    Tropical storm-induced turbulent mixing and chlorophyll-a enhancement in the continental shelf southeast of Hainan Island

    AbstractBased on moored observations and remote sensing data in July and August 2005, energy sources for enhancing turbulent mixing and possible mechanisms of phytoplankton bloom in the continental shelf southeast of Hainan Island under the influence of Washi, a fast-moving and weak tropical storm, are analyzed in this paper. Observations show that strong near-inertial internal waves were generated by the rapidly changing wind stress and the near-inertial energy was dissipated quickly across the thermocline. The strong turbulent mixing associated with the near-inertial baroclinic shear instability occurred with maximum eddy diffusivity above 3.2×10−4m2s−1, and the surface chlorophyll-a (Chl-a) concentration after the storm increased by 22.2%. The Chl-a concentration augment was inferred to be an upper ocean biophysical response to the enhanced near-inertial turbulent mixing which could increase the upward nutrient flux into the surface low eutrophic zone during the passage of Washi

    Characterization of vascular endothelial progenitor cells from chicken bone marrow

    BACKGROUND: Endothelial progenitor cells (EPC) are a type of stem cell used in the treatment of atherosclerosis, vascular injury and regeneration. At present, most of the EPCs studied are from human and mouse, whereas the study of poultry-derived EPCs has rarely been reported. In the present study, chicken bone marrow-derived EPCs were isolated and studied at the cellular level using immunofluorescence and RT-PCR. RESULTS: We found that the majority of chicken EPCs were spindle shaped. The growth-curves of chicken EPCs at passages (P) 1, -5 and -9 were typically “S”-shaped. The viability of chicken EPCs, before and after cryopreservation was 92.2% and 81.1%, respectively. Thus, cryopreservation had no obvious effects on the viability of chicken EPCs. Dil-ac-LDL and FITC-UAE-1 uptake assays and immunofluorescent detection of the cell surface markers CD34, CD133, VEGFR-2 confirmed that the cells obtained in vitro were EPCs. Observation of endothelial-specific Weibel-Palade bodies using transmission electron microscopy further confirmed that the cells were of endothelial lineage. In addition, chicken EPCs differentiated into endothelial cells and smooth muscle cells upon induction with VEGF and PDGF-BB, respectively, suggesting that the chicken EPCs retained multipotency in vitro. CONCLUSIONS: These results suggest that chicken EPCs not only have strong self-renewal capacity, but also the potential to differentiate into endothelial and smooth muscle cells. This research provides theoretical basis and experimental evidence for potential therapeutic application of endothelial progenitor cells in the treatment of atherosclerosis, vascular injury and diabetic complications

    The miR-1204 regulates apoptosis in NSCLC cells by targeting DEK

    Introduction. This study endeavors to analyze the effects of miR-1204 on the expression of DEK oncogene in non-small cell lung cancer (NSCLC) cell lines and to study the molecular mechanisms of these effects. Material and methods. The miR-1204 mimics and inhibitors were transfected into the (A549 and SPC) NSCLC cells. Then the mRNA levels, cell viability, apoptosis rate, morphology and caspase activity were determined. The expression of apoptosis-related proteins Bcl-2 and Bax was also analyzed. Results. In NSCLC cell lines (A549 and SPC), DEK mRNA levels were down-regulated in miR-1204 overex­pression group. In miR-1204 inhibition group, the expression of DEK mRNA showed an opposite trend. The overexpression of miR-1204 increases the apoptosis rate in NSCLC cells. The Bcl-2 levels in the miR-1204 over­expression group were decreased, while the Bax level was increased. In the miR-1204 inhibition group, expression of Bcl-2 and Bax showed opposite trends. Cell staining revealed cell’s morphological changes; the apoptosis in the miR-1204 overexpression group revealed significant morphological features, such as brighter nuclei and nu­clear condensation. Results indicated a typical characteristic of apoptosis in the miR-1204 overexpression group. Caspase-9 and Caspase-3 were involved in the apoptosis pathway, which was mediated by miR-1204 and DEK. Conclusions. The miR-1204 induces apoptosis of NSCLC cells by inhibiting the expression of DEK. The mech­anism of apoptosis involves down-regulation of Bcl-2 and up-regulation of Bax expression. Moreover, the apoptosis was mediated by mitochondria-related caspase 9/3 pathway

    O Serviço Social nas Autarquias e a sua Importância para o Desenvolvimento Social Local

    Aprofundar o conhecimento do Serviço Social e reflectir sobre a prática profissional do Assistente Social e a criação de políticas sociais com vista ao desenvolvimento social local, nomeadamente, na Câmara Municipal da Batalha, constituíram o objectivo de estudo. A descentralização do Poder Central para o Poder Local, assente na proximidade ao cidadão, mantém-se em discussão na agenda política e ganha maior relevo na conjuntura actual, com a reforma do Poder Local. Contudo, até ao momento, as transferências no âmbito da Acção Social mantêm-se bastante genéricas e sem regulamentação. Por essa razão, o Poder Local intervém na área social sem que essas competências estejam delineadas pela tutela e muitas vezes sem o devido financiamento, deixando aos Executivos Municipais a decisão sobre a criação de políticas sociais. Neste sentido, com o intuito de assegurarem os interesses das suas populações, as Câmaras Municipais implementam medidas sociais de âmbito local, que se revelam mais ou menos intensas, consoante o importância que lhes é dada por cada Executivo, que define as áreas de intervenção prioritárias e quais os recursos disponíveis para investir no domínio social. O Serviço Social revela ser um importante recurso das autarquias na criação das políticas sociais locais, na medida em que o Assistente Social, ao conhecer o território e intervir mais próximo dos cidadãos, pode propor programas de desenvolvimento local, adequados aos interesses da população. No caso particular da Câmara Municipal da Batalha, reflectiu-se sobre a prática da Assistente Social e evocaram-se as políticas sociais por esta planeadas e desenvolvidas, revelando o seu contributo para o desenvolvimento social do concelho. Atestou-se, em género de conclusão, que, apesar do Assistente Social ter um papel cada vez mais preponderante na execução das políticas de desenvolvimento local, a sua prática profissional tem limitações por não ser capaz, por si só, de resolver problemas sociais de génese estrutural, influenciados pela conjuntura nacional e internacional
