    Functional diversification of the nematode mbd2/3 gene between Pristionchus pacificus and Caenorhabditis elegans

    Abstract Background Several members of the Methyl-Binding Domain protein family link DNA methylation with chromatin remodeling complexes in vertebrates. Amongst the four classes of MBD proteins, MBD2/3 is the most highly conserved and widespread in metazoans. We have previously reported that an mbd2/3 like gene (mbd-2) is encoded in the genomes of the nematodes Pristionchus pacificus, Caenorhabditis elegans and Caenorhabditis briggsae. RNAi knock-down of mbd-2 in the two Caenorhabditis species results in varying percentages of lethality. Results Here, we report that a general feature of nematode MBD2/3 proteins seems to be the lack of a bona fide methyl-binding domain. We isolated a null allele of mbd-2 in P. pacificus and show that Ppa-mbd-2 mutants are viable, fertile and display a fully penetrant egg laying defect. This egg laying defect is partially rescued by treatment with acetylcholine or nicotine suggesting a specific function of this protein in vulval neurons. Using Yeast-two-hybrid screens, Ppa-MBD-2 was found to associate with microtubule interacting and vesicle transfer proteins. Conclusion These results imply that MBD2/3 proteins in nematodes are more variable than their relatives in insects and vertebrates both in structure and function. Moreover, nematode MBD2/3 proteins assume functions independent of DNA methylation ranging from the indispensable to the non-essential.</p

    Sensory Perception of Food and Insulin-Like Signals Influence Seizure Susceptibility

    Food deprivation is known to affect physiology and behavior. Changes that occur could be the result of the organism's monitoring of internal and external nutrient availability. In C. elegans, male mating is dependent on food availability; food-deprived males mate with lower efficiency compared to their well-fed counterparts, suggesting that the mating circuit is repressed in low-food environments. This behavioral response could be mediated by sensory neurons exposed to the environment or by internal metabolic cues. We demonstrated that food-deprivation negatively regulates sex-muscle excitability through the activity of chemosensory neurons and insulin-like signaling. Specifically, we found that the repressive effects of food deprivation on the mating circuit can be partially blocked by placing males on inedible food, E. coli that can be sensed but not eaten. We determined that the olfactory AWC neurons actively suppress sex-muscle excitability in response to food deprivation. In addition, we demonstrated that loss of insulin-like receptor (DAF-2) signaling in the sex muscles blocks the ability of food deprivation to suppress the mating circuit. During low-food conditions, we propose that increased activity by specific olfactory neurons (AWCs) leads to the release of neuroendocrine signals, including insulin-like ligands. Insulin-like receptor signaling in the sex muscles then reduces cell excitability via activation of downstream molecules, including PLC-γ and CaMKII

    Learning to Think Iconically in the Human and Social Sciences: Iconic Standards of Understanding as a Pivotal Challenge for Method Development

    Theoretically as well as alongside an empirical research idea, this paper outlines conditions for the development of social scientific empirical methods able to further exploit the iconic potential of the image. Reconstructing the role of formal pictorial elements for the standards of understanding within the medium “image” is considered pivotal in this endeavor. Within the context of language, standards of communication have already been extensively researched. The linguistic format of the narrative, for instance, is well studied. Up to now, though, comparable formal vehicles of iconic semantics have only been examined in aesthetics and art history. Nevertheless, standards of iconic understanding are part of our implicit knowledge, are incessantly in use in everyday practice and, thus, the basis of everyday identity formation. With the help of empirical methods based on an iconic logos we can deepen our understanding of orientations, longings, and anxieties of our time that are often silently conveyed by images.     Snowmass2021 Cosmic Frontier Dark Matter Direct Detection to the Neutrino Fog

    We present a summary of future prospects for direct detection of dark matter within the GeV/c2 to TeV/c2 mass range. This is paired with a new definition of the neutrino fog in order to better quantify the rate of diminishing returns on sensitivity due to irreducible neutrino backgrounds. A survey of dark matter candidates predicted to fall within this mass range demonstrates that fully testing multiple well-motivated theo-ries will require expanding the currently-funded generation of experiments down to and past the neutrino fog. We end with the status and plans for next-generation exper-iments and novel R&amp;D concepts which will get us there
