Growth factor/growth factor receptor loops in autocrine growth regulation of human prostate cancer DU145 cells
Autocrine growth factors produced by epithelial cells mediate the development and proliferation of neoplastic human prostate tissue. Various approaches have been used to down-regulate neoplastic growth of prostate cancer using natural flavonoids, soluble receptors, pseudo-ligands, monoclonal antibodies and tyrosine kinase inhibitors (tyrphostins). Selected growth factor/growth factor receptor loops (mainly TGFα/EGFR and IGFs/IGFIR) have been proposed as regulators of prostate cancer cell growth. We have previously determined that blockade of IGFIR or VEGF2R signaling pathways by tyrphostin AG1024 and SU1498 inhibits autocrine growth and viability of DU145 cells in vitro. Recently, we compared the activity of AG1024 and SU1498 with the inhibiting effect of tyrphostin A23 (a selective inhibitor of EGFR). The results described in this paper confirm that DU145 cells do not produce IGFI or EGF. In contrast, DU145 cells produce a great amount of VEGF, much more than TGFα (about 60-fold), and VEGF may be the real autocrine growth factor of the investigated cells. The results indicate that the growth of DU145 may be regulated by at least three autocrine loops: TGFα/EGFR, IGFII/IGFIR and VEGF/VEGFR2. Neither AG1024 nor SU1498 affected the production of TGFα substantially, which excludes the possibility that IGFRs or VEGFR2 inhibitors arrest the growth of these cells by inhibition of synthesis and/or secretion of TGFα. The obtained data indicate that all tree investigated tyrphostins (AG1024, SU1498 and A23) inhibit signal transmission by Akt (PKB), ERK(1/2), Src and STAT in a similar manner. A comparison of the effects of the investigated tyrphostins indicates that TGFα, IGFII and VEGF stimulate cell growth by affecting the same signaling pathway. The hypothesis was confirmed by the effect of the investigated tyrphostins on activation of EGFR. All these inhibitors decreased phosphorylation of EGFR to the same extent, and after the same time of incubation with cell culture. These results strongly suggest that stimulation of EGFR kinase is the main step in the initiation of mitogen signaling in DU145 cells, regardless of the type of ligand (TGFα, IGFs or VEGF) and their specific receptors
The effect of tyrosine kinase inhibitors, tyrphostins : AG1024 and SU1498, on autocrine growth of prostate cancer cells (DU145)
It is well established that autocrine growth of human prostate cancer cell line DU145 is dependent on TGF (EGF)/EGFR loop. However, the participation of several other growth factors in proliferation of DU145 cells has been also proposed. We employed two selective tyrosine kinase inhibitors (tyrphostins): AG1024 (an IGFIR inhibitor) and SU1498 (a VEGFR2 inhibitor) for growth regulation of DU145 cells, cultured in chemically defined DMEM/F12 medium. Both the tested compounds inhibited autocrine growth of DU145 cells at similar concentration values (IC50 approximately 2.5 microM). The tyrphostins arrested cell growth of DU145 in G1 phase, similarly as inhibitors of EGFR. However, in contrast to selective inhibitors of EGFR, neither AG1024, nor SU1498 (at concentration < or =10 microM) decreased the viability of the investigated cells. These results strongly suggest that autocrine growth of DU145 cells is stimulated by, at least, three autocrine loops: TGFalpha(EGF)/EGFR, IGFII/IGFIr and VEGF/VEGFR2(VEGFR1). These data support the hypothesis of multi-loops growth regulation of metastatic prostate cancer cell lines
The effect of tyrphostins AG494 and AG1478 on the autocrine growth regulation of A549 and DU145 cells
We employed two selective EGFR tyrosine kinase inhibitors: AG494 (reversible) and AG1478 (irreversible) for growth regulation of human lung (A549) and prostate (DU145) cancer cell lines, cultured in chemically defined DMEM/F12 medium. Both tested tyrphostins significantly inhibited autocrine growth of the investigated cell lines. The action of AG494 was dose dependent, and at highest concentrations led to complete inhibition of growth. AG1478 seemed to be more effective at lower concentrations, but was unable to completely inhibit growth of A549 cells. Inhibition of EGFR kinase activity by AG494 in contrast to AG1478 had no effect on the activity of ERK in both cell lines. Both EGFR’s inhibitors induced apoptosis of the investigated lung and prostate cancer cell lines, but the proapoptotic effect of the investigated tyrphostins was greater in A549 than in DU145 cells. The tyrphostins arrested cell growth of DU145 and A549 cells in the G1 phase, similarly to other known inhibitors of EGFR. The influence of AG494 and AG1478 on the activity of two signaling proteins (AKT and ERK) was dependent upon the kind of investigated cells. In the case of DU145 cells, there was an evident decline in enzymatic activity of both kinases (stronger for AG1478), while in A549, only AG1478 effectively inhibited the phosphorylation of Akt. Tyrphostins AG494 and AG1478 are ATP-competitors and are supposed to have a similar mechanism of action, but our results suggest that this is not quite true
Zakażenie Chlamydia pneumoniae u chorych z miażdżycowym zwężeniem tętnicy szyjnej wewnętrznej
Background. Infection with Chlamydia pneumoniae, a common human respiratory
tract pathogen, has been recently associated with cardiovascular syndromes, such
as atherosclerosis, myocardial infarction and stroke. According to the literature
data significantly high percentage of CAD patients exhibited serological markers
of chronic Chlamydia pneumoniae infection. Chlamydia pneumoniae DNA and antigens
were also detected (using variety of methods) in carotid arteries atherosclerotic
plaques. The aim of the study was to evaluate the frequency of Chlamydia pneumoniae
infection in CAD patients by measuring specific serum IgG, IgM and IgA levels
and by detection of Chlamydia pneumoniae DNA in carotid atherosclerotic plaques.
Material and methods. 28 patients operated for CAD (8 female, 20 male;
mean age 64.5 years) and 20 control subjects matched for age and sex (4 female,
16 male) without clinical signs and symptoms of cardiovascular and pulmonary disease
were enrolled in the study. Microimmunofluorescence method was applied to evaluate
the level of anti-Chlamydia pneumoniae IgG, IgA and IgM. Chlamydia pneumoniae
micro-IF test (Labsystem) was used. Nested PCR was used for Chlamydia pneumoniae
DNA detection in atherosclerotic plaques, obtained during carotid endartherectomy.
The results were evaluated by means of STATISTICA software programme and considered
statistically significant when was less than 0.05 (p < 0.05).
Results. Serologic markers of chronic Chlamydia pneumoniae infection were
detected in 22 of 28 (78.6%) patients and in 6 of 20 (30%) healthy controls. In
36.4% (8/22) of patients with serologic markers of chronic Chlamydia pneumoniae
infection we observed high titers of specific IgG and IgA, which can indicate
reinfection or exacerbation of chronic infection. Interestingly, all patients
in this group presented to have TIA symptoms. Chlamydia pneumoniae DNA was present
in carotid atherosclerotic plaques obtained from 17 (60.7%) patients.
Conclusions. Serological signs of chronic Chlamydia pneumoniae infection
occur statistically more frequent in CAD patients compared to healthy controls.
Active Chlamydia pneumoniae infection is associated with TIA symptoms. There is
an urgent need of standardization of PCR methods detecting DNA Chlamydia pneumoniae
in atherosclerotic plaques.Wstęp. Wzrastająca liczba danych seroepidemiologicznych i doświadczalnych
wskazuje na możliwy udział Chlamydia pneumoniae, niedawno odkrytego ludzkiego
patogenu wewnątrzkomórkowego, w patogenezie chorób układu sercowo-naczyniowego,
takich jak miażdżyca, zawał serca i udar mózgu. Według danych z piśmiennictwa
u znacznego odsetka chorych z miażdżycowym zwężeniem tętnicy szyjnej wewnętrznej
wykryto serologiczne wskaźniki przewlekłego zakażenia Chlamydia pneumoniae. Korzystając
z różnych metod, wykryto również DNA i antygeny Chlamydia pneumoniae w blaszkach
miażdżycowych zlokalizowanych w obrębie tętnic szyjnych wewnętrznych. Celem pracy
była ocena częstości występowania zakażenia Chlamydia pneumoniae u chorych z miażdżycowym
zwężeniem tętnicy szyjnej wewnętrznej poprzez oznaczenie miana swoistych przeciwciał
klasy IgG, IgM i IgA oraz poprzez detekcję DNA Chlamydia pneumoniae w blaszkach
miażdżycowych, usuniętych podczas zabiegu udrożnienia tętnic szyjnych wewnętrznych.
Materiał i metody. Badaniem objęto 28 chorych z miażdżycowym zwężeniem
tętnicy szyjnej wewnętrznej (8 kobiet, 20 mężczyzn), średni wiek chorych wynosił
64,5 roku. Grupa kontrolna składała się z 20 osób, zbliżonych pod względem wieku
i płci do grupy badanej, bez objawów chorób układu sercowo-naczyniowego i oddechowego.
Ocenę miana swoistych przeciwciał anty-Chlamydia pneumoniae klasy IgG, IgA i IgM
przeprowadzono, stosując metodę mikroimmunofluorescencji, za pomocą testu Chlamydia
pneumoniae Micro-IF firmy Labsystems (Finlandia). Do wykrywania DNA Chlamydia
pneumoniae w blaszkach miażdżycowych użyto metody nested PCR. W analizie statystycznej
wykorzystano program Statistica. Za istotne statystycznie przyjęto p < 0,05.
Wyniki. Serologiczne wskaźniki przewlekłego zakażenia Chlamydia pneumoniae
stwierdzono u 22 spośród 28 chorych (78,6%) oraz u 6 spośród 20 osób z grupy kontrolnej
(30%). U 36,4% pacjentów (8/22), u których rozpoznano serologicznie przewlekłe
zakażenie Chlamydia pneumoniae, zaobserwowano wysokie miana swoistych przeciwciał
klasy IgG i IgA, wskazujące na reinfekcję lub zaostrzenie przewlekłego zakażenia.
U wszystkich chorych w tej grupie odnotowano objawy przemijającego niedokrwienia
mózgu. DNA Chlamydia pneumoniae stwierdzono u 17 (60,7%) chorych z miażdżycowym
zwężeniem tętnicy szyjnej wewnętrznej.
Wnioski Serologiczne wskaźniki przewlekłego zakażenia Chlamydia
pneumoniae występują statystycznie częściej u chorych z miażdżycowym zwężeniem
tętnicy szyjnej wewnętrznej niż w grupie kontrolnej. Aktywne zakażenie Chlamydia
pneumoniae wiąże się z występowaniem objawów przemijającego niedokrwienia mózgu.
Istnieje pilna potrzeba standaryzacji metod PCR wykrywających DNA Chlamydia pneumoniae
w blaszkach miażdżycowych

Ze wstępu: "W ostatnich latach termin „bezpieczeństwo narodowe” odmieniany był w licznych
publikacjach naukowych i publicystycznych we wszystkich przypadkach i w każdej
prawie możliwej konfiguracji. Pisano zatem i rozprawiano na konferencjach oraz
seminariach naukowych o bezpieczeństwie zewnętrznym i wewnętrznym, ekonomicznym
i energetycznym, społecznym, demograficznym i wielu innych. Praktycznie
każdy obszar problemowy, któremu można było przypisać, z większym lub
mniejszym sensem, rzeczownik „bezpieczeństwo” stawał się przedmiotem rozważań
w ramach dyscypliny naukowej „nauki o bezpieczeństwie”. W tym natłoku
„gatunków” bezpieczeństwa coraz mniej widoczne było bezpieczeństwo militarne,
czyli innymi słowy obronność państwa. Dopiero wydarzenia na wschód od naszej
granicy doprowadziły do swego rodzaju przebudzenia i od dwóch lat ponownie
zaczęto głośno zastanawiać się nad istotą, poziomem i organizacją obronności
Rzeczypospolitej. Wszystkie te wydarzenia doprowadziły do powstania swego
rodzaju sytuacji problemowej, którą można by opisać w postaci pytania: w jakiej
