    What’s in Your Water? Development and Evaluation of the Virginia Household Water Quality Program and Virginia Master Well Owner Network

    Approximately one-fifth of Virginians (about 1.7 million people) rely on private water supplies (e.g., wells, springs, cisterns) for their household water. Unlike public water systems, the Environmental Protection Agency (EPA) does not regulate private systems. As a result, private water system owners are solely responsible for system maintenance and water quality but are often unaware of common issues and lack access to objective information. We report on the development and evaluation of the Virginia Household Water Quality Program (VAHWQP), an ongoing Virginia Cooperative Extension (VCE) program that provides affordable water testing and education about private water supply system maintenance and groundwater protection. A companion capacity-building program, the Virginia Master Well Owner Network (VAMWON), provides training to volunteers, agency collaborators, and VCE agents who support the goals and objectives of the VAHWQP by conducting VAHWQP drinking water clinics and other outreach efforts. Program assessment findings indicate that VAHWQP drinking water clinic participants regard this programming favorably and are taking recommended actions. We discuss the program assessment framework and continued efforts to improve these programs to achieve long-term behavioral changes regarding water testing and system maintenance, which will yield safer private water supplies and improved environmental stewardship

    Self-organizing hair peg-like structures from dissociated skin progenitor cells: New insights for human hair follicle organoid engineering and Turing patterning in an asymmetric morphogenetic field

    Human skin progenitor cells will form new hair follicles, although at a low efficiency, when injected into nude mouse skin. To better study and improve upon this regenerative process, we developed an in vitro system to analyze the morphogenetic cell behavior in detail and modulate physical-chemical parameters to more effectively generate hair primordia. In this three-dimensional culture, dissociated human neonatal foreskin keratinocytes self-assembled into a planar epidermal layer while fetal scalp dermal cells coalesced into stripes, then large clusters, and finally small clusters resembling dermal condensations. At sites of dermal clustering, subjacent epidermal cells protruded to form hair peg-like structures, molecularly resembling hair pegs within the sequence of follicular development. The hair peg-like structures emerged in a coordinated, formative wave, moving from periphery to center, suggesting that the droplet culture constitutes a microcosm with an asymmetric morphogenetic field. In vivo, hair follicle populations also form in a progressive wave, implying the summation of local periodic patterning events with an asymmetric global influence. To further understand this global patterning process, we developed a mathematical simulation using Turing activator-inhibitor principles in an asymmetric morphogenetic field. Together, our culture system provides a suitable platform to 1) analyze the self-assembly behavior of hair progenitor cells into periodically arranged hair primordia, and 2) identify parameters that impact the formation of hair primordia in an asymmetric morphogenetic field. This understanding will enhance our future ability to successfully engineer human hair follicle organoids

    Caloric Restriction Alters Postprandial Responses of Essential Brain Metabolites in Young Adult Mice

    Caloric restriction (CR) has been shown to extend longevity and protect brain function in aging. However, the effects of CR in young adult mice remain largely unexplored. In addition to the fundamental, long-term changes, recent studies demonstrate that CR has a significant impact on transient, postprandial metabolic flexibility and turnover compared to control groups. The goal of this study was to identify the brain metabolic changes at a transient (2 h) and steady (6 h) postprandial state in young mice (5–6 months of age) fed with CR or ad libitum (AL; free eating). Using metabolomics profiling, we show that CR mice had significantly higher levels of neurotransmitters (e.g., glutamate, N-acetylglutamate), neuronal integrity markers (e.g., NAA and NAAG), essential fatty acids (e.g., DHA and DPA), and biochemicals associated carnitine metabolism (related to reduced oxidative stress and inflammation) in the cerebral cortex and hippocampus at 2-h. These biochemicals remained at high levels at the 6-h postprandial time-point. The AL mice did not show the similar increases in essential fatty acid and carnitine metabolism until the 6-h time-point, and failed to show increases in neurotransmitters and neuronal integrity markers at any time-point. On the other hand, metabolites related to glucose utilization—glycolysis and pentose phosphate pathway (PPP)—were low in the CR mice throughout the 6-h period and significantly increased at the 6-h time-point in the AL mice. Our findings suggest that CR induces distinct postprandial responses in metabolites that are essential to maintain brain functions. CR mice produced higher levels of essential brain metabolites in a shorter period after a meal and sustained the levels for an extended period, while maintaining a lower level of glucose utilization. These early brain metabolism changes in the CR mice might play a critical role for neuroprotection in aging. Understanding the interplay between dietary intervention and postprandial metabolic responses from an early age may have profound implications for impeding brain aging and reducing risk for neurodegenerative disorders

    The role of atmospheric CO2 in controlling sea surface temperature change during the Pliocene,

    We present the role of CO2 forcing in controlling Late Pliocene sea surface temperature (SST) change using six models from Phase 2 of the Pliocene Model Intercomparison Project (PlioMIP2) and palaeoclimate proxy data from the PlioVAR working group. At a global scale, SST change in the Late Pliocene relative to the pre-industrial is predominantly driven by CO2 forcing in the low and mid-latitudes and non-CO2 forcing in the high latitudes. We find that CO2 is the dominant driver of SST change at the vast majority of proxy data sites assessed (17 out of 19), but the relative dominance of this forcing varies between all proxy sites, with CO2 forcing accounting for between 27 % and 82 % of the total change seen. The dearth of proxy data sites in the high latitudes means that only two sites assessed here are predominantly forced by non-CO2 forcing (such as changes to ice sheets and orography), both of which are in the North Atlantic Ocean.We extend the analysis to show the seasonal patterns of SST change and its drivers at a global scale and at a site-specific level for three chosen proxy data sites. We also present a new estimate of Late Pliocene climate sensitivity using site-specific proxy data values. This is the first assessment of site-specific drivers of SST change in the Late Pliocene and highlights the strengths of using palaeoclimate proxy data alongside model outputs to further develop our understanding of the Late Pliocene. We use the best available proxy and model data, but the sample sizes remain limited, and the confidence in our results would be improved with greater data availability
