BACKGROUND: Larix gmelinii is a dominant tree species in China’s boreal forests and plays an important role in the coniferous ecosystem. It is also one of the most economically important tree species in the Chinese timber industry due to excellent water resistance and anti-corrosion of its wood products. Unfortunately, in Northeast China, L. gmelinii often suffers from serious attacks by diseases and insects. The application of exogenous volatile semiochemicals may induce and enhance its resistance against insect or disease attacks; however, little is known regarding the genes and molecular mechanisms related to induced resistance. RESULTS: We performed de novo sequencing and assembly of the L. gmelinii transcriptome using a short read sequencing technology (Illumina). Chemical defenses of L. gmelinii seedlings were induced with jasmonic acid (JA) or methyl jasmonate (MeJA) for 6 hours. Transcriptomes were compared between seedlings induced by JA, MeJA and untreated controls using a tag-based digital gene expression profiling system. In a single run, 25,977,782 short reads were produced and 51,157 unigenes were obtained with a mean length of 517 nt. We sequenced 3 digital gene expression libraries and generated between 3.5 and 5.9 million raw tags, and obtained 52,040 reliable reference genes after removing redundancy. The expression of disease/insect-resistance genes (e.g., phenylalanine ammonialyase, coumarate 3-hydroxylase, lipoxygenase, allene oxide synthase and allene oxide cyclase) was up-regulated. The expression profiles of some abundant genes under different elicitor treatment were studied by using real-time qRT-PCR. The results showed that the expression levels of disease/insect-resistance genes in the seedling samples induced by JA and MeJA were higher than those in the control group. The seedlings induced with MeJA elicited the strongest increases in disease/insect-resistance genes. CONCLUSIONS: Both JA and MeJA induced seedlings of L. gmelinii showed significantly increased expression of disease/insect-resistance genes. MeJA seemed to have a stronger induction effect than JA on expression of disease/insect-resistance related genes. This study provides sequence resources for L. gmelinii research and will help us to better understand the functions of disease/insect-resistance genes and the molecular mechanisms of secondary metabolisms in L. gmelinii

Effects of Nanoparticles on Double-Stranded RNA Stability in Moth Hemolymph

RNA interference (RNAi) is an immune response in which double-stranded RNA (dsRNA) suppresses a target gene. By designing dsRNA to target genes that are necessary for life, dsRNA can potentially be used as an insecticide. RNAi-based insecticides are badly needed because they are more specific than conventional pesticides and because many insects have developed resistance to pesticides. Unfortunately, some insects produce enzymes that degrade dsRNA and prevent the RNAi response (Cooperet al., 2018). Therefore, RNAi-based insecticides currently cannot be used to control all insects. Here we investigate dsRNA stability when incubated in hemolymph ex vivoto determine if degradation of dsRNA is contributing to the inadequate RNAi response exhibited by lepidopterans, such as the European corn borer (ECB, Ostrinia nubilalis). Our findings indicate that dsRNA is significantly degraded in ECB hemolymph, but encapsulation of dsRNA in chitosan-based nanoparticles (CB-NPs) enhances stability. These findings provide insight into RNAi efficiency limitations in insects, and may provide a method to enhance RNAi efficiency in lepidopterans and other RNAi-refractory pests

Yan Li, Amy Z Fan, Lina S BalluzBehavioral Surveillance Branch, Division of Adult and Community Health, National Center for Chronic Disease Prevention and Health Promotion, Centers for Disease Control and Prevention, Atlanta, GA, USAPurpose: To compare the prevalence of age-related eye disease, visual impairment, and eye care service utilization among adults aged 65 and older in Florida with eight other states. Methods: In 2006, nine states conducted the visual impairment and access to eye care module using the Behavioral Risk Factors Surveillance System (BRFSS) survey (N = 62,750). Visual impairment was based on self-reported ability to see distant and near objects. Age-related eye diseases including cataract, glaucoma, macular degeneration, and diabetic retinopathy were self-reported with diagnosis confirmed by a health care professional. Eye care visit or examination was assessed by whether a respondent reported an eye visit or dilated eye examination within the past year. Results: The estimated prevalence of distant and near visual impairment was lower in Florida than in the eight other states (distant: 11.5% vs 15.2%, P < 0.001; near: 22.3% vs 28.7%, P < 0.001). There was no significant difference with the prevalence of age-related macular degeneration and diabetic retinopathy between these two groups. The prevalence of glaucoma and cataract was higher in Florida. The rates of eye care visits (80.5% vs 74.8%, P < 0.01) and dilated eye examinations (74.7% vs 64.0%, P < 0.01) were higher in Florida. After controlling for demographic variables, chronic conditions, insurance, and eye examination, results for elderly in Florida continued to demonstrate less visually impaired. Conclusion: Fewer elderly in Florida reported visual impairment in spite of comparable or higher prevalence of age-related eye diseases with other states. Health care utilization and health insurance for eye care coverage were also higher in Florida, which may account for the phenomena. More research is needed to investigate the association. Keywords: visual impairment, age-related eye disease, eye care service utilization, health insurance, BRFS
