    GALEX Observations of CS and OH Emission in Comet 9P/Tempel 1 During Deep Impact

    GALEX observations of comet 9P/Tempel 1 using the near ultraviolet (NUV) objective grism were made before, during and after the Deep Impact event that occurred on 2005 July 4 at 05:52:03 UT when a 370 kg NASA spacecraft was maneuvered into the path of the comet. The NUV channel provides usable spectral information in a bandpass covering 2000 - 3400 A with a point source spectral resolving power of approximately 100. The primary spectral features in this range include solar continuum scattered from cometary dust and emissions from OH and CS molecular bands centered near 3085 and 2575 A, respectively. In particular, we report the only cometary CS emission detected during this event. The observations allow the evolution of these spectral features to be tracked over the period of the encounter. In general, the NUV emissions observed from Tempel 1 are much fainter than those that have been observed by GALEX from other comets. However, it is possible to derive production rates for the parent molecules of the species detected by GALEX in Tempel 1 and to determine the number of these molecules liberated by the impact. The derived quiescent production rates are Q(H2O) = 6.4e27 molecules/s and Q(CS2) = 6.7e24 molecules/s, while the impact produced an additional 1.6e32 H2O molecules and 1.3e29 CS2 molecules, a similar ratio as in quiescent outgassing.Comment: 15 pages, 4 figures, accepted for publication in the Astrophysical Journa

    Understanding the association between skin involvement and joint activity in patients with psoriatic arthritis: experience from the Corrona Registry.

    Objective: To compare the characteristics of patients with psoriatic arthritis among patient groups stratified by degree of skin and joint involvement, and to evaluate the relationship between skin severity and joint activity. Methods: Body surface area (BSA) and Clinical Disease Activity Index (CDAI) at enrolment were analysed. Patient characteristics were stratified by skin severity and joint activity. Baseline patient characteristics, clinical and disease characteristics and patient-reported outcomes were compared. The strength of the relationship of skin severity and joint activity was evaluated using methods for categorical variables (χ Results: 1542 adult patients in the Corrona Psoriatic Arthritis/Spondyloarthritis Registry enrolled between 21 May 2013 and 20 September 2016 were analysed.     Synergies between interstellar dust and heliospheric science with an Interstellar Probe

    We discuss the synergies between heliospheric and dust science, the open science questions, the technological endeavors and programmatic aspects that are important to maintain or develop in the decade to come. In particular, we illustrate how we can use interstellar dust in the solar system as a tracer for the (dynamic) heliosphere properties, and emphasize the fairly unexplored, but potentially important science question of the role of cosmic dust in heliospheric and astrospheric physics. We show that an Interstellar Probe mission with a dedicated dust suite would bring unprecedented advances to interstellar dust research, and can also contribute-through measuring dust - to heliospheric science. This can, in particular, be done well if we work in synergy with other missions inside the solar system, thereby using multiple vantage points in space to measure the dust as it `rolls' into the heliosphere. Such synergies between missions inside the solar system and far out are crucial for disentangling the spatially and temporally varying dust flow. Finally, we highlight the relevant instrumentation and its suitability for contributing to finding answers to the research questions.Comment: 18 pages, 7 Figures, 5 Tables.     Molecular preservation by extraction and fixation, mPREF: a method for small molecule biomarker analysis and histology on exactly the same tissue

    <p>Abstract</p> <p>Background</p> <p>Histopathology is the standard method for cancer diagnosis and grading to assess aggressiveness in clinical biopsies. Molecular biomarkers have also been described that are associated with cancer aggressiveness, however, the portion of tissue analyzed is often processed in a manner that is destructive to the tissue. We present here a new method for performing analysis of small molecule biomarkers and histology in exactly the same biopsy tissue.</p> <p>Methods</p> <p>Prostate needle biopsies were taken from surgical prostatectomy specimens and first fixed, each in a separate vial, in 2.5 ml of 80% methanol:water. The biopsies were fixed for 24 hrs at room temperature and then removed and post-processed using a non-formalin-based fixative (UMFIX), embedded, and analyzed by hematoxylin and eosin (H&E) and by immunohistochemical (IHC) staining. The retained alcohol pre-fixative was analyzed for small molecule biomarkers by mass spectrometry.</p> <p>Results</p> <p>H&E analysis was successful following the pre-fixation in 80% methanol. The presence or absence of tumor could be readily determined for all 96 biopsies analyzed. A subset of biopsy sections was analyzed by IHC, and cancerous and non-cancerous regions could be readily visualized by PIN4 staining. To demonstrate the suitability for analysis of small molecule biomarkers, 28 of the alcohol extracts were analyzed using a mass spectrometry-based metabolomics platform. All extracts tested yielded successful metabolite profiles. 260 named biochemical compounds were detected in the alcohol extracts. A comparison of the relative levels of compounds in cancer containing <it>vs</it>. non-cancer containing biopsies showed differences for 83 of the compounds. A comparison of the results with prior published reports showed good agreement between the current method and prior reported biomarker discovery methods that involve tissue destructive methods.</p> <p>Conclusions</p> <p>The Molecular Preservation by Extraction and Fixation (mPREF) method allows for the analysis of small molecule biomarkers from exactly the same tissue that is processed for histopathology.</p

    Arthritis Increases the Risk for Fractures: Results from the Women's Health Initiative

    OBJECTIVE: The relationship between arthritis and fracture was examined in the Women’s Health Initiative (WHI). METHODS: Women were classified into three self-reported groups at baseline: no arthritis (n=83,295), osteoarthritis (OA) (n=63,402), and rheumatoid arthritis (RA) (n=960). Incident fractures were self-reported throughout follow-up. Age-adjusted fracture rates by arthritis category were generated, and Cox-proportional hazards model was used to test the association between arthritis and fracture. RESULTS: After an average of 7.80 years, 24,137 total fractures were reported including 2,559 self-reported spinal fractures and 1,698 adjudicated hip fractures. For each fracture type, age-adjusted fracture rates were highest in the RA group and lowest in the non-arthritic group. After adjustment for several covariates, report of arthritis was associated with increased risk for spine, hip, and any clinical fractures. Compared to the non-arthritis group, the risk [HR (95% CI)] of sustaining any clinical fracture in the OA group was 1.09 (1.05, 1.13) (p<0.001) and 1.49 (1.26, 1.75) (p<0.001) in the RA group. The risk of sustaining a hip fracture was not statistically increased in the OA group [1.11 (0.98, 1.25)] (p=0.122) compared to the non-arthritis group; however the risk of hip fracture significantly increased [3.03 (2.03, 4.51)] (p<0.001) in the RA group compared to the non-arthritis group. CONCLUSION: The increase in fracture risk found in this study confirms the importance of fracture prevention in patients with both RA and OA

    Recapture and retreatment rates with ixekizumab after withdrawal of therapy in patients with axial spondyloarthritis: Results at week 104 from a randomised placebo-controlled withdrawal study

    Objectives: To evaluate the recapture of response with open-label (OL) ixekizumab (IXE) retreatment at week 104 in patients with axial spondyloarthritis who flared after withdrawal of IXE therapy. Methods: COAST-Y (NCT03129100) is a phase III extension study that included a double-blind, placebo-controlled, randomised withdrawal-retreatment period (RWRP). Patients who achieved remission (Ankylosing Spondylitis Disease Activity Score (ASDAS) <1.3 (inactive disease, ID) at least once at week 16 or 20 and <2.1 (low disease activity, LDA) at both visits) were randomised 2:1 at week 24 to continue IXE or withdraw to placebo. Patients who subsequently flared were switched to OL IXE every 2 or 4 weeks (Q2W or Q4W) at the next visit. The proportions of patients who recaptured ASDAS LDA and ID were summarised for those who experienced flare. Results: Of the 155 patients who entered the RWRP (placebo, n=53; IXE Q4W, n=48; IXE Q2W, n=54), 138 (89%) completed week 104. Of the placebo-Treated patients (n=53), 28 (53%) experienced a flare during weeks 24-104; of these, 4 (14%) recaptured ASDAS LDA before retreatment with OL IXE, and 23 (82%) recaptured ASDAS LDA and 19 (68%) met ASDAS ID after retreatment. Of the continuously treated IXE patients (n=102), 13 experienced flare; 7 of 13 (54%) recaptured ASDAS LDA before switching to OL IXE retreatment, while 5 of 13 (38%) recaptured ASDAS LDA and 4 of 13 (31%) met ID after switching. Conclusions: Ninety-six per cent of patients withdrawn to placebo recaptured at least ASDAS LDA and 71% recaptured ASDAS ID with IXE retreatment at week 104. This may provide support to patients who may require a brief interruption in therapy

    Rotationally Resolved 8-35 Micron Spitzer Space Telescope Observations of the Nucleus of Comet 9P/Tempel 1

    International audienceWe have utilized the Spitzer Space Telescope (SST) Infrared Spectrograph (IRS) to directly observe thermal emission from the nucleus of comet 9P/Tempel 1 on UT 2004 March 25-27. We obtained 8-35 mum low-resolution (R~100) spectra and contemporaneous 16 and 22 mum photometric imaging over a 39 hr period. The comet was 3.7 AU from the Sun at the time, approximately 464 days before perihelion on 2005 July 5, and showed no evidence of extended emission beyond a point source. Visual inspection of the absolute photometry implies a rotation period of 40 +/- 2 hr, consistent with earlier results. Snapshot photometry by Spitzer at 8 and 24 mum, taken on UT 2004 March 10 and 15, respectively, are consistent with this light-curve phasing and with the IRS-measured flux. The spectra agree well with the predictions of the standard thermal model for a slowly rotating body with thermal inertia between 0 and 50 J K-1 m-2 s-1/2, and are inconsistent with any rapid rotator model. The mean effective radius at the middle of the light curve is 3.3 +/- 0.2 km. The maximum-to-minimum flux ratio of 1.8 in the light curve is consistent with an axial ratio a/b of 3.2 +/- 0.4, implying a=7.2+/-0.9 km and b=2.3+/-0.3 km. Combining our SST infrared light curve with visible observations of the nucleus, we obtain a visible geometric albedo of 0.04 +/- 0.01. With this sized nucleus and the published water production rates, we estimate that 9% +/- 2% of the surface area is actively emitting volatile material at perihelion
