The Buenos Aires water concession
The signing of a concession contract for the Buenos Aires water and sanitation system in December 1992, attracted worldwide attention, and caused considerable controversy in Argentina. It was one of the world's largest concessions, but the case was also interesting for other reasons. The concession was implemented rapidly, in contrast with slow implementation of privatization in Santiago, for example. And reform generated major improvements in the sector, including wider coverage, better service, more efficient company operations, and reduced waste. Moreover, the winning bid brought an immediate 26.9 percent reduction in water system tariffs. Consumers benefited from the system's expansion and from the immediate drop in real prices, which was only partly reversed by subsequent changes in tariffs, and access charges. And these improvements would probably not have occurred under public administration of the system. Still, the authors show information asymmetries, perverse incentives, and weak regulatory institutions could threaten the concession's sustainability. Opportunities for the company to act opportunistically - and the regulator, arbitrarily - exist, because of politicized regulation, a poor information base, serious flaws in the concession contract, a lumpy and ad hoc tariff system, and a general lack of transparency in the regulatory process. Because of these circumstances, public confidence in the process has eroded. A Tuned and Scalable Fast Multipole Method as a Preeminent Algorithm for Exascale Systems
Among the algorithms that are likely to play a major role in future exascale
computing, the fast multipole method (FMM) appears as a rising star. Our
previous recent work showed scaling of an FMM on GPU clusters, with problem
sizes in the order of billions of unknowns. That work led to an extremely
parallel FMM, scaling to thousands of GPUs or tens of thousands of CPUs. This
paper reports on a a campaign of performance tuning and scalability studies
using multi-core CPUs, on the Kraken supercomputer. All kernels in the FMM were
parallelized using OpenMP, and a test using 10^7 particles randomly distributed
in a cube showed 78% efficiency on 8 threads. Tuning of the
particle-to-particle kernel using SIMD instructions resulted in 4x speed-up of
the overall algorithm on single-core tests with 10^3 - 10^7 particles. Parallel
scalability was studied in both strong and weak scaling. The strong scaling
test used 10^8 particles and resulted in 93% parallel efficiency on 2048
processes for the non-SIMD code and 54% for the SIMD-optimized code (which was
still 2x faster). The weak scaling test used 10^6 particles per process, and
resulted in 72% efficiency on 32,768 processes, with the largest calculation
taking about 40 seconds to evaluate more than 32 billion unknowns. This work
builds up evidence for our view that FMM is poised to play a leading role in
exascale computing, and we end the paper with a discussion of the features that
make it a particularly favorable algorithm for the emerging heterogeneous and
massively parallel architectural landscape

Modest Declines in Proteome Quality Impair Hematopoietic Stem Cell Self-Renewal.
Low protein synthesis is a feature of somatic stem cells that promotes regeneration in multiple tissues. Modest increases in protein synthesis impair stem cell function, but the mechanisms by which this occurs are largely unknown. We determine that low protein synthesis within hematopoietic stem cells (HSCs) is associated with elevated proteome quality in vivo. HSCs contain less misfolded and unfolded proteins than myeloid progenitors. Increases in protein synthesis cause HSCs to accumulate misfolded and unfolded proteins. To test how proteome quality affects HSCs, we examine Aarssti/sti mice that harbor a tRNA editing defect that increases amino acid misincorporation. Aarssti/sti mice exhibit reduced HSC numbers, increased proliferation, and diminished serial reconstituting activity. Misfolded proteins overwhelm the proteasome within Aarssti/sti HSCs, which is associated with increased c-Myc abundance. Deletion of one Myc allele partially rescues serial reconstitution defects in Aarssti/sti HSCs. Thus, HSCs are dependent on low protein synthesis to maintain proteostasis, which promotes their self-renewal
Purification and characterization of a polygalacturonase produced by Wickerhamomyces anomalus
The aim of this work was to study the purification and physicochemical properties of an endo-polygalacturonase (PG) produced by Wickerhamomyces anomalus isolated from the citrus fruit peels. The enzyme was purified to homogeneity from the culture filtrate of W. anomalus grown on the yeast nitrogen base medium with glucose as carbon and energy source and citrus pectin as inductor. After anion-exchange chromatography and gel filtration chromatography, PG activity was eluted as a single peak, yielding 21% of the original activity. After dialysis and cation-exchange chromatography, only one fraction with PG activity was obtained, recovering 56% of initial enzyme activity and 1.3-fold increase in specific activity. The molecular weight of the enzyme was estimated as 43 kDa by the SDS-PAGE. The enzyme exhibited maximal activity at pH 4.2 and was stable over a pH range from 3.5 to 6.0 and up to 49°C for 10 h. The Vmax and Km values with polygalacturonic acid as substrate were 0.26 mmol/L. min and 0.173 mg/mL, respectively. Cations such as Cu+2, Fe+3, Mg+2, Mn+2 and Zn+2 did not show any significant effect on PG activity but K+ and Ca+2 reduced it. A conceptual map of invasion biology: Integrating hypotheses into a consensus network
Background and aims
Since its emergence in the mid‐20th century, invasion biology has matured into a productive research field addressing questions of fundamental and applied importance. Not only has the number of empirical studies increased through time, but also has the number of competing, overlapping and, in some cases, contradictory hypotheses about biological invasions. To make these contradictions and redundancies explicit, and to gain insight into the field’s current theoretical structure, we developed and applied a Delphi approach to create a consensus network of 39 existing invasion hypotheses.
Results
The resulting network was analysed with a link‐clustering algorithm that revealed five concept clusters (resource availability, biotic interaction, propagule, trait and Darwin’s clusters) representing complementary areas in the theory of invasion biology. The network also displays hypotheses that link two or more clusters, called connecting hypotheses, which are important in determining network structure. The network indicates hypotheses that are logically linked either positively (77 connections of support) or negatively (that is, they contradict each other; 6 connections).
Significance
The network visually synthesizes how invasion biology’s predominant hypotheses are conceptually related to each other, and thus, reveals an emergent structure – a conceptual map – that can serve as a navigation tool for scholars, practitioners and students, both inside and outside of the field of invasion biology, and guide the development of a more coherent foundation of theory. Additionally, the outlined approach can be more widely applied to create a conceptual map for the larger fields of ecology and biogeography
