    Investigation of a New Flux-Modulated Permanent Magnet Brushless Motor for EVs

    This paper presents a flux-modulated direct drive (FMDD) motor. The key is to integrate the magnetic gear with the PM motor while removing the gear inner-rotor. Hence, the proposed FMDD motor can achieve the low-speed high-torque output and high-speed compact design requirements as well as high-torque density with a simple structure. The output power equation is analytically derived. By using finite element analysis (FEA), the static characteristics of the proposed motor are obtained. Based on these characteristics, the system mathematical model can be established. Hence, the evaluation of system performances is conducted by computer simulation using the Matlab/Simulink. A prototype is designed and built for experimentation. Experimental results are given to verify the theoretical analysis and simulation

    Effects of biochar and vermicompost on growth and economic benefits of continuous cropping pepper at karst yellow soil region in Southwest China

    Recently, biochar (B) and vermicompost (V) have been widely used as amendments to improve crop productivity and soil quality. However, the ameliorative effects of biochar and vermicompost on the continuous cropping of pepper under open-air conditions, particularly in the karst areas of southwestern China, remain unclear. A field experiment was conducted to study the effects of biochar and vermicompost application, alone or in combination, on the yield, quality, nutrient accumulation, fertilizer utilization, and economic benefits of continuous pepper cropping from 2021 to 2022. The experiment included six treatments: CK (no fertilizer), TF (traditional fertilization of local farmers), TFB (TF combined with biochar of 3000 kg·ha-1), TFV (TF combined with vermicompost of 3000 kg·ha-1), TFBV1 (TF combined with biochar of 1500 kg·ha-1 and vermicompost of 1500 kg·ha-1), and TFBV2 (TF combined with biochar of 3000 kg·ha-1 and vermicompost of 3000 kg·ha-1). Compared with the TF treatment, biochar and vermicompost application alone or in combination increased the yield of fresh pod pepper by 24.38–50.03% and 31.61–88.92% in 2021 and 2022, respectively, whereas the yield of dry pod pepper increased by 14.69–40.63% and 21.44–73.29% in 2021 and 2022, respectively. The application of biochar and vermicompost reduced the nitrate content and increased the vitamin C (VC) and soluble sugar content of the fruits, which is beneficial for improving their quality. Biochar and vermicompost application alone or in combination not only increased nutrient uptake but also significantly improved agronomic efficiency (AE) and recovery efficiency (RE). In addition, although the application of biochar or vermicompost increased production costs, the increase in yield improved net income (ranging from 0.77 to 22.34% in 2021 and 8.82 to 59.96% in 2022), particularly in the TFBV2 treatment. In conclusion, the use of biochar and vermicompost amendments had a positive effect on the productivity and economic benefits of continuous pepper cropping, and the co-application of biochar and vermicompost could be an effective nutrient management strategy for the continuous cropping of pepper in the karst mountain areas of southwest China

    Case report: Effectiveness of sirolimus in a de novo FAS mutation leading to autoimmune lymphoproliferative syndrome-FAS and elevated DNT/Treg ratio

    BackgroundThe autoimmune lymphoproliferative syndrome (ALPS) is a rare disease characterized by defective function of the FAS death receptor, which results in chronic, non-malignant lymphoproliferation and autoimmunity accompanied by elevated numbers of double-negative (DN) T cells (T-cell receptor α/β + CD4–CD8–) and an increased risk of developing malignancies later in life.Case descriptionHere, we report a patient with a de novo FAS mutation with a severe phenotype of ALPS-FAS. The FAS gene identified as a novel spontaneous germline heterozygous missense mutation (c.857G > A, p.G286E) in exon 9, causing an amino acid exchange and difference in hydrogen bond formation. Consequently, the treatment with sirolimus was initiated. Subsequently, the patient’s clinical condition improved rapidly. Moreover, DNT ratio continuously decreased during sirolimus application.ConclusionWe described a novel germline FAS mutation (c.857G > A, p.G286E) associated with a severe clinical phenotype of ALPS-FAS. Sirolimus effectively improved the patient clinical manifestations with obvious reduction of the DNT ratio

    Protein phosphatase 5 and the tumor suppressor p53 down-regulate each other's activities in mice

    Protein phosphatase 5 (PP5), a serine/threonine phosphatase, has a wide range of biological functions and exhibits elevated expression in tumor cells. We previously reported that pp5-deficient mice have altered ataxia-telangiectasia mutated (ATM)-mediated signaling and function. However, this regulation was likely indirect, as ATM is not a known PP5 substrate. In the current study, we found that pp5-deficient mice are hypersensitive to genotoxic stress. This hypersensitivity was associated with the marked up-regulation of the tumor suppressor tumor protein p53 and its downstream targets cyclin-dependent kinase inhibitor 1A (p21), MDM2 proto-oncogene (MDM2), and phosphatase and tensin homolog (PTEN) in pp5-deficient tissues and cells. These observations suggested that PP5 plays a role in regulating p53 stability and function. Experiments conducted with p53 +/- pp5 +/- or p53 +/- pp5 -/- mice revealed that complete loss of PP5 reduces tumorigenesis in the p53 +/- mice. Biochemical analyses further revealed that PP5 directly interacts with and dephosphorylates p53 at multiple serine/threonine residues, resulting in inhibition of p53-mediated transcriptional activity. Interestingly, PP5 expression was significantly up-regulated in p53-deficient cells, and further analysis of pp5 promoter activity revealed that p53 strongly represses PP5 transcription. Our results suggest a reciprocal regulatory interplay between PP5 and p53, providing an important feedback mechanism for the cellular response to genotoxic stress

    The Effect of Raw Soybean on Oxidative Status of Digestive Organs in Mice

    The present study was undertaken to specify the effect of raw soybean on oxidative status of digestive organs in mice. For this purpose, thirty male (C57BL/6J) mice were randomly divided into three groups and fed on different diets as follows: Group 1 was fed on control diet, Group 2 was fed on raw soybean diet and Group 3 was fed on raw soybean diet supplemented with 30 mg/kg cysteamine. After two weeks of feeding, duodenum, liver and pancreas samples were collected to measure oxidative and antioxidative parameters. The results show that ingestion of raw soybean markedly increased contents of superoxide anion and malondialdehyde (MDA) and activity of inducible nitric oxide synthase (iNOS), decreased activity of superoxide dismutase (SOD), T-AOC and content of reduced glutathione (GSH) in digestive organs of mice (P < 0.05). In the group fed with raw soybean diet supplemented with cysteamine, oxidative stress was mitigated. However, oxidative parameter levels were still higher than those of control diet-fed group. The present study indicates that ingestion of raw soybean could result in an imbalance between oxidant and antioxidant, and thus induce oxidative stress in digestive organs of mice

    Evaluating the safety and efficacy of cryopreserved ovarian tissue transplantation in leukemia patients with different bone marrow remission status using xenotransplantation

    BackgroundLeukemia patients undergoing cryopreserved ovarian tissue transplantation (OTT) may carry a high risk of disease induction. Measurable residual disease (MRD) in bone marrow is linked to an elevated risk of relapse. It is controversial whether leukemia patients must be allowed to achieve measurable residual disease negative (MRD-negative) status instead of measurable residual disease positive (MRD-positive) status before ovarian tissue cryopreservation (OTC).ObjectiveTo explore the safety and efficacy of OTT in acute leukemia patients with different MRD status by using xenotransplantation.MethodCryopreserved ovarian tissue from 19 leukemia patients was thawed and xenotransplanted to ovariectomized BALB/C nude mice (n=36). The mice were divided into 2 groups based on the patient’s MRD status before OTC: MRD-negative group (n=18) and MRD-positive group (n=18), additionally, a control group consisted of ovariectomized mice (n=9). Body weight was measured weekly and mortality, emaciation, and other abnormalities were recorded. Twenty-six weeks post-surgery, livers, spleens, uteruses, and ovarian grafts were removed for macroscopic and histological examinations to evaluate the efficacy of xenotransplantation and assess malignant cell contamination in mice.ResultsFollicle growth was visible in the ovarian grafts of the MRD-negative and MRD-positive groups. Compared with the ovariectomized group, a significant decrease in body weight (p&lt;0.01) was noted, the uterine volume was notably larger, estradiol (E2) levels were significantly higher (p&lt;0.01), and follicle-stimulating hormone (FSH) levels were significantly lower (p&lt;0.001) in the other two groups. Mice in the MRD-positive group showed a significantly higher incidence of death (p&lt;0.001) and emaciation (p&lt;0.01), compared to the MRD-negative group. Histological observation revealed the presence of malignant cells in the grafts, livers, and spleens of 3 mice in the MRD-positive group. No abnormalities were observed in the mice from the MRD-negative group in both macroscopic and histological observations except one mouse was sacrificed for ascites unrelated to leukemia relapse.ConclusionFor leukemia patients having ovarian tissue preserved in the first and only centralized human ovarian tissue cryobank in China, immunodeficient mice xenotransplantation can be a method to evaluate the safety and efficacy of OTT; the risk of malignant cell reimplantation due to OTT is higher in leukemia patients with MRD-positive status than those with MRD-negative status before OTC
