Effect of endogenous purines on electrically evoked ACh release at the mouse neuromuscular junction
At the mouse neuromuscular junction, adenosine triphosphate (ATP), which is co-released with the neurotransmitter acetylcholine (ACh), and its metabolite adenosine, modulate neurotransmitter release by activating presynaptic inhibitory P2Y13 receptors (a subtype of ATP/adenosine diphosphate [ADP] receptor), inhibitory A1 and A3 adenosine receptors, and excitatory A2A adenosine receptors. To study the effect of endogenous purines, when phrenic-diaphragm preparations are depolarized by different nerve stimulation patterns, we analyzed the effect of the antagonists for P2Y13, A1, A3, and A2A receptors (AR-C69931MX, 8-cyclopentyl-1,3-dipropylxanthine, MRS-1191, and SCH-58261, respectively) on the amplitude of the end-plate potentials of the trains, and contrasted these results with those obtained with the selective agonists of these receptors (2-methylthioadenosine 5âČ-diphosphate trisodium salt hydrate, 2-chloro-N6-cyclopentyl-adenosine, inosine, and PSB-0777, respectively). During continuous 0.5-Hz stimulation, the amount of endogenous purines was not enough to activate purinergic receptors, while at continuous 5-Hz stimulation, an incipient action of endogenous purines on P2Y13, A1 and A3 receptors might be evident just at the end of the trains. During continuous 50-Hz stimulation, the concentration of endogenous ATP/ADP and adenosine exerted an inhibitory action on ACh release after of the initial phase of the train, but when the nerve was stimulated at intermittent 50 Hz (5 bursts), this behavior was not observed. Excitatory A2A receptors were only activated when continuous 100-Hz stimulation was applied. In conclusion, when motor nerve terminals are depolarized by repetitive stimulation of the phrenic nerve, endogenous ATP/ADP and adenosine are able to fine-tune neurosecretion depending on the frequency and pattern of stimulation.Fil: GonzĂĄlez Sanabria, Javier A. Effects of mixed of a ketogenic diet in overweight and obese women with polycystic ovary syndrome
Polycystic ovary syndrome (PCOS) is a commonly occurring endocrine disorder characterized by hirsutism, anovulation, and polycystic ovaries. Often comorbid with insulin resistance, dyslipidemia, and obesity, it also carries significant risk for the development of cardiovascular and metabolic sequelae, including diabetes and metabolic syndrome. The relationship between central obesity and the development of insulin resistance is widely verified. Adipose tissue excess and the coexistent dysregulation of adipocyte functions directly contribute to the pathogenesis of the metabolic complications observed in women with PCOS. In the light of these evidence, the most therapeutic option prescribed to obese women with PCOS, regardless of the phenotype e from the severity of clinical expression, is lifestyle correction by diet and physical activity. The aim of this study is to evaluate the beneficial effects of ketogenic diet in 17 obese women with PCOS. Our results showed that the ketogenic diet inducing therapeutic ketosis, improves the anthropometric and many biochemical parameters such as LH, FSH, SHBG, insulin sensitivity and HOMA index. In addition, it induces a reduction in androgenic production, whereas the contextual reduction of fat mass reduced the acyclic production of estrogens deriving from the aromatization in the adipose tissue of the androgenic excess, with an improvement of the LH/FSH ratio. This is the first study on the effects of the ketogenic diet on PCOS, however, further studies are needed to elucidate the mechanism underlying ketogenic diet effects
Lessons Learned from the September 11th Disaster: A State Health Agency Perspective
The New York State Department of Health has conducted a number of studies over the past 10 years investigating health impacts related to the September 11, 2001 (9/11) disaster among New York City residents and New York State World Trade Center (WTC) responders. Efforts to evaluate the health effects of WTC exposures in these cohorts presented numerous challenges, including study design and associated concerns about bias, identifying the affected populations, gaining community support and participation, and determining the most appropriate clinical testing and follow-up approaches. The unique position of a state public health agency provided multiple points of support for these efforts. An overview of what was found and the lessons learned during the response to the 9/11 disaster is presented, from the viewpoint of a state public health agency
Produzione, qualitĂ e analisi della filiera produttiva del lino da fibra in Italia
Fibre flax (Linum usitatissimum L.) is disappeared from the ltalian crop systems, while the textile industry has reached a leading position. The reintroduction of the crop is than potentially promising but the crop techniques and the production processes have to be set up.
In 12 environments: crop development and growth, cultivar response, straw, fibre and seed yields, and fibre quality were analysed in the 1992-98 period.
The data pointed out crop cycles of 176 and 110 d for the autumn and the spring sowing time respectively, corresponding to 1125 and 990°C d cumulated growing degree. Straw yield at pulling was on average of 6.3 t ha-1, with relevant differences among years and environrnents. Early maturity cultivar yielded more in the less favourable sites. N fertilization was occasionally relevant, and a generally distributions of 60 kg N ha-1 was the more effective. In the Mediterranean environments, with the autumn, sowing flax was more productive and the yield steady. The long fibre ratio was often low (10-13%) and the quality uneven because of the insufficient cleanliness due to retting difficulties. Occasionally, the value of the fibre was affected by the insufficient stem length. In the less dense crop, the seed yield was on average 0.9 t ha-1. Such productions were similar to those assessed for the more dense crop.
In conclusion: because of the complexity of the interactions among the crop growth, the yield, the retting process and the fibre quality flax reintroduction is doubtful. In the production processes retting seems to be the crucial phase.
Il lino da fibra (Linum usitatissimum L.) Ăš da tempo assente negli ordinamenti colturali italiani, mentre Ăš assai importante l'industria di filatura. La reintroduzione della coltura appare quindi potenzialmente promettente, ma richiede la messa a punto dell'agrotecnica e della filiera produttiva.
Nel periodo 1992-1998 in 12 ambienti sono stati esaminati: lo sviluppo e la crescita della coltura, l'adattamento di cultivar, la produzione di paglia, fibra e seme, la resa alla stigliatura, la qualitĂ della fibra.
I dati raccolti evidenziano cicli colturali in media di 176 e 110 d rispettivamente per le semine autunnali (ambienti del centro-sud) e vemino-primaverili; corrispondenti a somme termiche di 1125 e 990°C d. La produzione di paglia alla estirpatura Ú stata in media di 6.3 t ha-1 con valori assai variabili tra annate e ambienti. Le varietà a ciclo corto sono apparse migliori nelle condizioni difficili. L'effetto della concimazione azotata Ú apparsa talvolta rilevante e comunque con dosi ottimali prossime a 60 kg ha-1 di N. Negli ambienti del centro-sud le semine autunnali si sono
dimostrate spesso le piĂč interessanti e sicure. La resa in fibra lunga Ăš stata bassa (10-13%) e la qualitĂ variabile per la scarsa pulizia dovuta alle difficoltĂ di macerazione. La modesta lunghezza tecnica dello stelo sovente riduce il valore del prodotto. In coltura rada, la produzione di seme Ăš variata da 0.5 a 1.5 t ha-1; tali valori non si sono discostati in modo apprezzabile da quelli ottenuti in coltura fitta.
In conclusione, la diffusione del lino appare problematica per le complesse interazioni tra la produzione, il processo di macerazione e la qualitĂ della fibra. Soprattutto la macerazione appare il passaggio chiave
Microservice Transition and its Granularity Problem: A Systematic Mapping Study
Microservices have gained wide recognition and acceptance in software
industries as an emerging architectural style for autonomic, scalable, and more
reliable computing. The transition to microservices has been highly motivated
by the need for better alignment of technical design decisions with improving
value potentials of architectures. Despite microservices' popularity, research
still lacks disciplined understanding of transition and consensus on the
principles and activities underlying "micro-ing" architectures. In this paper,
we report on a systematic mapping study that consolidates various views,
approaches and activities that commonly assist in the transition to
microservices. The study aims to provide a better understanding of the
transition; it also contributes a working definition of the transition and
technical activities underlying it. We term the transition and technical
activities leading to microservice architectures as microservitization. We then
shed light on a fundamental problem of microservitization: microservice
granularity and reasoning about its adaptation as first-class entities. This
study reviews state-of-the-art and -practice related to reasoning about
microservice granularity; it reviews modelling approaches, aspects considered,
guidelines and processes used to reason about microservice granularity. This
study identifies opportunities for future research and development related to

Precise Spatiotemporal Control of Optogenetic Activation Using an Acousto-Optic Device
Light activation and inactivation of neurons by optogenetic techniques has emerged as an important tool for studying neural circuit function. To achieve a high resolution, new methods are being developed to selectively manipulate the activity of individual neurons. Here, we report that the combination of an acousto-optic device (AOD) and single-photon laser was used to achieve rapid and precise spatiotemporal control of light stimulation at multiple points in a neural circuit with millisecond time resolution. The performance of this system in activating ChIEF expressed on HEK 293 cells as well as cultured neurons was first evaluated, and the laser stimulation patterns were optimized. Next, the spatiotemporally selective manipulation of multiple neurons was achieved in a precise manner. Finally, we demonstrated the versatility of this high-resolution method in dissecting neural circuits both in the mouse cortical slice and the Drosophila brain in vivo. Taken together, our results show that the combination of AOD-assisted laser stimulation and optogenetic tools provides a flexible solution for manipulating neuronal activity at high efficiency and with high temporal precision
Ubiquitous Networks, Ubiquitous Sensors: Issues Of Security, Reliability And Privacy In The Internet Of Things
The growth of ubiquitous networks moves ever more personal data into collectible and computational opportunities. This changes assumptions based on past latency and limits on data analysis and brings challenges to the reliability of data acquisition principles and practices and their proper use in societies. System engineers must consider outcomes and related regulation in the design and use of these systems. It also cautions us as to overreaching. We examine the general legal sphere within which ubiquitous networks and associated data exist, mapping some technological outcomes to legal consequences. We consider the impact and legal decisions regarding ubiquitous networks in the United States as an indicator of future direction and legal entanglements of these technological systems. In particular, we consider security and privacy principles that may regulate computational use through ubiquitous networks, and the competing interests/benefits/detriments in their use. This informs as to possible future regulation that may be needed or required and offer guidance with the growing data sphere and ubiquitous networking, and advises that the engineering of such systems should be flexible enough to accommodate new regulatory regimes as to deployment, access and use
