    A CASE STUDY ON SUPPORT VECTOR MACHINES VERSUS ARTIFICIAL NEURAL NETWORKS

    The capability of artificial neural networks for pattern recognition of real world problems is well known. In recent years, the support vector machine has been advocated for its structure risk minimization leading to tolerance margins of decision boundaries. Structures and performances of these pattern classifiers depend on the feature dimension and training data size. The objective of this research is to compare these pattern recognition systems based on a case study. The particular case considered is on classification of hypertensive and normotensive right ventricle (RV) shapes obtained from Magnetic Resonance Image (MRI) sequences. In this case, the feature dimension is reasonable, but the available training data set is small, however, the decision surface is highly nonlinear.For diagnosis of congenital heart defects, especially those associated with pressure and volume overload problems, a reliable pattern classifier for determining right ventricle function is needed. RV¡¦s global and regional surface to volume ratios are assessed from an individual¡¦s MRI heart images. These are used as features for pattern classifiers. We considered first two linear classification methods: the Fisher linear discriminant and the linear classifier trained by the Ho-Kayshap algorithm. When the data are not linearly separable, artificial neural networks with back-propagation training and radial basis function networks were then considered, providing nonlinear decision surfaces. Thirdly, a support vector machine was trained which gives tolerance margins on both sides of the decision surface. We have found in this case study that the back-propagation training of an artificial neural network depends heavily on the selection of initial weights, even though randomized. The support vector machine where radial basis function kernels are used is easily trained and provides decision tolerance margins, in spite of only small margins

    Curvelet-Based Texture Classification in Computerized Critical Gleason Grading of Prostate Cancer Histological Images

    Classical multi-resolution image processing using wavelets provides an efficient analysis of image characteristics represented in terms of pixel-based singularities such as connected edge pixels of objects and texture elements given by the pixel intensity statistics. Curvelet transform is a recently developed approach based on curved singularities that provides a more sparse representation for a variety of directional multi-resolution image processing tasks such as denoising and texture analysis. The objective of this research is to develop a multi-class classifier for the automated classification of Gleason patterns of prostate cancer histological images with the utilization of curvelet-based texture analysis. This problem of computer-aided recognition of four pattern classes between Gleason Score 6 (primary Gleason grade 3 plus secondary Gleason grade 3) and Gleason Score 8 (both primary and secondary grades 4) is of critical importance affecting treatment decision and patients’ quality of life. Multiple spatial sampling within each histological image is examined through the curvelet transform, the significant curvelet coefficient at each location of an image patch is obtained by maximization with respect to all curvelet orientations at a given location which represents the apparent curved-based singularity such as a short edge segment in the image structure. This sparser representation reduces greatly the redundancy in the original set of curvelet coefficients. The statistical textural features are extracted from these curvelet coefficients at multiple scales. We have designed a 2-level 4-class classification scheme, attempting to mimic the human expert’s decision process. It consists of two Gaussian kernel support vector machines, one support vector machine in each level and each is incorporated with a voting mechanism from classifications of multiple windowed patches in an image to reach the final decision for the image. At level 1, the support vector machine with voting is trained to ascertain the classification of Gleason grade 3 and grade 4, thus Gleason score 6 and score 8, by unanimous votes to one of the two classes, while the mixture voting inside the margin between decision boundaries will be assigned to the third class for consideration at level 2. The support vector machine in level 2 with supplemental features is trained to classify an image patch to Gleason grade 3+4 or 4+3 and the majority decision from multiple patches to consolidate the two-class discrimination of the image within Gleason score 7, or else, assign to an Indecision category. The developed tree classifier with voting from sampled image patches is distinct from the traditional voting by multiple machines. With a database of TMA prostate histological images from Urology/Pathology Laboratory of the Johns Hopkins Medical Center, the classifier using curvelet-based statistical texture features for recognition of 4-class critical Gleason scores was successfully trained and tested achieving a remarkable performance with 97.91% overall 4-class validation accuracy and 95.83% testing accuracy. This lends to an expectation of more testing and further improvement toward a plausible practical implementation

    A peroxiredoxin cDNA from Taiwanofungus camphorata: role of Cys31 in dimerization

    Peroxiredoxins (Prxs) play important roles in antioxidant defense and redox signaling pathways. A Prx isozyme cDNA (TcPrx2, 745 bp, EF552425) was cloned from Taiwanofungus camphorata and its recombinant protein was overexpressed. The purified protein was shown to exist predominantly as a dimer by sodium dodecyl sulfate-polyacrylamide gel electrolysis in the absence of a reducing agent. The protein in its dimeric form showed no detectable Prx activity. However, the protein showed increased Prx activity with increasing dithiothreitol concentration which correlates with dissociation of the dimer into monomer. The TcPrx2 contains two Cys residues. The Cys(60) located in the conserved active site is the putative active peroxidatic Cys. The role of Cys(31) was investigated by site-directed mutagenesis. The C31S mutant (C(31) → S(31)) exists predominantly as a monomer with noticeable Prx activity. The Prx activity of the mutant was higher than that of the corresponding wild-type protein by nearly twofold at 12 μg/mL. The substrate preference of the mutant was H2O2 > cumene peroxide > t-butyl peroxide. The Michaelis constant (K M) value for H2O2 of the mutant was 0.11 mM. The mutant enzyme was active under a broad pH range from 6 to 10. The results suggest a role of Cys(31) in dimerization of the TcPrx2, a role which, at least in part, may be involved in determining the activity of Prx. The C(31) residue does not function as a resolving Cys and therefore the TcPrx2 must follow the reaction mechanism of 1-Cys Prx. This TcPrx2 represents a new isoform of Prx family

    Neutrino-photon scattering and its crossed processes in a background magnetic field

    We study the neutrino-photon processes such as γγννˉ\gamma\gamma\to \nu\bar{\nu} and νγνγ\nu\gamma\to \nu\gamma in a background magnetic field smaller than the critical magnetic field Bcme2/eB_c\equiv m_e^2/e. Using Schwinger's proper-time method, we extract leading magnetic-field contributions to the above processes. Our result is valid throughout the kinematic regime where both neutrino and photon energies are significantly smaller than mWm_W. A self-transferable blaCMY-2-harboring IncI1 plasmid was identified in S. enterica serotypes Choleraesuis, Typhimurium, Agona, and Enteritidis and contributed to the overall increase of ceftriaxone resistance in salmonellae

    The effect of cigarette price increase on the cigarette consumption in Taiwan: evidence from the National Health Interview Surveys on cigarette consumption

    BACKGROUND: This study uses cigarette price elasticity to evaluate the effect of a new excise tax increase on cigarette consumption and to investigate responses from various types of smokers. METHODS: Our sample consisted of current smokers between 17 and 69 years old interviewed during an annual face-to-face survey conducted by Taiwan National Health Research Institutes between 2000 to 2003. We used Ordinary Least Squares (OLS) procedure to estimate double logarithmic function of cigarette demand and cigarette price elasticity. RESULTS: In 2002, after Taiwan had enacted the new tax scheme, cigarette price elasticity in Taiwan was found to be -0.5274. The new tax scheme brought about an average annual 13.27 packs/person (10.5%) reduction in cigarette consumption. Using the cigarette price elasticity estimate from -0.309 in 2003, we calculated that if the Health and Welfare Tax were increased by another NT$ 3 per pack and cigarette producers shifted this increase to the consumers, cigarette consumption would be reduced by 2.47 packs/person (2.2%). The value of the estimated cigarette price elasticity is smaller than one, meaning that the tax will not only reduce cigarette consumption but it will also generate additional tax revenues. Male smokers who had no income or who smoked light cigarettes were found to be more responsive to changes in cigarette price. CONCLUSIONS: An additional tax added to the cost of cigarettes would bring about a reduction in cigarette consumption and increased tax revenues. It would also help reduce incidents smoking-related illnesses. The additional tax revenues generated by the tax increase could be used to offset the current financial deficiency of Taiwan's National Health Insurance program and provide better public services

    Acute Kidney Injury Biomarkers for Patients in a Coronary Care Unit: A Prospective Cohort Study

    Background: Renal dysfunction is an established predictor of all-cause mortality in intensive care units. This study analyzed the outcomes of coronary care unit (CCU) patients and evaluated several biomarkers of acute kidney injury (AKI), including neutrophil gelatinase-associated lipocalin (NGAL), interleukin-18 (IL-18) and cystatin C (CysC) on the first day of CCU admission. Methodology/Principal Findings: Serum and urinary samples collected from 150 patients in the coronary care unit of a tertiary care university hospital between September 2009 and August 2010 were tested for NGAL, IL-18 and CysC. Prospective demographic, clinical and laboratory data were evaluated as predictors of survival in this patient group. The most common cause of CCU admission was acute myocardial infarction (80%). According to Acute Kidney Injury Network criteria, 28.7 % (43/150) of CCU patients had AKI of varying severity. Cumulative survival rates at 6-month follow-up following hospital discharge differed significantly (p,0.05) between patients with AKI versus those without AKI. For predicting AKI, serum CysC displayed an excellent areas under the receiver operating characteristic curve (AUROC) (0.89560.031, p,0.001). The overall 180-day survival rate was 88.7 % (133/150). Multiple Cox logistic regression hazard analysis revealed that urinary NGAL, serum IL-18, Acute Physiology, Age and Chronic Health Evaluation II (APACHE II) and sodium on CCU admission day one were independent risk factors for 6-month mortality. In terms of 6-month mortality, urinary NGAL had the best discriminatory power, the best Youden index, and the highest overall correctness of prediction

    Expression Profiles of Phosphoenolpyruvate Carboxylase and Phosphoenolpyruvate Carboxylase Kinase Genes in Phalaenopsis, Implications for Regulating the Performance of Crassulacean Acid Metabolism

    Phalaenopsis is one of the most important potted plants in the ornamental market of the world. Previous reports implied that crassulacean acid metabolism (CAM) orchids at their young seedling stages might perform C3 or weak CAM photosynthetic pathways, but the detailed molecular evidence is still lacking. In this study, we used a key species in white Phalaenopsis breeding line, Phalaenopsis aphrodite subsp. formosana, to study the ontogenetical changes of CAM performance in Phalaenopsis. Based on the investigations of rhythms of day/night CO2 exchange, malate contents and phosphoenolpyruvate carboxylase (PEPC) activities, it is suggested that a progressive shift from C3 to CAM occurred as the protocorms differentiated the first leaf. To understand the role of phosphoenolpyruvate carboxylase kinase (PEPC kinase) in relation to its target PEPC in CAM performance in Phalaenopsis, the expression profiles of the genes encoding PEPC (PPC) and PEPC kinase (PPCK) were measured in different developmental stages. In Phalaenopsis, two PPC isogenes were constitutively expressed over a 24-h cycle similar to the housekeeping genes in all stages, whereas the significant day/night difference in PaPPCK expression corresponds to the day/night fluctuations in PEPC activity and malate level. These results suggest that the PaPPCK gene product is most likely involved in regulation of CAM performance in different developmental stages of Phalaenopsis seedlings

    Association between genetic variant on chromosome 12p13 and stroke survival and recurrence: a one year prospective study in Taiwan

    <p>Abstract</p> <p>Background</p> <p>The association between ischemic stroke and 2 single nucleotide polymorphisms (SNPs) on chromosome 12p13, rs12425791 and rs11833579 appears inconsistent across different samples. These SNPs are close to the ninjurin2 gene which may alter the risk of stroke by affecting brain response to ischemic injury. The purpose of this study was to investigate the association between these two SNPs and ischemic stroke risk, as well as prognostic outcomes in a Taiwanese sample.</p> <p>Methods</p> <p>We examined the relations of these two SNPs to the odds of new-onset ischemic stroke, ischemic stroke subtypes, and to the one year risk of stroke-related death or recurrent stroke following initial stroke in a case-control study. A total of 765 consecutive patients who had first-ever ischemic stroke were compared to 977 stroke-free, age-matched controls. SNPs were genotyped by Taqman fluorescent allelic discrimination assay. The association between ischemic stroke and SNPs were analyzed by multivariate logistic regression. Cox proportional hazard model was used to assess the effect of individual SNPs on stroke-related mortality or recurrent stroke.</p> <p>Results</p> <p>There was no significant association between SNP rs12425791 and rs11833579 and ischemic stroke after multiple testing corrections. However, the marginal significant association was observed between SNP rs12425791 and large artery atherosclerosis under recessive model (OR, 2.30; 95%CI, 1.22-4.34; q-value = 0.062). Among the 765 ischemic stroke patients, 59 died or developed a recurrent stroke. After adjustment for age, sex, vascular risk factors and baseline stroke severity, Cox proportional hazard analysis indicated that the hazard ratios were 2.76 (95%CI, 1.34-5.68; q-value, 0.02) and 2.15 (95%CI, 1.15-4.02; q-value, 0.03) for individuals with homozygous variant allele of rs12425791 and rs11833579, respectively.</p> <p>Conclusions</p> <p>This is a precedent study that found genetic variants of rs12425791 and rs11833579 on chromosome 12p13 are independent predictors of stroke-related mortality or stroke recurrence in patients with incident ischemic stroke in Taiwan. Further study is needed to explore the details of the physiological function and the molecular mechanisms underlying the association of this genetic locus with ischemic stroke.</p
