When some parameters are properly chosen as special values, the known double solitary-like wave solutions are derived from the double hyperbolic function solutions

    Effects of supplemental lighting with different light qualities on growth and secondary metabolite content of Anoectochilus roxburghii

    Background Anoectochilus roxburghii is a widespread herbaceous plant with high medicinal value. Wild A. roxburghii resources face extinction due to their slow growth rate and over exploitation. The growing market demand has led to advances in the field of artificial planting of A. roxburghii. Methods to increase the economic benefits of cultivation and the production of medicinal ingredients are very useful. Methods A. roxburghii was exposed to red light, blue light (BL), yellow light (YL), green light, or white light as supplemental lighting at night (18:00–02:00) in a greenhouse or were left in darkness (control, CK) to investigate the effects of various light qualities on growth indices, photosynthetic pigments, chlorophyll fluorescence, root vitality, stomatal density, soluble proteins, sugars, and the accumulation of secondary metabolites. Results Supplementation of BL had a positive effect on A. roxburghii growth and secondary metabolite accumulation. Leaf number, stem diameter, fresh weight, dry weight, chlorophyll a content, and secondary metabolite (total flavonoids, total polyphenols) content increased significantly. YL treatment showed significantly higher soluble sugar and polysaccharide contents than the control. Discussion BL treatment was conducive to promoting the growth and accumulation of secondary metabolites (total flavonoids, total polyphenols); YL treatment significantly increased the content of soluble sugar and polysaccharides more than the control. Polysaccharides and total flavonoids are important medicinal ingredients of Anoectochilus, so future research will focus on the combination of blue and YL

    Perturb-Seq: Dissecting Molecular Circuits with Scalable Single-Cell RNA Profiling of Pooled Genetic Screens

    Genetic screens help infer gene function in mammalian cells, but it has remained difficult to assay complex phenotypes—such as transcriptional profiles—at scale. Here, we develop Perturb-seq, combining single-cell RNA sequencing (RNA-seq) and clustered regularly interspaced short palindromic repeats (CRISPR)-based perturbations to perform many such assays in a pool. We demonstrate Perturb-seq by analyzing 200,000 cells in immune cells and cell lines, focusing on transcription factors regulating the response of dendritic cells to lipopolysaccharide (LPS). Perturb-seq accurately identifies individual gene targets, gene signatures, and cell states affected by individual perturbations and their genetic interactions. We posit new functions for regulators of differentiation, the anti-viral response, and mitochondrial function during immune activation. By decomposing many high content measurements into the effects of perturbations, their interactions, and diverse cell metadata, Perturb-seq dramatically increases the scope of pooled genomic assays.     Identification and characterization of miRNA169 family members in banana (Musa acuminata L.) that respond to fusarium oxysporum f. sp. cubense infection in banana cultivars

    MicroRNAs (miRNAs) play an important role in plant resistance to pathogen infections. However, little is known about the role of miRNAs in banana Fusarium wilt, which is the most economically devastating disease in banana production. In the present study, we identified and characterized a total of 18 miR169 family members in banana (Musa acuminata L.) based on small RNA sequencing. The banana miR169 family clustered into two groups based on miRNA evolutionary analysis. Multiple sequence alignment indicated a high degree of sequence conservation in miRNA169 family members across 28 plant species. Computational target prediction algorithms were used to identify 25 targets of miR169 family members in banana. These targets were enriched in various metabolic pathways that include the following molecules: glycine, serine, threonine, pentose, glycerolipids, nucleotide sugars, starch, and sucrose. Through miRNA transcriptomic analysis, we found that ma-miR169a and ma-miR169b displayed high expression levels, whereas the other 16 ma-miR169 members exhibited low expression in the HG and Baxi banana cultivars. Further experiments indicate that there were negative relationships between ma-miR169a, ma-miR169b and their targets basing on their expression levels to Foc4 (Fusarium oxysporum f. sp. cubense tropical race 4) infection in resistant cultivars. But they were low expressed in susceptive cultivars. These results suggested that the expression levels of ma-miR169a and ma-miR169b were consistent with the resistance degree of the banana cultivars to Foc4. The analysis presented here constitutes a starting point to understand ma-miR169-mediated Fusarium wilt resistance at the transcriptional level in banana and predicts possible candidate targets for the genetic improvement of banana resistance to Foc4

    Abnormal Degree Centrality Associated With Cognitive Dysfunctions in Early Bipolar Disorder

    Delayed diagnosis of bipolar disorder (BD) is common. However, diagnostic validity may be enhanced using reliable neurobiological markers for BD. Degree centrality (DC) is one such potential marker that enables researchers to visualize neuronal network abnormalities in the early stages of some neuropsychiatric disorders. In the present study, we measured resting-state DC abnormalities and cognitive deficits in order to identify early neurobiological markers for BD. We recruited 23 patients with BD who had recently experienced manic episodes (duration of illness <2 years) and 46 matched healthy controls. Our findings indicated that patients with BD exhibited DC abnormalities in frontal areas, temporal areas, the right postcentral gyrus, and the posterior lobe of the cerebellum. Moreover, correlation analysis revealed that psychomotor speed indicators were associated with DC in the superior temporal and inferior temporal gyri, while attention indicators were associated with DC in the inferior temporal gyrus, in patients with early BD. Our findings suggest that DC abnormalities in neural emotion regulation circuits are present in patients with early BD, and that correlations between attention/psychomotor speed deficits and temporal DC abnormalities may represent early markers of BD

    With the help of big data and Citespace software, this research makes a statistical analysis of the journals and dissertations on College English teaching and learning materials collected by CNKI from 2011 to 2020. This paper, based on the knowledge map drawn by the visualized analysis of literatures volume, authors, research institutions and keywords clustering, analyzes the current research status and hotspots in the compilation of China’s College English textbooks. It is found that there are six problems in the compilation. Because of the complexity of teaching and learning materials and the dynamic progress of language learning, the nature of English teaching and learning materials is bound to turn from a learning tool to learning resources. Thus, this research, from the perspective of Complex Dynamic System Theory, attempts to develop big data-based College English learning materials with digital, individual and multi-dimensional characteristics by three paths: the establishment of big data-based English learning behaviors index system, the development of big data-based College English learning materials and the application of big data-based College English learning materials. This paper will explore a new way of developing China’s College English learning materials, improve and optimize the compilation and development of College English learning materials in China
