    Development of flow cytometry-based algal bioassays for assessing toxicity of copper in natural waters

    Copper toxicity to the freshwater algae Selenastrum capricornutum and Chlorella sp. and the marine algae Phaeodactylum tricornutum and Dunaliella tertiolecta was investigated using different parameters measured by flow cytometry: cell division rate inhibition, chlorophyll a fluorescence, cell size (i.e., light-scattering), and enzyme activity. These parameters were assessed regarding their usefulness as alternative endpoints for acute (1-24 h) and chronic (48-72 h) toxicity tests. At copper concentrations of 10 μg/L or less, significant inhibition (50%) of the cell division rate was observed after 48- and 72-h exposures for Chlorella sp., S. capricornutum, and P. tricornutum. Bioassays based on increases in algal cell size were also sensitive for Chlorella sp. and P. tricornutum. Copper caused both chlorophyll a fluorescence stimulation (48-h EC50 of 10 ± 1 μg Cu/L for P. tricornutum) and inhibition (48-h EC50 of 14 ± 6 μg Cu/L for S. capricornutum). For acute toxicity over short exposure periods, esterase activity in S. capricornutum using fluorescein diacetate offered a rapid alternative (3-h EC50 of 90 ± 40 μg Cu/L) to growth inhibition tests for monitoring copper toxicity in mine-impacted waters. For all the effect parameters measured, D. tertiolecta was tolerant to copper at concentrations up to its solubility limit in seawater. These results demonstrate that flow cytometry is a useful technique for toxicity testing with microalgae and provide additional information regarding the general mode of action of copper (II) to algal species

    Effects of river water and salinity on the toxicity of deltamethrin to freshwater shrimp, cladoceran, and fish

    Deltamethrin is a pyrethroid insecticide used extensively to control invertebrate pests on cotton and other crops. It is acutely toxic to nontarget aquatic organisms, but existing toxicity data are mostly from toxicity tests using purified laboratory water that differs greatly from the turbid, high-conductivity rivers in the cotton-growing regions of Australia. The aim of this study was to determine whether the water quality variables conductivity, suspended particles, and dissolved organic matter alter the toxicity of deltamethrin to freshwater crustaceans and a fish. We tested three Australian native species: a cladoceran (Ceriodaphnia cf. dubia), a freshwater shrimp (Paratya australiensis), and larvae of the eastern rainbow fish (Melanotaenia duboulayi). Conductivity of the test solutions ranged from 200 to 750 μS/cm, but such changes did not modify the toxicity of deltamethrin to any of the test species. However, the toxicity of deltamethrin to C. cf. dubia and P. australiensis in river water was significantly decreased (1.8-fold to 6.3-fold reduction) compared to that in laboratory water. Variability in the toxicity data limited our ability to detect differences between laboratory and river water for M. duboulayi. Despite reductions in toxicity in natural waters, deltamethrin remained highly toxic [all L(E)C50 values <0.26 μg/L] to all organisms tested; thus, further investigation of the hazard of deltamethrin is warranted. © 2008 Springer Science+Business Media, LLC

    Tropical cyanobacterial blooms: A review of prevalence, problem taxa, toxins and influencing environmental factors

    © 2015, Page Press Publications. All rights reserved. Toxic cyanobacterial blooms are a major issue in freshwater systems in many countries. The potentially toxic species and their ecological causes are likely to be different in tropical zones from those in temperate water bodies; however, studies on tropical toxic cyanobacterial blooms are sporadic and currently there is no global synthesis. In this review, we examined published information on tropical cyanobacterial bloom occurrence and toxin production to investigate patterns in their growth and distribution. Microcystis was the most frequently occurring bloom genus throughout tropical Asia, Africa and Central America, while Cylindrospermopsis and Anabaena blooms occurred in various locations in tropical Australia, America and Africa. Microcystis blooms were more prevalent during the wet season while Cylindrospermopsis blooms were more prevalent during the dry period. Microcystin was the most encountered toxin throughout the tropics. A meta-analysis of tropical cyanobacterial blooms showed that Microcystis blooms were more associated with higher total nitrogen concentrations, while Cylindrospermopsis blooms were more associated with higher maximum temperatures. Meta-analysis also showed a positive linear relationship between levels of microcystin and N:P (nitrate:phosphate) ratio. Tropical African Microcystis blooms were found to have the lowest microcystin levels in relation to biomass and N:P (nitrate:phosphate) compared to tropical Asian, Australian and American blooms. There was also no significant correlation between microcystin concentration and cell concentration for tropical African blooms as opposed to tropical Asian and American blooms. Our review illustrates that some cyanobacteria and toxins are more prevalent in tropical areas.     Orbital superfluidity in the PP-band of a bipartite optical square lattice

    The successful emulation of the Hubbard model in optical lattices has stimulated world wide efforts to extend their scope to also capture more complex, incompletely understood scenarios of many-body physics. Unfortunately, for bosons, Feynmans fundamental "no-node" theorem under very general circumstances predicts a positive definite ground state wave function with limited relevance for many-body systems of interest. A promising way around Feynmans statement is to consider higher bands in optical lattices with more than one dimension, where the orbital degree of freedom with its intrinsic anisotropy due to multiple orbital orientations gives rise to a structural diversity, highly relevant, for example, in the area of strongly correlated electronic matter. In homogeneous two-dimensional optical lattices, lifetimes of excited bands on the order of a hundred milliseconds are possible but the tunneling dynamics appears not to support cross-dimensional coherence. Here we report the first observation of a superfluid in the PP-band of a bipartite optical square lattice with SS-orbits and PP-orbits arranged in a chequerboard pattern. This permits us to establish full cross-dimensional coherence with a life-time of several ten milliseconds. Depending on a small adjustable anisotropy of the lattice, we can realize real-valued striped superfluid order parameters with different orientations Px±PyP_x \pm P_y or a complex-valued Px±iPyP_x \pm i P_y order parameter, which breaks time reversal symmetry and resembles the π\pi-flux model proposed in the context of high temperature superconductors. Our experiment opens up the realms of orbital superfluids to investigations with optical lattice models.Comment: 5 pages, 5 figure

    Correlates of Complete Childhood Vaccination in East African Countries.

    Despite the benefits of childhood vaccinations, vaccination rates in low-income countries (LICs) vary widely. Increasing coverage of vaccines to 90% in the poorest countries over the next 10 years has been estimated to prevent 426 million cases of illness and avert nearly 6.4 million childhood deaths worldwide. Consequently, we sought to provide a comprehensive examination of contemporary vaccination patterns in East Africa and to identify common and country-specific barriers to complete childhood vaccination. Using data from the Demographic and Health Surveys (DHS) for Burundi, Ethiopia, Kenya, Rwanda, Tanzania, and Uganda, we looked at the prevalence of complete vaccination for polio, measles, Bacillus Calmette-Guérin (BCG) and DTwPHibHep (DTP) as recommended by the WHO among children ages 12 to 23 months. We conducted multivariable logistic regression within each country to estimate associations between complete vaccination status and health care access and sociodemographic variables using backwards stepwise regression. Vaccination varied significantly by country. In all countries, the majority of children received at least one dose of a WHO recommended vaccine; however, in Ethiopia, Tanzania, and Uganda less than 50% of children received a complete schedule of recommended vaccines. Being delivered in a public or private institution compared with being delivered at home was associated with increased odds of complete vaccination status. Sociodemographic covariates were not consistently associated with complete vaccination status across countries. Although no consistent set of predictors accounted for complete vaccination status, we observed differences based on region and the location of delivery. These differences point to the need to examine the historical, political, and economic context of each country in order to maximize vaccination coverage. Vaccination against these childhood diseases is a critical step towards reaching the Millennium Development Goal of reducing under-five mortality by two-thirds by 2015 and thus should be a global priority

    &lt;p&gt;Background: High blood pressure is an important determinant of cardiovascular disease risk. Treated hypertensives do not attain a risk level equivalent to normotensives. This may be a consequence of suboptimal blood pressure control to which indiscriminate use of antihypertensive drugs may contribute. Indeed the recent ALLHAT[1]study suggests that thiazides should be given first to virtually all hypertensives. Whether this is correct or whether different antihypertensive therapies should be targeted towards different patients is a major unresolved issue, which we address in this study.&lt;/p&gt; &lt;p&gt;The measurement of the ratio of aldosterone: renin is used to identify hypertensive subjects who may respond well to treatment with the aldosterone antagonist spironolactone. It is not known if subjects with a high ratio have aldosteronism or aldosterone-sensitive hypertension is debated but it is important to know whether spironolactone is superior to other diuretics such as bendroflumethiazide in this setting.&lt;/p&gt; &lt;p&gt;Methods/design: The study is a double-blind, randomised, crossover, controlled trial that will randomise 120 hypertensive subjects to 12 weeks treatment with spironolactone 50 mg once daily and 12 weeks treatment with bendroflumethiazide 2.5 mg once daily. The 2 treatment periods are separated by a 2-week washout period. Randomisation is stratified by aldosterone: renin ratio to include equal numbers of subjects with high and low aldosterone: renin ratios.&lt;/p&gt; &lt;p&gt;Primary Objective – To test the hypothesis that the aldosterone: renin ratio predicts the antihypertensive response to spironolactone, specifically that the effect of spironolactone 50 mg is greater than that of bendroflumethiazide 2.5 mg in hypertensive subjects with high aldosterone: renin ratios.&lt;/p&gt; &lt;p&gt;Secondary Objectives – To determine whether bendroflumethiazide induces adverse metabolic abnormalities, especially in subjects with high aldosterone: renin ratios and if baseline renin measurement predicts the antihypertensive response to spironolactone and/or bendrofluazide.&lt;/p&gt; &lt;p&gt;Discussion: The numerous deleterious effects of hypertension dictate the need for a systematic approach for its treatment. In spite of various therapies, resistant hypertension is widely prevalent. Among various factors, primary aldosteronism is an important cause of resistant hypertension and is now more commonly recognised. More significantly, hypertensives with primary aldosteronism are also exposed to various other deleterious effects of excess aldosterone. Hence treating hypertension with specific aldosterone antagonists may be a better approach in this group of patients. It may lead on to better blood pressures with fewer medications.&lt;/p&gt

However, currently available treatments have low efficacy in achieving sustained response among patients in the early phase of chronic hepatitis B infection when the immune response to hepatitis B virus is weak.     Assessment of the efficacy of an advanced tertiary sewage treatment plant to remove biologically active chemicals using endocrine and genotoxicity bioassays

    There has been much effort to assess the efficacy of sewage treatment plants to remove chemicals with estrogenic activity, but other modes of toxicity have received less attention. This study assessed the efficacy of advanced tertiary technologies to remove estrogen receptor (ER), aryl hydrocarbon receptor (AhR), retinoic receptor (RAR) agonists and genotoxicity using recombinant gene bioassays. Untreated sewage contained ER, AhR and RAR agonists and was genotoxic. Activated sludge treatment removed over 69% of the ER, AhR and RAR aqueous phase activities. Ozonation and biologically activated carbon processes removed genotoxicity and most receptor agonists to below detection limits. Estrogenic activity was associated with the semi-polar fraction of the aqueous phase and non-polar phase of the influent (maximum 30%). AhR and RAR activities were found in both the aqueous and particulate phases of influent. Only RAR activity was frequently found in the polar fraction of aqueous and particulate phases of the process effluent. In the influent, the greatest RAR activity was found in the polar fraction of the aqueous phase (>65%). Genotoxic activity was detected in all influent fractions but rapidly decreased. The results showed that ER, AhR and RAR agonists were greatly reduced by the advanced tertiary treatment processes
