    SYBR Green-based Real-Time PCR targeting kinetoplast DNA can be used to discriminate between the main etiologic agents of Brazilian cutaneous and visceral leishmaniases

    <p>Abstract</p> <p>Background</p> <p>Leishmaniases control has been hampered by the unavailability of rapid detection methods and the lack of suitable therapeutic and prophylactic measures. Accurate diagnosis, which can distinguish between <it>Leishmania </it>isolates, is essential for conducting appropriate prognosis, therapy and epidemiology. Molecular methods are currently being employed to detect <it>Leishmania </it>infection and categorize the parasites up to genus, complex or species level. Real-time PCR offers several advantages over traditional PCR, including faster processing time, higher sensitivity and decreased contamination risk.</p> <p>Results</p> <p>A SYBR Green real-time PCR targeting the conserved region of kinetoplast DNA minicircles was able to differentiate between <it>Leishmania </it>subgenera. A panel of reference strains representing subgenera <it>Leishmania </it>and <it>Viannia </it>was evaluated by the derivative dissociation curve analyses of the amplified fragment. Distinct values for the average melting temperature were observed, being 78.95°C ± 0.01 and 77.36°C ± 0.02 for <it>Leishmania </it>and <it>Viannia</it>, respectively (p < 0.05). Using the Neighbor-Joining method and Kimura 2-parameters, the alignment of 12 sequences from the amplified conserved minicircles segment grouped together <it>L</it>. (<it>V</it>.) <it>braziliensis </it>and <it>L</it>. (<it>V</it>.) <it>shawii </it>with a bootstrap value of 100%; while for <it>L</it>. (<it>L</it>.) <it>infantum </it>and <it>L</it>. (<it>L</it>.) <it>amazonensis</it>, two groups were formed with bootstrap values of 100% and 62%, respectively. The lower dissociation temperature observed for the subgenus <it>Viannia </it>amplicons could be due to a lower proportion of guanine/cytosine sites (43.6%) when compared to species from subgenus <it>Leishmania </it>(average of 48.4%). The method was validated with 30 clinical specimens from visceral or cutaneous leishmaniases patients living in Brazil and also with DNA samples from naturally infected <it>Lutzomyia </it>spp. captured in two Brazilian localities.</p> <p>Conclusions</p> <p>For all tested samples, a characteristic amplicon melting profile was evidenced for each <it>Leishmania </it>subgenus, corroborating the data from reference strains. Therefore, the analysis of thermal dissociation curves targeting the conserved kinetoplast DNA minicircles region is able to provide a rapid and reliable method to identify the main etiologic agents of cutaneous and visceral leishmaniases in endemic regions of Brazil.</p

    Advancing South American water and climate science through multi-decadal convection-permitting modeling

    The South America Affinity Group (SAAG) was established in early 2019 by the National Center for Atmospheric Research (NCAR) Water Systems Program as a community effort focused on improving hydroclimate science over South America. SAAG supports large research efforts such as the ANDEX Regional Hydroclimate Program (Espinoza et al. 2020) as well as individual research groups. The group started with a dozen members and quickly grew to over 100 participants from more than ten countries. For the past four years, the SAAG has been meeting online every two weeks and has organized sessions at international conferences such as the American Geophysical Union Fall Meeting and the Convection-Permitting Climate Workshop (Prein et al. 2022). At the core of the SAAG effort are two multi-decadal convection-permitting (CP) model simulations with 4-km grid spacing for historical and future climates over the South American continent. Additionally, a major observational data collection effort has been undertaken, including in-situ station data from South American meteorological and water services, gridded products, satellite-based observations, and field campaign data (NCAR, 2023a).     VIII Congresso Estadual Paulista sobre Formação de Educadores

    O tema proposto para discussão durante o VIII Congresso Estadual Paulista sobre a Formação de Educadores – Modos de ser Educador: Artes e Técnicas – Ciências e Políticas envolve a busca de integração entre as diferentes dimensões que compõem a formação do profissional da educação. O trabalho e o sentido da intervenção social desse profissional se constituem em objeto em torno do qual as análises e interpretações vão recolher os fundamentos de debate sobre cultura como totalidade, incluindo: Artes e Técnicas – Ciências e Políticas. A dinâmica que caracteriza a formação de educadores requer a interpretaçãodesses fundamentos para o constante redimensionamento e definição dos objetivos visando aperfeiçoar o papel do profissional educador na sociedade. Por meio de discussões entrediferentes setores ligados à educação é que podem ser avaliados, preservados ou transformados os móveis que norteiam a formação do educador dentro de contextos culturais/sociais específicos, nos diversos campos, artes, técnicas, ciências e políticas. A disponibilidade de um espaço como o VIII Congresso Estadual Paulista sobre a Formação de Educadores para que estudiosos do tema - Educação - se congreguem e debatam a produção de conhecimento sob diversos ângulos, insere e qualifica esse evento como um processo de contribuição para o aprofundamento de estudos na área educacional. Os Congressos Estaduais Paulistas sobre a Formação de Educadores (CEPFE) vêm acontecendo desde 1990 e têm dedicado ênfase especial à formação de profissionais da educação, nos níveis de ensino fundamental, médio e universitário. Tal conformação do Congresso procura extrapolar questões próprias do padrão formal, para a abordagem de formação do profissional da educação embasada na crítica constante, capaz de dinamizar processos e resultados. Essa pretensão ressalta a importância do envolvimento de diferentes áreas do saber numa discussão sobre significados da produção de conhecimento.
