    Study on antioxidant activity of Echinacea purpurea L. extracts and its impact on cell viability

    This study investigates the antioxidant activity of Echinacea Purpurea L. (EP) extracts and its impact on cell viability. The polysaccharides content of EP was 159.8 ± 12.4 mg/g dry weight (DW), with extracts obtained by applying 55% ethanol at 55°C containing 11.0 ±1.0 mg gallic acid equivalent/g DW of total phenolic compound. Trolox equivalent antioxidant capacity, 0.1 mg/mL of EP extracts exhibited only 30% when compared to the ascorbic acid at the same concentration. Reducing power of extractsincreased linearly with its concentration and the concentration at 2.0 mg/mL reached about 65% of ascorbic acid at 0.3 mg/mL. The chelating capacity of ferrous iron (Fe2+) was 70% as good as that of thesynthetic metal chelater EDTA when added to 5.0 mg/mL of EP extracts. The DPPH scavenging capacity showed 85.1% at 0.5 mg/mL of extracts and with half-effective doses (ED50) was measured at 0.23mg/mL. The superoxide anions scavenging capacity of EP extracts was nearly equivalent to ascorbic acid (91.1% vs 93.0%) at the same concentration of 1.6 mg/mL and ED50 was 0.32 and 0.13 mg/mL, respectively. Microculture tetrazolium assays showed extracts had 92% cell viability at 1.6 mg/mL forchicken’s peripheral blood mononuclear cells (PBMCs) and 84% for RAW 264.7 macrophages, neither reaching the IC50 level. In summary, the EP extracts had antioxidant activity similar to that of ascorbic acid, but have no serious effect on inhibiting chicken’s PBMCs viability

    Chaotic motions in the real fuzzy electronic circuits

    Fuzzy electronic circuit (FEC) is firstly introduced, which is implementing Takagi-Sugeno (T-S) fuzzy chaotic systems on electronic circuit. In the research field of secure communications, the original source should be blended with other complex signals. Chaotic signals are one of the good sources to be applied to encrypt high confidential signals, because of its high complexity, sensitiveness of initial conditions, and unpredictability. Consequently, generating chaotic signals on electronic circuit to produce real electrical signals applied to secure communications is an exceedingly important issue. However, nonlinear systems are always composed of many complex equations and are hard to realize on electronic circuits. Takagi-Sugeno (T-S) fuzzy model is a powerful tool, which is described by fuzzy IF-THEN rules to express the local dynamics of each fuzzy rule by a linear system model. Accordingly, in this paper, we produce the chaotic signals via electronic circuits through T-S fuzzy model and the numerical simulation results provided by MATLAB are also proposed for comparison. T-S fuzzy chaotic Lorenz and Chen-Lee systems are used for examples and are given to demonstrate the effectiveness of the proposed electronic circuit. © 2013 Shih-Yu Li et al

    Integrated mRNA and microRNA transcriptome sequencing characterizes sequence variants and mRNA – microRNA regulatory network in nasopharyngeal carcinoma model systems

    Nasopharyngeal carcinoma (NPC) is a prevalent malignancy in Southeast Asia among the Chinese population. Aberrant regulation of transcripts has been implicated in many types of cancers including NPC. Herein, we characterized mRNA and miRNA transcriptomes by RNA sequencing (RNASeq) of NPC model systems. Matched total mRNA and small RNA of undifferentiated Epstein-Barr virus (EBV)-positive NPC xenograft X666 and its derived cell line C666, well-differentiated NPC cell line HK1, and the immortalized nasopharyngeal epithelial cell line NP460 were sequenced by Solexa technology. We found 2812 genes and 149 miRNAs (human and EBV) to be differentially expressed in NP460, HK1, C666 and X666 with RNASeq; 533 miRNA-mRNA target pairs were inversely regulated in the three NPC cell lines compared to NP460. Integrated mRNA/miRNA expression profiling and pathway analysis show extracellular matrix organization, Beta-1 integrin cell surface interactions, and the PI3K/AKT, EGFR, ErbB, and Wnt pathways were potentially deregulated in NPC. Real-time quantitative PCR was performed on selected mRNA/miRNAs in order to validate their expression. Transcript sequence variants such as short insertions and deletions (INDEL), single nucleotide variant (SNV), and isomiRs were characterized in the NPC model systems. A novel TP53 transcript variant was identified in NP460, HK1, and C666. Detection of three previously reported novel EBV-encoded BART miRNAs and their isomiRs were also observed. Meta-analysis of a model system to a clinical system aids the choice of different cell lines in NPC studies. This comprehensive characterization of mRNA and miRNA transcriptomes in NPC cell lines and the xenograft provides insights on miRNA regulation of mRNA and valuable resources on transcript variation and regulation in NPC, which are potentially useful for mechanistic and preclinical studies. © 2014 The Authors.published_or_final_versio

    Shunting of recurrent post-traumatic syringomyelia into the fourth ventricle: a case report

    <p>Abstract</p> <p>Introduction</p> <p>Post-traumatic syringomyelia is a progressive degenerative disorder that is a well-recognized sequela of spinal cord injury. There is currently no optimal intervention capable of producing satisfactory long-term clinical results.</p> <p>Case presentation</p> <p>In this report, we present a 55-year-old Asian man with recurrent syringomyelia after shunt treatment. The syrinx extended from the thoracic cord into the medulla. We used a silicone tube to create a channel connecting the syrinx cavity directly to the fourth ventricle. The patient made a good recovery and follow-up magnetic resonance imaging revealed a considerable diminution in the size of the syrinx.</p> <p>Conclusions</p> <p>We present a new approach that has the potential to improve the outcome of patients with recurrent post-traumatic syringomyelia, who cannot be treated by conventional methods.</p

    Flat bands as a route to high-temperature superconductivity in graphite

    Superconductivity is traditionally viewed as a low-temperature phenomenon. Within the BCS theory this is understood to result from the fact that the pairing of electrons takes place only close to the usually two-dimensional Fermi surface residing at a finite chemical potential. Because of this, the critical temperature is exponentially suppressed compared to the microscopic energy scales. On the other hand, pairing electrons around a dispersionless (flat) energy band leads to very strong superconductivity, with a mean-field critical temperature linearly proportional to the microscopic coupling constant. The prize to be paid is that flat bands can generally be generated only on surfaces and interfaces, where high-temperature superconductivity would show up. The flat-band character and the low dimensionality also mean that despite the high critical temperature such a superconducting state would be subject to strong fluctuations. Here we discuss the topological and non-topological flat bands discussed in different systems, and show that graphite is a good candidate for showing high-temperature flat-band interface superconductivity.Comment: Submitted as a chapter to the book on "Basic Physics of functionalized Graphite", 21 pages, 12 figure

    Background: Prenatal environmental enrichment (EE) has been proven to positively affect but prenatal stress negatively influence the physiological and psychological processes in animals, whose trans-generational genetic mechanism remains unclearly defined. We aimed to investigate and find out key genes underlying the positive-negative effects derived from prenatal interventions.Materials and Methods: Pregnant rats were randomized into EE group (EEG), earthquake simulation group (ESG), herbal group (HG) received herbal supplements in feed after earthquake simulation, and control group (CG).Results: Light Box Defecation Test (LBDT) showed EEG offspring presented less fecal pellets than CG offspring, ESG’s more than CG’s, and HG’s less than ESG (p’s&lt;0.05). Open-field Test (OFT) score of EEG was higher than CG offspring, of ESG’s was lower than CG’s, and HG’s higher than ESG’s. Irf7 and Ninj were screened, which were up-regulated in EEG, down-regulated in ESG (FC&lt;0.5), and were neutralized in HG. Prenatal EE could positively promote the nervous system development, prenatal earthquake simulation could retard the nervous system development and Chinese herbal remedy (JKSQW) which could correct the retardation.Conclusion: The negative-positive prenatal effect could contribute to altered gene expression of Irf7 and Ninj2 which also could play a key role in the improving function of JKSQWfor the kidneys.Keywords: Prenatal stress; Earthquake simulation; Light Box Defecation Test; Open-field Test; Irf7; Ninj

    Kikuchi-Fujimoto Disease: A case report and review of the literature

    Kikuchi-Fujimoto Disease is a rare benign, condition of necrotising histiocytic lymphadenitis. A case of a 55 year old gentleman is described here. He presented with fevers, weight loss and tender cervical lymph nodes. Kikuchi-Fujimoto disease was diagnosed after cervical lymph node biopsy. Symptomatic treatment was provided and an uneventful full recovery was made. Of significant note, this patient's daughter had Kikuchi-Fujimoto disease almost a decade earlier. Although Kikuchi-Fujimoto disease has been reported widely, we believe this is the first familial report in the literature of a case of Kikuchi-Fujimoto disease occurring in a parent and child

    Evaluation of the effectiveness of a novel brain-computer interface neuromodulative intervention to relieve neuropathic pain following spinal cord injury: Protocol for a single-case experimental design with multiple baselines

    Background: Neuropathic pain is a debilitating secondary condition for many individuals with spinal cord injury. Spinal cord injury neuropathic pain often is poorly responsive to existing pharmacological and nonpharmacological treatments. A growing body of evidence supports the potential for brain-computer interface systems to reduce spinal cord injury neuropathic pain via electroencephalographic neurofeedback. However, further studies are needed to provide more definitive evidence regarding the effectiveness of this intervention. Objective: The primary objective of this study is to evaluate the effectiveness of a multiday course of a brain-computer interface neuromodulative intervention in a gaming environment to provide pain relief for individuals with neuropathic pain following spinal cord injury. Methods: We have developed a novel brain-computer interface-based neuromodulative intervention for spinal cord injury neuropathic pain. Our brain-computer interface neuromodulative treatment includes an interactive gaming interface, and a neuromodulation protocol targeted to suppress theta (4-8 Hz) and high beta (20-30 Hz) frequency powers, and enhance alpha (9-12 Hz) power. We will use a single-case experimental design with multiple baselines to examine the effectiveness of our self-developed brain-computer interface neuromodulative intervention for the treatment of spinal cord injury neuropathic pain. We will recruit 3 participants with spinal cord injury neuropathic pain. Each participant will be randomly allocated to a different baseline phase (ie, 7, 10, or 14 days), which will then be followed by 20 sessions of a 30-minute brain-computer interface neuromodulative intervention over a 4-week period. The visual analog scale assessing average pain intensity will serve as the primary outcome measure. We will also assess pain interference as a secondary outcome domain. Generalization measures will assess quality of life, sleep quality, and anxiety and depressive symptoms, as well as resting-state electroencephalography and thalamic γ-aminobutyric acid concentration. Results: This study was approved by the Human Research Committees of the University of New South Wales in July 2019 and the University of Technology Sydney in January 2020. We plan to begin the trial in October 2020 and expect to publish the results by the end of 2021. Conclusions: This clinical trial using single-case experimental design methodology has been designed to evaluate the effectiveness of a novel brain-computer interface neuromodulative treatment for people with neuropathic pain after spinal cord injury. Single-case experimental designs are considered a viable alternative approach to randomized clinical trials to identify evidence-based practices in the field of technology-based health interventions when recruitment of large samples is not feasible

    Population seroprevalence of antibody to influenza A(H7N9) virus, Guangzhou, China

    BACKGROUND: Since the identification in early 2013 of severe disease caused by influenza A(H7N9) virus infection, there have been few attempts to characterize the full severity profile of human infections. Our objective was to estimate the number and severity of H7N9 infections in Guangzhou, using a serological study. METHODS: We collected residual sera from patients of all ages admitted to a hospital in the city of Guangzhou in southern China in 2013 and 2014. We screened the sera using a haemagglutination inhibition assay against a pseudovirus containing the H7 and N9 of A/Anhui/1/2013(H7N9), and samples with a screening titer ≥10 were further tested by standard hemagglutination-inhibition and virus neutralization assays for influenza A(H7N9). We used a statistical model to interpret the information on antibody titers in the residual sera, assuming that the residual sera provided a representative picture of A(H7N9) infections in the general population, accounting for potential cross-reactions. RESULTS: We collected a total of 5360 residual sera from December 2013 to April 2014 and from October 2014 to December 2014, and found two specimens that tested positive for H7N9 antibody at haemagglutination inhibition titer ≥40 and a neutralization titer ≥40. Based on this, we estimated that 64,000 (95 % credibility interval: 7300, 190,000) human infections with influenza A(H7N9) virus occurred in Guangzhou in early 2014, with an infection-fatality risk of 3.6 deaths (95 % credibility interval: 0.47, 15) per 10,000 infections. CONCLUSIONS: Our study suggested that the number of influenza A(H7N9) virus infections in Guangzhou substantially exceeded the number of laboratory-confirmed cases there, albeit with considerable imprecision. Our study was limited by the small number of positive specimens identified, and larger serologic studies would be valuable.
