    Impaired Functional Criticality of Human Brain during Alzheimer's Disease Progression

    The progression of Alzheimer's Disease (AD) has been proposed to comprise three stages, subjective cognitive decline (SCD), mild cognitive impairment (MCI), and AD. Was brain dynamics across the three stages smooth? Was there a critical transition? How could we characterize and study functional criticality of human brain? Based on dynamical characteristics of critical transition from nonlinear dynamics, we proposed a vertex-wise Index of Functional Criticality (vIFC) of fMRI time series in this study. Using 42 SCD, 67 amnestic MCI (aMCI), 34 AD patients as well as their age-, sex-, years of education-matched 54 NC, our new method vIFC successfully detected significant patient-normal differences for SCD and aMCI, as well as significant negative correlates of vIFC in the right middle temporal gyrus with total scores of Montreal Cognitive Assessment (MoCA) in SCD. In comparison, standard deviation of fMRI time series only detected significant differences between AD patients and normal controls. As an index of functional criticality of human brain derived from nonlinear dynamics, vIFC could serve as a sensitive neuroimaging marker for future studies; considering much more vIFC impairments in aMCI compared to SCD and AD, our study indicated aMCI as a critical stage across AD progression

    A Sentiment-Change-Driven Event Discovery System

    We present a system that automatically discovers important events that have significantly driven peopleā€™s sentiment changes towards a target using Twitter data (i.e. tweets). This system can also provide the time, importance, and description of events that are associated with peopleā€™s sentiment changes. In this system, a sentiment classifier is used as the sensor to detect the time points of those changes. It is also used as the filter to effectively eliminate a considerable amount of noisy information and select the most informative tweets to be further analyzed for event descriptions. Discovered events are described from the following aspects, 1) the most important tweets ranked by tweet-based TextRank algorithm, 2) the topics generated by the nonnegative matrix factorization, and 3) the most important keywords generated by word-based TextRank algorithm. Compared with traditional event discovery techniques, the experimental results show that this system can effectively discover important patterns from tweets and unveil 3Ws of an event (i.e. what happens, when it happens, what its effect is), which provides good reference on understanding behavior changes and making strategies. Furthermore, the system was applied to analyze peopleā€™s sentiment changes towards the two candidates during the 2016 U.S. presidential election. It can also be applied in other scenarios where peopleā€™s attitude plays an important role like the brand influence marketing and financial investment markets

    Nucleus accumbens shell small conductance potassium channels underlie adolescent ethanol exposure-induced anxiety

    Alcohol use typically begins in adolescence, increasing the likelihood of adult mental disorders such as anxiety. However, the cellular mechanisms underlying the consequences of adolescent alcohol exposure as well as the behavioral consequences remain poorly understood. We examined the effects of adolescent or adult chronic intermittent ethanol (CIE) exposure on intrinsic excitability of striatal medium-sized spiny neurons (MSNs) and anxiety levels. Rats underwent one of the following procedures: (1) light-dark transition (LDT) and open-field (OF) tests to evaluate anxiety levels and general locomotion; (2) whole-cell patch clamp recordings and biocytin labeling to assess excitability of striatal MSNs, as well as morphological properties; and (3) western blot immunostaining to determine small conductance (SK) calcium-activated potassium channel protein levels. Three weeks, but not 2 days, after CIE treatment, adolescent CIE-treated rats showed shorter crossover latency from the light to dark side in the LDT test and higher MSN excitability in the nucleus accumbens shell (NAcS). Furthermore, the amplitude of the medium afterhyperpolarization (mAHP), mediated by SK channels, and SK3 protein levels in the NAcS decreased concomitantly. Finally, increased anxiety levels, increased excitability, and decreased amplitude of mAHP of NAcS MSNs were reversed by SK channel activator 1-EBIO and mimicked by the SK channel blocker apamin. Thus, adolescent ethanol exposure increases adult anxiety-like behavior by downregulating SK channel function and protein expression, which leads to an increase of intrinsic excitability in NAcS MSNs. SK channels in the NAcS may serve as a target to treat adolescent alcohol binge exposure-induced mental disorders, such as anxiety in adulthood

    A class of pseudoinverse-free greedy block nonlinear Kaczmarz methods for nonlinear systems of equations

    In this paper, we construct a class of nonlinear greedy average block Kaczmarz methods to solve nonlinear problems without computing the Moore-Penrose pseudoinverse. This kind of methods adopts the average technique of Gaussian Kaczmarz method and combines with the greedy strategy, which greatly reduces the amount of computation. The convergence analysis and numerical experiments of the proposed method are given. The numerical results show the effectiveness of the proposed methods

    Sliding at first order: Higher-order momentum distributions for discontinuous image registration

    In this paper, we propose a new approach to deformable image registration that captures sliding motions. The large deformation diffeomorphic metric mapping (LDDMM) registration method faces challenges in representing sliding motion since it per construction generates smooth warps. To address this issue, we extend LDDMM by incorporating both zeroth- and first-order momenta with a non-differentiable kernel. This allows to represent both discontinuous deformation at switching boundaries and diffeomorphic deformation in homogeneous regions. We provide a mathematical analysis of the proposed deformation model from the viewpoint of discontinuous systems. To evaluate our approach, we conduct experiments on both artificial images and the publicly available DIR-Lab 4DCT dataset. Results show the effectiveness of our approach in capturing plausible sliding motion

    Thickness dependence of superconductivity and superconductor-insulator transition in ultrathin FeSe films on SrTiO3(001) substrate

    Interface-enhanced high-temperature superconductivity in one unit-cell (UC) FeSe film on SrTiO3(001) (STO) substrate has recently attracted much attention in condensed matter physics and material science. Here, by ex situ transport measurements, we report on the superconductivity in FeSe ultra-thin films with different thickness on STO substrate. We find that the onset superconducting transition temperature (Tc) decreases with increasing film thickness of FeSe, which is opposite to the behavior usually observed in traditional superconductor films. By systematic post-annealing of 5 UC FeSe films, we observe an insulator to superconductor transition, which is accompanied with a sign change of the dominated charge carriers from holes to electrons at low temperatures according to the corresponding Hall measurement

    MicroRNA-29a Inhibits Growth, Migration and Invasion of Melanoma A375 Cells in Vitro by Directly Targeting BMI1

    Background/Aims: Melanoma is one of the most aggressive malignant tumors, with increasing incidence, poor prognosis, and lack of any effective targeted therapies. Abnormal expression of miR-29a has been found in several types of cancers, including melanoma. In this study, experiments were performed to investigate the role of miR-29a in melanoma, and the molecular mechanism by which miR-29a represses melanoma. Methods: miR-29 and Bmi1 expression was examined by quantitative real-time polymerase chain reaction (qRT-PCR). The cell viability, apoptosis, migration and invasion were respectively determined by Cell Counting Kit-8 assay, Propidium iodide (PI) fluorescein isothiocynate (FITC)-Annexin V staining assay, wound healing assay and transwell assay. Luciferase reporter assay was performed to determine a target gene of miR-29a. Western blot was used to analyze protein expression of apoptosis-related proteins, Bmi1, Wnt/Ī²-catenin and Nuclear factor-ĪŗB (NF-ĪŗB) pathway target genes. Results: miR-29a was down-regulated in all tested melanoma cell lines. Up-regulation of miR-29a effectively inhibited cell viability, migration, and invasion, but promoted apoptosis in A375 cells. Bmi1 was a direct target gene of miR-29a. Transfection with miR-29a mimic decreased cell migration and invasion and Bmi1 expression in Malme-3M cells, SK-MEL-2, SK-MEL-5, and M14 cell lines. Moreover, miR-29a might suppress growth, migration and invasion of A375 cells by negatively regulating Bmi1. In addition, our results demonstrated that transfection with miR-29a mimic effectively blocked Wnt/Ī²-catenin and NF-ĪŗB pathways via down-regulating Bmi1. Conclusion: miR-29a could be functioned as a potential tumor suppressor through direct regulation of Bmi1 in melanoma cells
