    Bioequivalence and Population Pharmacokinetic Modeling of Two Forms of Antibiotic, Cefuroxime Lysine and Cefuroxime Sodium, after Intravenous Infusion in Beagle Dogs

    To investigate the bioequivalence and the population pharmacokinetics of cefuroxime lysine and cefuroxime sodium in healthy beagle dogs. A randomized 2-period crossover design in 18 healthy beagle dogs after receiving 20, 40, and 80 mg/kg of cefuroxime lysine or cefuroxime sodium was conducted. A 3-compartment open model was used as the basic model for the population pharmacokinetic study. Both of the antibiotics exhibited dose-proportional pharmacokinetics over the dose range of 20–80 mg/kg. The mean relative bioavailability of cefuroxime lysine versus cefuroxime sodium was 1.05 (range, 0.71 to 1.42), with a significant difference between males and females. The estimates of population pharmacokinetic of CL, V1, Q2, V2, Q3, V3 were 3.74 mL/h, 1.70 mL, 29.5 mL/min, 3.58 mL, 0.31 mL/min, and 158 mL for cefuroxime lysine and 4.10 mL/h, 1.00 mL, 38.5 mL/min, 4.19 mL, 0.06 mL/min, and 13.6 mL for cefuroxime sodium, respectively. The inter-individual variability was determined to be less than 29.1%. A linear pharmacokinetic was revealed for cefuroxime lysine and cefuroxime sodium in dogs after intravenous infusion, and the bioequivalence of these forms of the antibiotic was observed with the significant gender-related differences in mean relative bioavailability of cefuroxime lysine versus cefuroxime sodium

    Spatial genetic subdivision among populations of Pampus chinensis between China and Pakistan: testing the barrier effect of the Malay Peninsula

    Tissue samples from 84 Pampus chinensis individuals were collected from four geographic regions within the Indo–Pacific Ocean and analyzed using mitochondrial and nuclear DNA markers. Distinct genetic heterogeneity was found for both types of markers between Chinese and Pakistani populations, while the diversity of this species was high in all populations. In combination with published information on marine species with similar distributions, these results suggested that the Malay Peninsula, or a less effective supplement, played a role in shaping the contemporary genetic structure. This population structure was presumably reflected in P. chinensis, whose populations were genetically isolated during Pleistocene glaciations and then did not experience secondary contact between previous refuge populations. However, P. chinensis showed genetic continuity in China or Pakistan, which indicated that the populations in different geographical regions constituted a single panmictic stock with high gene flow, respectively. The spatial genetic subdivision evident among populations indicates that P. chinensis in this Indo–Pacific region should be managed as different independent stocks to guide the sustainability of this fisheries resource

    Advances in Chinese Arctic and subarctic research in marine biology and ecology with emphasis on the Pacific Arctic sector

    The Arctic is one of the most sensitive regions that respond through feedback to global climate changes. Climatic, hydrological and ecological changes in the Arctic are clear evidence of global warming. In 2012 and 2014, the 5th and 6th Chinese National Arctic Research Expeditions undertook studies in the Bering Sea, the Arctic Ocean (including the Chukchi Sea), and the Norwegian Sea. These studies provided us with a better understanding of the marine biology and ecology in the Arctic and subarctic regions, particularly in the Pacific Arctic sector. Rapid changes observed in the Arctic environment include the shrinking of cold-water masses in the Bering Sea in the summer, and elevated water temperatures promoting phytoplankton blooms, leading to an increase in phytoplankton transferred to higher trophic levels. As a result, the transfer efficiency of organic matter toward the bottom weakened, leading to a reduction in benthic biomass. This is consistent with expectations that the overall carbon and energy flux will ultimately switch from the dominant mode of sea ice–algae–benthos to one of phytoplankton–zooplankton. Influenced by Pacific water inflow, fluvial runoff and melting sea ice, the Chukchi Sea exhibited different responses to various environmental changes. Interactions between water masses led to other interannual ecological shifts. With the increase in sea ice melt and sunlight in the central region of the Arctic Ocean, the relative abundance of heterotrophic bacteria is expected to increase, and play a vital role in the Arctic microbial loop

    First record of Pampus minor (Actinopterygii: Perciformes: Stromateidae) from the coastal waters of Wenzhou, China

    Pampus fishes (Perciformes: Stromateidae) are important commercial species and include six valid species in China. The southern lesser pomfret (Pampus minor Liu and Li, 1998) is a species of Pampus for which knowledge is incomplete. This report confirms the occurrence of P. minor in the coastal waters of Wenzhou, China, by analyzing five specimens of P. minor obtained in Wenzhou in March 2019. Morphometric measurements and meristic counts were performed. The diagnostic morphological features of these species were consistent with those of the neotype specimen of P. minor and sufficient for separating the examined specimens from other Pampus species. This study was based on the Cytochrome Oxidase Subunit 1 (CO1) gene, which was sequenced for the purpose of identification. The genetic distances between P. minor and other Pampus species ranged from 13.4% to 15.5%, and the mean genetic distance within the P. minor group was 0.2%. Given that P. minor has not been reported in this region, our findings represent the first record from the coastal waters of Wenzhou and extend the distribution range of this species to the central and northern East China Sea. The reason for the observed northward migration of some P. minor individuals from their northernmost recorded habitat (Taiwan Strait) might be global warming. The collection of additional specimens is needed to further study the geographic limits of P. minor and its presumed northern expansion

    Bench surgery with autotransplantation for bilateral Wilms tumor—A feasible technique for renal sinus invasion

    PurposeBilateral Wilms tumor (BWT) with renal sinus invasion requires extremely difficult surgical care. This study presents an alternative strategy for tumor removal while at the same time preserving the renal parenchyma.Materials and methodsIn total, 9 cases of synchronous BWT were admitted to our hospital between May 2016 to Aug 2020. We retrospectively reviewed the clinical data, surgical technique, and functional and oncological outcomes of these cases.ResultsThe 9 cases included 3 males and 6 females, with a median age of 12 months at surgery (range 7–40). A total of 14 kidney units had renal sinus invasion (77.8%), whereas multifocal neoplasms were observed in 7 units (38.9%). The local stage distribution revealed 1 kidney with stage I, 10 kidneys with stage II, and 7 kidneys with stage III. Nephron-sparing surgery was performed on 15 kidney units (83.3%), among which 13 (72.2%) underwent bench surgery with autotransplantation (BS-AT), whereas 2 (11.1%) were subjected to tumor enucleation in vivo. Urinary leakage was the most prevalent postoperative complication. We observed negative margins. During the mean follow-up of 28.4 months, 2 patients (22.2%) succumbed from sepsis and renal failure, respectively, whereas the other 7 (77.8%) survived without recurrence. Survivors experienced an estimated glomerular filtration rate of 81 ± 15.4 ml/(min × 1.73 m2). The endpoint renal volume of 9 renal units receiving BS-AT significantly increased (P = 0.02).ConclusionsIn summary, the surgical management of bilateral Wilms tumor requires meticulous operative approach and technique. Besides, BS-AT provides a viable alternative to nephron-sparing surgery for BWT patients with renal sinus invasion

    Clinical Characteristics of 26 Human Cases of Highly Pathogenic Avian Influenza A (H5N1) Virus Infection in China

    BACKGROUND: While human cases of highly pathogenic avian influenza A (H5N1) virus infection continue to increase globally, available clinical data on H5N1 cases are limited. We conducted a retrospective study of 26 confirmed human H5N1 cases identified through surveillance in China from October 2005 through April 2008. METHODOLOGY/PRINCIPAL FINDINGS: Data were collected from hospital medical records of H5N1 cases and analyzed. The median age was 29 years (range 6-62) and 58% were female. Many H5N1 cases reported fever (92%) and cough (58%) at illness onset, and had lower respiratory findings of tachypnea and dyspnea at admission. All cases progressed rapidly to bilateral pneumonia. Clinical complications included acute respiratory distress syndrome (ARDS, 81%), cardiac failure (50%), elevated aminotransaminases (43%), and renal dysfunction (17%). Fatal cases had a lower median nadir platelet count (64.5 x 10(9) cells/L vs 93.0 x 10(9) cells/L, p = 0.02), higher median peak lactic dehydrogenase (LDH) level (1982.5 U/L vs 1230.0 U/L, p = 0.001), higher percentage of ARDS (94% [n = 16] vs 56% [n = 5], p = 0.034) and more frequent cardiac failure (71% [n = 12] vs 11% [n = 1], p = 0.011) than nonfatal cases. A higher proportion of patients who received antiviral drugs survived compared to untreated (67% [8/12] vs 7% [1/14], p = 0.003). CONCLUSIONS/SIGNIFICANCE: The clinical course of Chinese H5N1 cases is characterized by fever and cough initially, with rapid progression to lower respiratory disease. Decreased platelet count, elevated LDH level, ARDS and cardiac failure were associated with fatal outcomes. Clinical management of H5N1 cases should be standardized in China to include early antiviral treatment for suspected H5N1 cases

    Pampus argenteus is a broadly exploited pelagic fish species, commonly misidentified as Pampus echinogaster. Genetic variation and population structure in Pampus argenteus was studied based on seven microsatellite loci. The observed high average allele number, heterozygosity values, and polymorphism information content of P. argenteus suggested high genetic diversity. No population genetic differentiation was detected based on the results of pairwise Fst, three-dimensional factorial correspondence analysis (3D-FCA) and STRUCTURE analysis, which implied continuous gene flow. Wilcoxon signed rank tests did not indicate significant heterozygosity excess, and recent genetic bottleneck events were not detected. Coupled with previous mitochondrial DNA results, the findings presented here indicate that high gene flow characterizes the current phylogeographic pattern of the species

    Determination of Antioxidant 264 in the Butyl Rubber Stopper and the Compatibility with Recombinant Potent Antitumor and Antivirus Protein Injection

    Objective. To establish a method for extraction and determination of antioxidant 264 (2,6-di-tert-butyl-4-methylphenol) in the brominated butyl rubber stopper for injection and migration study in recombinant potent antitumor and antivirus protein injection (Novaferon). Methods. Dichloromethane-ethanol was adopted as the extraction solvent during the process of reflux extraction of antioxidant 264 in the brominated butyl rubber stopper. High-performance liquid chromatography (HPLC) was used for the determination of the migration of antioxidant 264 to Novaferon. The mobile phase consisted of acetonitrile-water (80 : 20, v/v). The flow rate was 1.5 mL/min. The detection wavelength was 280 nm. Results. The linearity range was from 4.003 to 200.150 μg/mL (r2 = 0.99996), and the average recovery of antioxidant 264 was 97.8%. The applicability of the methodology was good, which can be used for the determination of antioxidant 264. The results indicated that antioxidant 264 was not detected in Novaferon after the accelerated test and three months of long-term test. Conclusion. The established validated method in this study can be used for the determination of antioxidant 264 in the rubber stopper, and the brominated butyl rubber stopper has good compatibility with Novaferon
