Modification of Loop 1 Affects the Nucleotide Binding Properties of Myo1c, the Adaptation Motor in the Inner Ear
Myo1c is one of eight members of the mammalian myosin I family of actin-associated molecular motors. In stereocilia of the hair cells in the inner ear, Myo1c presumably serves as the adaptation motor, which regulates the opening and closing of transduction channels. Although there is conservation of sequence and structure among all myosins in the N-terminal motor domain, which contains the nucleotide- and actin-binding sites, some differences include the length and composition of surface loops, including loop 1, which lies near the nucleotide-binding domain. To investigate the role of loop 1, we expressed in insect cells mutants of a truncated form of Myo1c, Myo1c1IQ, as well as chimeras of Myo1c1IQ with the analogous loop from other myosins. We found that replacement of the charged residues in loop 1 with alanines or the whole loop with a series of alanines did not alter the ATPase activity, transient kinetics properties, or Ca2+ sensitivity of Myo1c1IQ. Substitution of loop 1 with that of the corresponding region from tonic smooth muscle myosin II (Myo1c1IQ-tonic) or replacement with a single glycine (Myo1c1IQ-G) accelerated the release of ADP from A.M 2?3-fold in Ca2+, whereas substitution with loop 1 from phasic muscle myosin II (Myo1c1IQ-phasic) accelerated the release of ADP 35-fold. Motility assays with chimeras containing a single ?-helix, or SAH, domain showed that Myo1cSAH-tonic translocated actin in vitro twice as fast as Myo1cSAH-WT and 3-fold faster than Myo1cSAH-G. The studies show that changes induced in Myo1c via modification of loop 1 showed no resemblance to the behavior of the loop donor myosins or to the changes previously observed with similar Myo1b chimeras
Laser induced fluorescence for axion dark matter detection: a feasibility study in YLiF:Er
We present a detection scheme to search for QCD axion dark matter, that is
based on a direct interaction between axions and electrons explicitly predicted
by DFSZ axion models. The local axion dark matter field shall drive transitions
between Zeeman-split atomic levels separated by the axion rest mass energy . Axion-related excitations are then detected with an upconversion scheme
involving a pump laser that converts the absorbed axion energy (
hundreds of eV) to visible or infrared photons, where single photon
detection is an established technique. The proposed scheme involves rare-earth
ions doped into solid-state crystalline materials, and the optical transitions
take place between energy levels of electron configuration. Beyond
discussing theoretical aspects and requirements to achieve a cosmologically
relevant sensitivity, especially in terms of spectroscopic material properties,
we experimentally investigate backgrounds due to the pump laser at temperatures
in the range K. Our results rule out excitation of the upper Zeeman
component of the ground state by laser-related heating effects, and are of some
help in optimizing activated material parameters to suppress the
multiphonon-assisted Stokes fluorescence.Comment: 8 pages, 5 figure
A Description Logic of Typicality for Conceptual Combination
We propose a nonmonotonic Description Logic of typicality able to
account for the phenomenon of combining prototypical concepts, an open problem
in the fields of AI and cognitive modelling. Our logic extends the logic of
typicality ALC + TR, based on the notion of rational closure, by inclusions
p :: T(C) v D (âwe have probability p that typical Cs are Dsâ), coming
from the distributed semantics of probabilistic Description Logics. Additionally,
it embeds a set of cognitive heuristics for concept combination. We show that the
complexity of reasoning in our logic is EXPTIME-complete as in ALC
Star Architecture as Socio-Material Assemblage
Taking inspiration from new materialism and assemblage, the chapter deals with star architects and iconic buildings as socio-material network effects that do not pre-exist action, but are enacted in practice, in the materiality of design crafting and city building. Star architects are here conceptualized as part of broader assemblages of actors and practices âmaking star architectureâ a reality, and the buildings they design are considered not just as unique and iconic objects, but dis-articulated as complex crafts mobilizing skills, technologies, materials, and forms of knowledge not necessarily ascribable to architecture. Overcoming narrow criticism focusing on the symbolic order of icons as unique creations and alienated repetitions of capitalist development, the chapterâs main aim is to widen the scope of critique by bridging culture and economy, symbolism and practicality, making star architecture available to a broad, fragmented arena of (potential) critics, unevenly equipped with critical tools and differentiated experiences
Particle detection through the quantum counter concept in YAG:Er
We report about a novel scheme for particle detection based on the infrared
quantum counter concept. Its operation consists of a two-step excitation
process of a four level system, that can be realized in rare earth-doped
crystals when a cw pump laser is tuned to the transition from the second to the
fourth level. The incident particle raises the atoms of the active material
into a low lying, metastable energy state, triggering the absorption of the
pump laser to a higher level. Following a rapid non-radiative decay to a
fluorescent level, an optical signal is observed with a conventional detectors.
In order to demonstrate the feasibility of such a scheme, we have investigated
the emission from the fluorescent level S (540 nm band) in an
Er-doped YAG crystal pumped by a tunable titanium sapphire laser when it
is irradiated with 60 keV electrons delivered by an electron gun. We have
obtained a clear signature this excitation increases the
metastable level population that can efficiently be exploited to generate a
detectable optical signal
Ontologies, Mental Disorders and Prototypes
As it emerged from philosophical analyses and cognitive research, most concepts exhibit typicality effects, and resist to the efforts of defining them in terms of necessary and sufficient conditions. This holds also in the case of many medical concepts. This is a problem for the design of computer science ontologies, since knowledge representation formalisms commonly adopted in this field do not allow for the representation of concepts in terms of typical traits. However, the need of representing concepts in terms of typical traits concerns almost every domain of real world knowledge, including medical domains. In particular, in this article we take into account the domain of mental disorders, starting from the DSM-5 descriptions of some specific mental disorders. On this respect, we favor a hybrid approach to the representation of psychiatric concepts, in which ontology oriented formalisms are combined to a geometric representation of knowledge based on conceptual spaces
