Substrate specificity provides insights into the sugar donor recognition mechanism of O-GlcNAc transferase (OGT).
O-Linked β-N-acetylglucosaminyl transferase (OGT) plays an important role in the glycosylation of proteins, which is involved in various cellular events. In human, three isoforms of OGT (short OGT [sOGT]; mitochondrial OGT [mOGT]; and nucleocytoplasmic OGT [ncOGT]) share the same catalytic domain, implying that they might adopt a similar catalytic mechanism, including sugar donor recognition. In this work, the sugar-nucleotide tolerance of sOGT was investigated. Among a series of uridine 5'-diphosphate-N-acetylglucosamine (UDP-GlcNAc) analogs tested using the casein kinase II (CKII) peptide as the sugar acceptor, four compounds could be used by sOGT, including UDP-6-deoxy-GlcNAc, UDP-GlcNPr, UDP-6-deoxy-GalNAc and UDP-4-deoxy-GlcNAc. Determined values of Km showed that the substitution of the N-acyl group, deoxy modification of C6/C4-OH or epimerization of C4-OH of the GlcNAc in UDP-GlcNAc decreased its affinity to sOGT. A molecular docking study combined with site-directed mutagenesis indicated that the backbone carbonyl oxygen of Leu653 and the hydroxyl group of Thr560 in sOGT contributed to the recognition of the sugar moiety via hydrogen bonds. The close vicinity between Met501 and the N-acyl group of GlcNPr, as well as the hydrophobic environment near Met501, were responsible for the selective binding of UDP-GlcNPr. These findings illustrate the interaction of OGT and sugar nucleotide donor, providing insights into the OGT catalytic mechanism
Superconducting properties of novel BiSe-based layered LaOFBiSe single crystals
F-doped LaOBiSe superconducting single crystals with typical size of
240.2 mm are successfully grown by flux method and the
superconducting properties are studied. Both the superconducting transition
temperature and the shielding volume fraction are effectively improved with
fluorine doping. The LaOFBiSe sample exhibits
zero-resistivity at 3.7 K, which is higher than that of the
LaOFBiSe polycrystalline sample (2.4K). Bulk
superconductivity is confirmed by a clear specific-heat jump at the associated
temperature. The samples exhibit strong anisotropy and the anisotropy parameter

Superconductivity and Charge Density Wave in ZrTeSe
Charge density wave (CDW), the periodic modulation of the electronic charge
density, will open a gap on the Fermi surface that commonly leads to decreased
or vanishing conductivity. On the other hand superconductivity, a commonly
believed competing order, features a Fermi surface gap that results in infinite
conductivity. Here we report that superconductivity emerges upon Se doping in
CDW conductor ZrTe when the long range CDW order is gradually suppressed.
Superconducting critical temperature in ZrTeSe
() increases up to 4 K plateau for
. Further increase in Se content results in
diminishing and filametary superconductivity. The CDW modes from Raman
spectra are observed in = 0.04 and 0.1 crystals, where signature of
ZrTe CDW order in resistivity vanishes. The electronic-scattering for
high crystals is dominated by local CDW fluctuations at high
temperures, the resistivity is linear up to highest measured and
contributes to substantial in-plane anisotropy.Comment: 15 pages, 4 figure
One Fish, Two Fish, Red Fish, Blue Fish: Effects of Price Frames, Brand Names, and Choice Set Size in Medicare Part D Insurance Plan Decisions
Because many seniors choose Medicare Part D plans offering poorer coverage at greater cost, the authors examined the effect of price frames, brand names, and choice set size on participants\u27 ability to choose the lowest cost plan. A 2×2×2 within-subjects design was used with 126 participants aged 18 to 91 years old. Mouselab, a web-based program, allowed participants to choose drug plans across eight trials that varied using numeric or symbolic prices, real or fictitious drug plan names, and three or nine drug plan options. Results from the multilevel models suggest numeric versus symbolic prices decreased the likelihood of choosing the lowest cost plan (-8.0 percentage points, 95% confidence interval=-14.7 to -0.9). The likelihood of choosing the lowest cost plan decreased as the amount of information increased suggesting that decision cues operated independently and collectively when selecting a drug plan. Redesigning the current Medicare Part D plan decision environment could improve seniors\u27 drug plan choices
Electron-hole bilayer quantum dots: Phase diagram and exciton localization
We studied a vertical ``quantum dot molecule'', where one of the dots is
occupied with electrons and the other with holes. We find that different phases
occur in the ground state, depending on the carrier density and the interdot
distance. When the system is dominated by shell structure, orbital degeneracies
can be removed either by Hund's rule, or by Jahn-Teller deformation. Both
mechanisms can lead to a maximum of the addition energy at mid-shell. At low
densities and large interdot distances, bound electron-hole pairs are formed.Comment: 10 pages, 3 figure
