    Drivers of consumers’ intention to adopt sustainable healthy dietary patterns: evidence from China

    IntroductionIn line with the shift towards sustainable consumption, sustainable healthy dietary patterns (SHDP) have received considerable attention, but no study has examined Chinese consumers’ intention to adopt SHDP.MethodsBased on the theory of planned behaviour (TPB), this study integrated health consciousness, environmental concerns, and past eating behaviour to construct an expanded TPB framework for analyzing the factors influencing Chinese consumers’ intention to adopt SHDP. The mediating role of attitude between perceived value and consumers’ intention to adopt SHDP was also analyzed. The study empirically tested the research model using structural equation modelling estimation, based on the data collected from 402 local consumers in Wuxi, China.Results and discussionThe results showed that attitude, perceived behavioural control, health consciousness, and past eating behaviuor positively and significantly influenced consumers’ intention to adopt SHDP, whereas perceived value indirectly influenced adoption intention through attitude. Subjective norms and environmental concerns had no significant influence on adoption intention. Based on these findings, interventions through dietary education and information campaigns are recommended to enhance consumers’ value awareness and attitudes towards SHDP. Interventions, such as nudging, should be designed to enhance consumers’ perceived behavioural control and dietary practises. The findings of this study provide important insights for the development of dietary change intervention strategies

    Consumers' decoy effect when purchasing pork with traceability technologies

    Despite government investment, policy guidance, and publicity, it has been difficult to establish a traceable food market in China over the past 2 decades. Once a food safety problem occurs, it is difficult to implement effective traceability, recall, and accountability along the food supply chain. How to use the decoy effect to promote the development of China traceable food market? As bounded rationality, a decoy effect exists when adding an alternative to a choice set increases the chance an existing alternative to be chosen. However, few studies have examined the decoy effect in food purchases. Based on consumers in Wuxi, Jiangsu Province, China, we show the decoy effect in traceable pork hindquarter purchases and that the effects differ across product quality and price attributes. The effects are heterogeneous across consumers and are less likely to occur among those who had a personal annual income of more than 50,000 yuan (USD $7,000), were married, and had minor children in the family. These findings have implications on leveraging the influence of the decoy effect on consumer behavior and facilitating the construction of food traceability systems

    Impact of online public opinion regarding the Japanese nuclear wastewater incident on stock market based on the SOR model

    The exposure of the Japanese nuclear wastewater incident has shaped online public opinion and has also caused a certain impact on stocks in aquaculture and feed industries. In order to explore the impact of network public opinion caused by public emergencies on relevant stocks, this paper uses the stimulus organism response(SOR) model to construct a framework model of the impact path of network public opinion on the financial stock market, and it uses emotional analysis, LDA and grounded theory methods to conduct empirical analysis. The study draws a new conclusion about the impact of online public opinion on the performance of relevant stocks in the context of the nuclear waste water incident in Japan. The positive change of media sentiment will lead to the decline of stock returns and the increase of volatility. The positive change of public sentiment will lead to the decline of stock returns in the current period and the increase of stock returns in the lag period. At the same time, we have proved that media attention, public opinion theme and prospect theory value have certain influences on stock performance in the context of the Japanese nuclear wastewater incident. The conclusion shows that after the public emergency, the government and investors need to pay attention to the changes of network public opinion caused by the event, so as to avoid the possible stock market risks

    Cytological DNA methylation for cervical cancer screening: a validation set

    BackgroundIn a previous training set with a case-controlled design, cutoff values for host EPB41L3 and JAM3 gene methylation were obtained for the detection of cervical intraepithelial neoplasia (CIN) 2 or more severe lesions (CIN2+). This validation trial was conducted to evaluate the role of DNA methylation in screening for CIN2+ by cervical cytology among unselected participants.MethodsFrom June 1, 2019, to September 1, 2019, in our study center, we collected liquid-based samples from cervical swabs for methylation assays and hrHPV testing in eligible patients. The primary endpoint was the diagnostic accuracy of DNA methylation and hrHPV genotyping for CIN2+ according to confirmed histology results.ResultsAmong 307 participants, compared with hrHPV testing, the methylation assay for CIN2+ had lower sensitivity (68.7% versus 86.1%, p=0.002) but higher specificity (96.7% versus 0.696, p<0.001). The methylation assay also had favorable sensitivity and specificity in patients with negative hrHPV testing (56.3% and 96.9%) and in patients with cervical adenocarcinoma (73.7% and 92.7%). DNA methylation had higher specificity than the hrHPV assay (100.0% versus 44.4%, p<0.001) for identifying residual CIN2+ in patients without residual lesions. Positive cervical DNA methylation was associated with a diagnostic probability of endometrial carcinoma (odds ratio 15.5 [95% confidence interval 4.1-58.6]) but not of ovarian epithelial carcinoma (1.4 [0.3-6.5]).ConclusionsThe host EPB41L3 and JAM3 gene methylation assay in cervical cytology had favorable diagnostic accuracy for CIN2+ and was highly specific for residual CIN2+ lesions The methylation assay is a promising triage tool in hrHPV+ women, or even an independent tool for cervical cancer screening. The methylation status in cervical cytology could also serve as a prognostic biomarker. Its role in detecting endometrial carcinomas is worthy of further exploration

    Multi-omics analyses of glucose metabolic reprogramming in colorectal cancer

    BackgroundGlucose metabolic reprogramming (GMR) is a cardinal feature of carcinogenesis and metastasis. However, the underlying mechanisms have not been fully elucidated. The aim of this study was to profile the metabolic signature of primary tumor and circulating tumor cells from metastatic colorectal cancer (mCRC) patients using integrated omics analysis.MethodsPET-CT imaging, serum metabolomics, genomics and proteomics data of 325 high 18F-fluorinated deoxyglucose (FDGhigh) mCRC patients were analyzed. The para-tumor, primary tumor and liver metastatic tissues of mCRC patients were used for proteomics analysis.ResultsThe glucose uptake in tumor tissues as per the PET/CT images was correlated to serum levels of glutamic-pyruvic transaminase (ALT), total bilirubin (TBIL), creatinine (CRE). Proteomics analysis indicated that several differentially expressed proteins were enriched in both GMR and epithelial-mesenchymal transition (EMT)-related pathways. Using a tissue-optimized proteomic workflow, we identified novel proteomic markers (e.g. CCND1, EPCAM, RPS6), a novel PCK1-CDK6-INSR protein axis, and a potential role for FOLR (FR) in GMR/EMT of CRC cells. Finally, CEA/blood glucose (CSR) was defined as a new index, which can be used to jointly diagnose liver metastasis of colorectal cancer.ConclusionsGMR in CRC cells is closely associated with the EMT pathway, and this network is a promising source of potential therapeutic targets

    A five-collagen-based risk model in lung adenocarcinoma: prognostic significance and immune landscape

    As part of the tumor microenvironment (TME), collagen plays a significant role in cancer fibrosis formation. However, the collagen family expression profile and clinical features in lung adenocarcinoma (LUAD) are poorly understood. The objective of the present work was to investigate the expression pattern of genes from the collagen family in LUAD and to develop a predictive signature based on collagen family. The Cancer Genome Atlas (TCGA) samples were used as the training set, and five additional cohort samples obtained from the Gene Expression Omnibus (GEO) database were used as the validation set. A predictive model based on five collagen genes, including COL1A1, COL4A3, COL5A1, COL11A1, and COL22A1, was created by analyzing samples from the TCGA cohort using LASSO Cox analysis and univariate/multivariable Cox regression. Using Collagen-Risk scores, LUAD patients were then divided into high- and low-risk groups. KM survival analysis showed that collagen signature presented a robust prognostic power. GO and KEGG analyses confirmed that collagen signature was associated with extracellular matrix organization, ECM-receptor interaction, PI3K-Akts and AGE-RAGE signaling activation. High-risk patients exhibited a considerable activation of the p53 pathway and cell cycle, according to GSEA analysis. The Collage-Risk model showed unique features in immune cell infiltration and tumor-associated macrophage (TAM) polarization of the TME. Additionally, we deeply revealed the association of collagen signature with immune checkpoints (ICPs), tumor mutation burden (TMB), and tumor purity.     Constructional Meaning and Inheritance in Mandarin Chinese LVCs: The Case of the GIVE Group

    This study focuses on aspectual properties of three light verb constructions in Mandarin Chinese involving the verbs jiyu, yuyi and jiayi, all meaning ‘give’. These verbs combine with an action nominal to form a complex predicate, but unlike jiyu and yuyi, the jiayi-construction is incompatible with any aspectual markers, as evidenced by my investigation into a million-word Mandarin corpus. Assuming that light verbs typically follow the grammaticalisation path from independent verbs to grammatical morphemes, I propose that the three GIVE verbs are at different stages of grammaticalisation. The verb jiayi is the most grammaticalised item. However, jiyu and yuyi have not fully grammaticalised as aspectual morphemes, so aspectuality has to be encoded by means of aspectual markers such as the perfective markers -le and -guo. Taking into account their different aspectual properties, this study gives a constructional presentation on the family of GIVE light verb constructions, which demonstrates both conventional meaning common to all GIVE light verb constructions and idiosyncratic features particular to each light verb construction

    ULK1 phosphorylates Exo70 to suppress breast cancer metastasis

    乳腺癌是威胁女性生命健康的“头号杀手”,而远处转移是乳腺癌患者死亡的主要原因。因此,了解乳腺癌如何发动侵袭和转移,对于有效治疗乳腺癌、延长病人生存期具有重要意义。本研究中,该团队发现ULK1通过结合并磷酸化胞泌蛋白复合体关键亚基Exo70来抑制乳腺癌转移。ULK1对Exo70上Ser47,Ser59和Ser89位点的磷酸化,严重地削弱了Exo70的自身寡聚化和与其它胞外分泌复合体亚基的结合,进而减少了细胞运动伪足形成以及基质金属蛋白酶的分泌,从而抑制乳腺癌细胞的迁移和侵袭。该论文首次揭示了胞外分泌复合体重要成员Exo70在乳腺癌中受到ULK1和ERK1/2的双重磷酸化调控,从而使得乳腺癌细胞可以根据外环境来决定潜伏还是发动侵袭转移,为乳腺癌的治疗提供了新的理论基础。 本论文的通讯作者为占艳艳副教授、郭巍教授和胡天惠教授。医学院博士生毛丽媛、占艳艳副教授、吴斌博士和医学院博士生于强为共同第一作者。【Abstract】Increased expression of protein kinase ULK1 was reported to negatively correlate with breast cancer metastasis. Here we report that ULK1 suppresses the migration and invasion of human breast cancer cells. The suppressive effect is mediated through direct phosphorylation of Exo70, a key component of the exocyst complex. ULK1 phosphorylation inhibits Exo70 homo-oligomerization as well as its assembly to the exocyst complex, which are needed for cell protrusion formation and matrix metalloproteinases secretion during cell invasion. Reversely, upon growth factor stimulation, Exo70 is phosphorylated by ERK1/2, which in turn suppresses its phosphorylation by ULK1.     Characterization of the sesame (Sesamum indicum L.) global transcriptome using Illumina paired-end sequencing and development of EST-SSR markers

    <p>Abstract</p> <p>Background</p> <p>Sesame is an important oil crop, but limited transcriptomic and genomic data are currently available. This information is essential to clarify the fatty acid and lignan biosynthesis molecular mechanism. In addition, a shortage of sesame molecular markers limits the efficiency and accuracy of genetic breeding. High-throughput transcriptomic sequencing is essential to generate a large transcriptome sequence dataset for gene discovery and molecular marker development.</p> <p>Results</p> <p>Sesame transcriptomes from five tissues were sequenced using Illumina paired-end sequencing technology. The cleaned raw reads were assembled into a total of 86,222 unigenes with an average length of 629 bp. Of the unigenes, 46,584 (54.03%) had significant similarity with proteins in the NCBI nonredundant protein database and Swiss-Prot database (E-value < 10<sup>-5</sup>). Of these annotated unigenes, 10,805 and 27,588 unigenes were assigned to gene ontology categories and clusters of orthologous groups, respectively. In total, 22,003 (25.52%) unigenes were mapped onto 119 pathways using the Kyoto Encyclopedia of Genes and Genomes Pathway database (KEGG). Furthermore, 44,750 unigenes showed homology to 15,460 <it>Arabidopsis </it>genes based on BLASTx analysis against The Arabidopsis Information Resource (TAIR, Version 10) and revealed relatively high gene coverage. In total, 7,702 unigenes were converted into SSR markers (EST-SSR). Dinucleotide SSRs were the dominant repeat motif (67.07%, 5,166), followed by trinucleotide (24.89%, 1,917), tetranucleotide (4.31%, 332), hexanucleotide (2.62%, 202), and pentanucleotide (1.10%, 85) SSRs. AG/CT (46.29%) was the dominant repeat motif, followed by AC/GT (16.07%), AT/AT (10.53%), AAG/CTT (6.23%), and AGG/CCT (3.39%). Fifty EST-SSRs were randomly selected to validate amplification and to determine the degree of polymorphism in the genomic DNA pools. Forty primer pairs successfully amplified DNA fragments and detected significant amounts of polymorphism among 24 sesame accessions.</p> <p>Conclusions</p> <p>This study demonstrates that Illumina paired-end sequencing is a fast and cost-effective approach to gene discovery and molecular marker development in non-model organisms. Our results provide a comprehensive sequence resource for sesame research.</p
