Egy új, a szimbiotikus gümőfejlődésben szerepet játszó ubiquitin ligáz funkcionális jellemzése. = Functional characterization of a newly identified ubiquitin ligase specialized for symbiotic nodule organogenesis.
A szimbiotikus nitrogĂ©nkötĂ©s kialakulása során egy speciális növĂ©nyi szerv, gĂĽmĹ‘ fejlĹ‘dik a pillangĂłsvirágĂşak gyökerĂ©n. A gĂĽmĹ‘ organogenezisĂ©hez Ă©s a szimbionta baktĂ©riumok inváziĂłjához a korábban általunk azonosĂtott Medicago gĂ©n, a LIN funkciĂłja is elengedhetetlen. A gĂ©n termĂ©ke egy ~ 1500 aminosavbĂłl állĂł, U-box domĂ©nt is hordozĂł fehĂ©rje, melyrĹ‘l a projekt során kĂsĂ©rletesen bizonyĂtottuk, hogy E3 ubiquitin ligáz aktivitással bĂr, valamint azonosĂtottunk egy vele kölcsönhatásba lĂ©pĹ‘ szimbiotikus transzkripciĂłs faktort. A kapott eredmĂ©nyek rĂ©vĂ©n a gĂĽmĹ‘ organogenezisĂ©nek egy Ăşj reguláciĂłjárĂłl nyertĂĽnk ismereteket, melyek arra is rávilágĂtanak, hogy az eukariĂłta sejtekben általános ubiqiuitináciĂłs folyamatok hogyan specializálĂłdhattak a növĂ©ny-mikrĂłba szimbiotikus interakciĂłkban. | Symbiotic root nodules are special organs developed on the roots of legume plants upon rhizobial inoculation, in which biological nitrogen fixation takes place. The Medicago LIN gene is proved to be indispensable for the nodule organogenesis and the bacterial invasion by its mutant phenotype. We have identified this gene by positional cloning in M. truncatula and it codes for a large protein of ~ 1500 AA containing a U-box domain. During this project we have demonstrated its E3 ubiquitin ligase activity experimentally. We have also identified a symbiotic transcription factor as an interactor partner of LIN as its potential target molecule. The expression pattern of LIN was explored, its transcriptional regulation was shown not to be important in the course of nodulation. On the other hand accumulated data strongly suggest the translational/post-translational regulation of LIN. Using deletion derivatives we have mapped the functional importance of the different domains of this protein. By analyzing the promoter activities of various symbiotic genes in the lin mutant background we have put its function into the symbiotic context. We have identified homologs of LIN in many different plant species, and trans-complementation experiments revealed that some of them could fully complement the symbiotic function of LIN. The knowledge we gained on the function of LIN shed light to a new type of regulatory pathway needed for the intermediate steps of nodule formation
