    Differential Effects of Buffer pH On Ca\u3csup\u3e2+\u3c/sup\u3e-Induced ROS Emission with Inhibited Mitochondrial Complexes I and III

    Excessive mitochondrial reactive oxygen species (ROS) emission is a critical component in the etiology of ischemic injury. Complex I and complex III of the electron transport chain are considered the primary sources of ROS emission during cardiac ischemia and reperfusion (IR) injury. Several factors modulate ischemic ROS emission, such as an increase in extra-matrix Ca2+, a decrease in extra-matrix pH, and a change in substrate utilization. Here we examined the combined effects of these factors on ROS emission from respiratory complexes I and III under conditions of simulated IR injury. Guinea pig heart mitochondria were suspended in experimental buffer at a given pH and incubated with or without CaCl2. Mitochondria were then treated with either pyruvate, a complex I substrate, followed by rotenone, a complex I inhibitor, or succinate, a complex II substrate, followed by antimycin A, a complex III inhibitor. H2O2 release rate and matrix volume were compared with and without adding CaCl2 and at pH 7.15, 6.9, or 6.5 with pyruvate + rotenone or succinate + antimycin A to simulate conditions that may occur during in vivo cardiac IR injury. We found a large increase in H2O2 release with high [CaCl2] and pyruvate + rotenone at pH 6.9, but not at pHs 7.15 or 6.5. Large increases in H2O2 release rate also occurred at each pH with high [CaCl2] and succinate + antimycin A, with the highest levels observed at pH 7.15. The increases in H2O2 release were associated with significant mitochondrial swelling, and both H2O2 release and swelling were abolished by cyclosporine A, a desensitizer of the mitochondrial permeability transition pore (mPTP). These results indicate that ROS production by complex I and by complex III is differently affected by buffer pH and Ca2+ loading with mPTP opening. The study suggests that changes in the levels of cytosolic Ca2+ and pH during IR alter the relative amounts of ROS produced at mitochondrial respiratory complex I and complex III

    Life interrupted and life regained? Coping with stroke at a young age

    Stroke is a leading cause of disability across the developed world, affecting an increasing number of younger people. In this article, we seek to understand the experience of stroke as a disabling life situation among young people and the strategies that they use to recover and cope. Directed content analysis was conducted from interviews with 17 community-dwelling stroke survivors aged 55 years and younger across the United Kingdom. The sample was drawn from a larger maximum variation sample of stroke survivors. Using the sociological concepts of biographical disruption and biographical repair as a guide, excerpts from the interviews pertaining to aspects of the patientsā€™ life that were interrupted, in addition to how they coped with the changes, were selected and analysed. All individuals described an ā€˜ā€˜altered sense of self,ā€™ā€™ a theme that included loss of identity, family disruption, and/or loss of valued activities. Individuals sought to adapt their sense of self by seeking external support, by restoring normality, and/or through positive reflection. Despite the adapted self that emerged, most individuals continued to experience impairments. While young stroke survivors adapt to their illness over time, they continue to experience impairments and disruptions in their personal and work lives.Aholistic model of rehabilitation that helps individuals regain the capacity for everyday activities related to work, family life, and leisure can begin to address the emotional ramifications of diseases such as stroke, restore wellness, and work towards minimizing the burden felt by family caregivers and children

    Volcano impacts on climate and biogeochemistry in a coupled carbonā€“climate model

    Volcanic eruptions induce a dynamical response in the climate system characterized by short-term global reductions in both surface temperature and precipitation, as well as a response in biogeochemistry. The available observations of these responses to volcanic eruptions, such as to Pinatubo, provide a valuable method to compare against model simulations. Here, the Community Climate System Model Version 3 (CCSM3) reproduces the physical climate response to volcanic eruptions in a realistic way, as compared to direct observations from the 1991 eruption of Mount Pinatubo. The model's biogeochemical response to eruptions is smaller in magnitude than observed, but because of the lack of observations, it is not clear why or where the modeled carbon response is not strong enough. Comparison to other models suggests that this model response is much weaker over tropical land; however, the precipitation response in other models is not accurate, suggesting that other models could be getting the right response for the wrong reason. The underestimated carbon response in the model compared to observations could also be due to the ash and lava input of biogeochemically important species to the ocean, which are not included in the simulation. A statistically significant reduction in the simulated carbon dioxide growth rate is seen at the 90% level in the average of 12 large eruptions over the period 1870ā€“2000, and the net uptake of carbon is primarily concentrated in the tropics, with large spatial variability. In addition, a method for computing the volcanic response in model output without using a control ensemble is tested against a traditional methodology using two separate ensembles of runs; the method is found to produce similar results in the global average.     Interactive effects of vascular risk burden and advanced age on cerebral blood flow.

    Vascular risk factors and cerebral blood flow (CBF) reduction have been linked to increased risk of cognitive impairment and Alzheimer's disease (AD); however the possible moderating effects of age and vascular risk burden on CBF in late life remain understudied. We examined the relationships among elevated vascular risk burden, age, CBF, and cognition. Seventy-one non-demented older adults completed an arterial spin labeling MR scan, neuropsychological assessment, and medical history interview. Relationships among vascular risk burden, age, and CBF were examined in a priori regions of interest (ROIs) previously implicated in aging and AD. Interaction effects indicated that, among older adults with elevated vascular risk burden (i.e., multiple vascular risk factors), advancing age was significantly associated with reduced cortical CBF whereas there was no such relationship for those with low vascular risk burden (i.e., no or one vascular risk factor). This pattern was observed in cortical ROIs including medial temporal (hippocampus, parahippocampal gyrus, uncus), inferior parietal (supramarginal gyrus, inferior parietal lobule, angular gyrus), and frontal (anterior cingulate, middle frontal gyrus, medial frontal gyrus) cortices. Furthermore, among those with elevated vascular risk, reduced CBF was associated with poorer cognitive performance. Such findings suggest that older adults with elevated vascular risk burden may be particularly vulnerable to cognitive change as a function of CBF reductions. Findings support the use of CBF as a potential biomarker in preclinical AD and suggest that vascular risk burden and regionally-specific CBF changes may contribute to differential age-related cognitive declines

    The Completely Hackable Amateur Radio Telescope (CHART) Project

    We present the Completely Hackable Amateur Radio Telescope (CHART), a project that provides hands-on radio instrumentation and design experience to undergraduates while bringing accessible radio astronomy experiments to high school students and teachers. Here we describe a system which can detect 21-cm emission from the Milky Way which is optimized for cost and simplicity of construction. Software, documentation, and tutorials are all completely open source to improve the user experience and facilitate community involvement. We demonstrate the design with several observations which we compare with state-of-the-art surveys. The system is shown to detect galactic 21-cm emission in both rural and urban settings

    Observing cosmic string loops with gravitational lensing surveys

    We show that the existence of cosmic strings can be strongly constrained by the next generation of gravitational lensing surveys at radio frequencies. We focus on cosmic string loops, which simulations suggest would be far more numerous than long (horizon-sized) strings. Using simple models of the loop population and minimal assumptions about the lensing cross section per loop, we estimate the optical depth to lensing and show that extant radio surveys such as CLASS have already ruled out a portion of the cosmic string model parameter space. Future radio interferometers, such as LOFAR and especially SKA, may constrain GĪ¼/c2<10āˆ’9G\mu/c^2 < 10^{-9} in some regions of parameter space, outperforming current constraints from pulsar timing and the CMB by up to two orders of magnitude. This method relies on direct detections of cosmic strings, and so is less sensitive to the theoretical uncertainties in string network evolution that weaken other constraints.Comment: 20 pages, 3 figures. v3: Some clarification of text, revised figure, appendix added.
