    Comparison of bacterial culture and 16S rRNA community profiling by clonal analysis and pyrosequencing for the characterization of the dentine caries-associated microbiome

    Culture-independent analyses have greatly expanded knowledge regarding the composition of complex bacterial communities including those associated with oral diseases. A consistent finding from such studies, however, has been the under-reporting of members of the phylum Actinobacteria. In this study, five pairs of broad range primers targeting 16S rRNA genes were used in clonal analysis of 6 samples collected from tooth lesions involving dentine in subjects with active caries. Samples were also subjected to cultural analysis and pyrosequencing by means of the 454 platform. A diverse bacterial community of 229 species-level taxa was revealed by culture and clonal analysis, dominated by representatives of the genera Prevotella, Lactobacillus, Selenomonas, and Streptococcus. The five most abundant species were: Lactobacillus gasseri, Prevotella denticola, Alloprevotella tannerae, S. mutans and Streptococcus sp. HOT 070, which together made up 31.6 % of the sequences. Two samples were dominated by lactobacilli, while the remaining samples had low numbers of lactobacilli but significantly higher numbers of Prevotella species. The different primer pairs produced broadly similar data but proportions of the phylum Bacteroidetes were significantly higher when primer 1387R was used. All of the primer sets underestimated the proportion of Actinobacteria compared to culture. Pyrosequencing analysis of the samples was performed to a depth of sequencing of 4293 sequences per sample which were identified to 264 species-level taxa, and resulted in significantly higher coverage estimates than the clonal analysis.     Volatile constituents and behavioral change induced by Cymbopogon winterianus leaf essential oil in rodents

    Cymbopogon winterianus Jowitt (‘Java citronella’) is an important essential oil yielding aromatic grass cultivated in India and Brazil and its volatile essential oils extracted from its leaves are used in perfumery, cosmetics, pharmaceuticals and flavoring industries. However, there is no report on any psychopharmacological study of C. winterianus leaf essential oil (LEO) available to date. In this study, the pharmacological effects of the LEO were investigated in animal models and its phytochemical analyses. GC-MS analysis showed a mixture of monoterpenes, as citronellal (36.19%), geraniol (32.82%) and citronellol (11.37%). LEO exhibited an inhibitory effect on the locomotor activity of mice, an antinociceptive effect by increasing the reaction time in the writhing and capsaicin tests. All doses induced a significant increase in the sleeping time of animals not having modified however, the latency. The LEO did not alter the remaining time of the animals on the rota-rod apparatus.     A equipe de enfermagem e a comunicação com o paciente traqueostomizado

    OBJETIVO: identificar as percepções da equipe de enfermagem acerca da importância da comunicação no cuidado ao paciente traqueostomizado, de modo a auxiliá-lo na sua recuperação. MÉTODOS: estudo de abordagem quantitativa e qualitativa. A amostra do estudo foi constituída por 51 integrantes da equipe de enfermagem (auxiliares de enfermagem, técnicos e enfermeiros), das Clínicas Médica Feminina e Masculina, Centro de Terapia Semi-Intensiva e Intensiva Adulto, e Neurocirurgia, de um hospital de ensino em Curitiba/PR. As informações foram coletadas por meio de questionário com questões fechadas e abertas. Na associação entre variáveis quantitativas foram considerados o teste Qui-Quadrado, valores de p < 0,05 indicaram significância estatística, os dados qualitativos foram submetidos à proposta de análise de conteúdo. RESULTADOS: os dados mostram que os participantes têm vivências variadas em sua área de atuação e constatam na sua prática cotidiana a necessidade e a importância da interação com o paciente traqueostomizado para sua recuperação. Os resultados apontam que os Auxiliares e Técnicos, desenvolvem mais orientações do que o enfermeiro (p=0,028). CONCLUSÃO: evidenciou-se no estudo que na impossibilidade de comunicação verbal pelo paciente, os participantes do estudo se utilizam de estratégias diferenciadas de comunicação, como gestos, sinalização e comunicação escrita

    The Drosophila melanogaster host model

    The deleterious and sometimes fatal outcomes of bacterial infectious diseases are the net result of the interactions between the pathogen and the host, and the genetically tractable fruit fly, Drosophila melanogaster, has emerged as a valuable tool for modeling the pathogen–host interactions of a wide variety of bacteria. These studies have revealed that there is a remarkable conservation of bacterial pathogenesis and host defence mechanisms between higher host organisms and Drosophila. This review presents an in-depth discussion of the Drosophila immune response, the Drosophila killing model, and the use of the model to examine bacterial–host interactions. The recent introduction of the Drosophila model into the oral microbiology field is discussed, specifically the use of the model to examine Porphyromonas gingivalis–host interactions, and finally the potential uses of this powerful model system to further elucidate oral bacterial-host interactions are addressed
