao
ambiente. O nosso trabalho mostra a stargazina como um novo regulador da
homeostase dos circuitos.Activity-dependent synaptic refinement during development occurs primarily
through Hebbian mechanisms of plasticity. However, this plasticity tends to
destabilize the neuronal circuits leading to runway excitation or inhibition. There is
now compelling evidence from a number of systems for homeostatic control of firing
rates in central neurons. Molecular control of homeostatic processes is an active
area of research.
We tested whether stargazin participates in homeostatic synaptic plasticity
by investigating how loss of stargazin function affects TTX-induced synaptic scaling
in cultured cortical neurons. We found that chronic activity blockade with TTX
increased stargazin levels and stargazin phosphorylation state, leading to increased
synaptic stargazin accumulation coincident with increased surface expression of
AMPA-type glutamate receptors, a hallmark of homeostatic plasticity induced by
chronic inhibition of neuronal activity. Stargazin phosphorylation was increased upon
chronic activity blockade and regulates synaptic scaling as expression of stargazin
phospho-mutants, S9A and S9D, disrupted increased surface expression of GluA1,
an AMPA receptor subunit. We were also able to show the involvement of PKC and
CaMKIIβ in synaptic scaling.
The role of stargazin in experience-dependent plasticity was studied by
characterizing the development of the retinogeniculate synapse of stargazer mice.
We found that stargazin is essential for synaptic maintenance and experiencedependent
plasticity, as stargazer mice showed disrupted retinogeniculate synapse
maturation at the vision sensitive period. Furthermore, we biochemically analyzed
TARPs and AMPA receptor subunits in the dorsal lateral geniculate nucleus (dLGN)
after different visual manipulations. Importantly, stargazin levels are regulated by
vision, and stargazin was up-regulated in the visual thalamus when experiencexvi
dependent plasticity was induced by late-dark rearing, without significant changes in
the thalamus-expressed family member TARPγ4. Finally, we found that, similarly to
what was previously described (Gainey et al. 2009) and confirmed in our in vitro
system, GluA2 subunit is increased in dLGN after late dark-rearing mice, showing
similar molecular effectors of homeostatic plasticity in vitro and in vivo.
To our knowledge this is the first evidence for homeostatic plasticity
mechanisms being induced in sub-cortical regions in response to binocular visual
deprivation. Herein we propose a model for stargazin phosphorylation as the main
switch controlling AMPAR synaptic accumulation and trapping during homeostatic
plasticity. This form of plasticity is essential for circuit development and adaptation to
the ever-changing environment, making stargazin a new regulator of circuit homeostasis
Produção de extratos bioativos a partir de resíduos da CMC-Biomassa S.A. por extração supercrítica com solventes verdes
Mestrado em Engenharia QuímicaEsta dissertação teve como objetivo estudar a extração e caraterização de compostos bioativos com valor acrescentado, nomeadamente os ácidos triterpénicos (TTAs) (ácidos betulínico, betulónico, oleanólico, ursólico e derivados acetilados dos dois últimos) da folhagem de Eucalyptus globulus. Efectuaram-se extracções com metanol, etanol e diclorometano, pelo método Soxhlet, e com dióxido de carbono na extração supercrítica (SFE), modificado ou não com etanol (cossolvente). Analisaram-se os extractos por FTIR-ATR, GC-MS e SEM, em conjunto com métodos hierárquicos de análise de agrupamentos (clusters) com vista à comparação dos diferentes extractos entre si, com a biomassa original e com os TTAs puros.
Verificou-se que as folhas de eucalipto contêm uma quantidade elevada de extractáveis, com rendimentos totais de 30.34 % (m/m) para extrações Soxhlet com metanol, e 7.32 % com diclorometano. Por SFE os rendimentos totais variaram entre 1.52 e 3.16 % para extracções realizadas com CO2 a 200 bar com 0 e 5.0 % (m/m) de etanol, respectivamente. Após optimização alcançaram-se rendimentos totais de 3.95 % e de TTAs de 0.67 % (m/m), para extracções a 250 bar com CO2 contendo 5.0 wt.% etanol, a 40 ᵒC e caudal de 12 gCO2 min-1.
A remoção específica de ceras das folhas foi estudada com êxito obtendo-se posteriormente extratos particularmente ricos em TTAs. Realizou-se um estudo cinético para as condições ótimas de SFE, usando biomassa tal e qual (estilhas sem pré-tratamento) ou pré-tratada para redução de granulometria (moagem) ou remoção de ceras. Destes resultados destaca-se o elevado rendimento total obtido com a amostra moída (5.90 %, ao fim de 6 h) e o bom desempenho da biomassa sem tratamento em termos de extração de TTAs (rendimento 0.64 % e concentração de 16.5 %). A biomassa com pré-remoção das ceras gerou extratos com concentrações de TTAs similares aos da biomassa não tratada.
No geral, o presente trabalho fornece informação útil para o estudo da extração de compostos bioativos da folhagem de E. globulus, assim como detalhes de composição e rendimentos que podem constituir o ponto de partida para estudos ainda mais detalhados a realizar futuramente.This work focuses the extraction and characterization of high added value bioactive compounds obtained from Eucalyptus globulus leaves, namely triterpenic acids (TTAs) such as betulonic, betulinic, oleanolic and ursolic acids, and the acetylated derivates of the last two. The extractions were done using methanol, ethanol, and dichloromethane for the Soxhlet method, and carbon dioxide for the supercritical fluid extraction (SFE), modified or not with ethanol (cosolvent). The extracts were characterized by FTIR-ATR, GC-MS and SEM, together with the hierarchical cluster analysis method for comparison of the different extracts between themselves and with the original biomass and with the pure TTAs.
The results revealed Eucalyptus leaves have high content of extractives, namely 30.34 wt.% for Soxhlet extraction with methanol and 7.32 wt.% with dichloromethane. With SFE the total extraction yield varied between 1.52 and 3.16 wt.%, for extractions at 200 bar with CO2 containing 0 wt.% and 5.0 wt.% of ethanol, respectively. After optimization, total extraction yields of 3.95 wt.% and 0.67 wt.% for TTAs were attained, at 250 bar with CO2 containing 5.0 wt.% ethanol, temperature of 40 ᵒC and flow rate of 12 gCO2 min-1
The removal of waxes from the leaves was successfully studied enabling the production of extracts rich in TTAs. Kinetic studies were performed at the SFE optimized conditions to compare the effect of crushing or dewaxing the leaves in relation to untreated leaves. Highest yields were obtained with the crushed leaves (5.90 wt.%, after 6 h) while in terms of TTAs extraction the best results were obtained with untreated biomass (yield 0.64 wt.%, and concentration 16.5 wt.%). The extracts obtained with untreated and dewaxed biomass samples presented similar concentrations of TTAs.
The present work presents useful information for the extraction of bioactive compounds from E. globulus leaves as well as attainable yields and composition that may guide future comprehensive studies
Excess ribosomal protein production unbalances translation in a model of Fragile X Syndrome
Dysregulated protein synthesis is a core pathogenic mechanism in Fragile X Syndrome (FX). The mGluR Theory of FX predicts that pathological synaptic changes arise from the excessive translation of mRNAs downstream of mGlu1/5 activation. Here, we use a combination of CA1 pyramidal neuron-specific TRAP-seq and proteomics to identify the overtranslating mRNAs supporting exaggerated mGlu1/5 -induced long-term synaptic depression (mGluR-LTD) in the FX mouse model (Fmr1−/y). Our results identify a significant increase in the translation of ribosomal proteins (RPs) upon mGlu1/5 stimulation that coincides with a reduced translation of long mRNAs encoding synaptic proteins. These changes are mimicked and occluded in Fmr1−/y neurons. Inhibiting RP translation significantly impairs mGluR-LTD and prevents the length-dependent shift in the translating population. Together, these results suggest that pathological changes in FX result from a length-dependent alteration in the translating population that is supported by excessive RP translation
Nanotoxicology and Nanosafety: Safety-by-Design and Testing at a Glance
This review offers a systematic discussion about nanotoxicology and nanosafety associated with nanomaterials during manufacture and further biomedical applications. A detailed introduction on nanomaterials and their most frequently uses, followed by the critical risk aspects related to regulatory uses and commercialization, is provided. Moreover, the impact of nanotoxicology in research over the last decades is discussed, together with the currently available toxicological methods in cell cultures (in vitro) and in living organisms (in vivo). A special focus is given to inorganic nanoparticles such as titanium dioxide nanoparticles (TiO2NPs) and silver nanoparticles (AgNPs). In vitro and in vivo case studies for the selected nanoparticles are discussed. The final part of this work describes the significance of nano-security for both risk assessment and environmental nanosafety. “Safety-by-Design” is defined as a starting point consisting on the implementation of the principles of drug discovery and development. The concept “Safety-by-Design” appears to be a way to “ensure safety”, but the superficiality and the lack of articulation with which it is treated still raises many doubts. Although the approach of “Safety-by-Design” to the principles of drug development has helped in the assessment of the toxicity of nanomaterials, a combination of scientific efforts is constantly urgent to ensure the consistency of methods and processes. This will ensure that the quality of nanomaterials is controlled and their safe development is promoted.
