    Sediment and nutrient retention capacity of natural riverine wetlands in Southwest Ethiopia

    Although wetlands in temperate regions have been studied for their pollutant remediation potential, the sediment and nutrient retention capacity of natural wetlands in tropical environments remains understudied. In this study, a mass balance approach was used to estimate the amount of sediment and nutrients retained at 40 different study sites located in four natural riverine wetlands in Southwest Ethiopia. Awetu and Boye wetlands retained a substantial amount of total suspended solids (TSS) and nutrients whereas Kofe and Kito wetlands had a net release. Overall, the amount of TSS retained by the four wetlands was estimated to be 6266.5 kg/ha/day. On the other hand, the net retention of Total Organic Nitrogen (TON) and Total Phosphorous (TP) was estimated to be 37.6 kg/ha/day and 40.9 kg/ha/day, respectively. A stepwise multiple regression analysis revealed that anthropogenic activities such as farming, grazing, waste dumping and clay mining were the main variables that negatively affected the retention of TSS and nutrients. Farming alone explained 58% of the variation in TSS retention and cattle grazing explained 25% of the variation in TON retention.     Anti-apoptotic B-cell lymphoma 2 (Bcl-2) family members target several intracellular Ca2+-transport systems. Bcl-2, via its N-terminal Bcl-2 homology (BH) 4 domain, inhibits both inositol 1,4,5-trisphosphate receptors (IP(3)Rs) and ryanodine receptors (RyRs), while Bcl-X-L, likely independently of its BH4 domain, sensitizes IP3Rs. It remains elusive whether Bcl-XL can also target and modulate RyRs. Here, Bcl-X-L co-immunoprecipitated with RyR3 expressed in HEK293 cells. Mammalian protein-protein interaction trap (MAPPIT) and surface plasmon resonance (SPR) showed that Bcl-XL bound to the central domain of RyR3 via its BH4 domain, although to a lesser extent compared to the BH4 domain of Bcl-2. Consistent with the ability of the BH4 domain of Bcl-X-L to bind to RyRs, loading the BH4-Bcl-X-L peptide into RyR3-overexpressing HEK293 cells or in rat hippocampal neurons suppressed RyR-mediated Ca2+ release. In silico superposition of the 3D-structures of Bcl-2 and Bcl-XL indicated that Lys87 of the BH3 domain of Bcl-XL could be important for interacting with RyRs. In contrast to Bcl-X-L, the Bcl-X-L(K87D) mutant displayed lower binding affinity for RyR3 and a reduced inhibition of RyR-mediated Ca2+ release. These data suggest that Bcl-X-L binds to RyR channels via its BH4 domain, but also its BH3 domain, more specific Lys87, contributes to the interaction

    Enzymatic Regulation of Protein-Protein Interactions in Artificial Cells

    Membraneless organelles are important for spatial organization of proteins and regulation of intracellular processes. Proteins can be recruited to these condensates by specific protein–protein or protein–nucleic acid interactions, which are often regulated by post-translational modifications. However, the mechanisms behind these dynamic, affinity-based protein recruitment events are not well understood. Here, a coacervate system that incorporates the 14-3-3 scaffold protein to study enzymatically regulated recruitment of 14-3-3-binding proteins is presented, which mostly bind in a phosphorylation-dependent manner. Synthetic coacervates are efficiently loaded with 14-3-3, and phosphorylated binding partners, such as the c-Raf pS233/pS259 peptide (c-Raf), show 14-3-3-dependent sequestration with up to 161-fold increase in local concentration. The c-Raf domain is fused to green fluorescent protein (GFP-c-Raf) to demonstrate recruitment of proteins. In situ phosphorylation of GFP-c-Raf by a kinase leads to enzymatically regulated uptake. The introduction of a phosphatase into coacervates preloaded with the phosphorylated 14-3-3-GFP-c-Raf complex results in a significant cargo efflux mediated by dephosphorylation. Finally, the general applicability of this platform to study protein–protein interactions is demonstrated by the phosphorylation-dependent and 14-3-3-mediated active reconstitution of a split-luciferase inside artificial cells. This work presents an approach to study dynamically regulated protein recruitment in condensates, using native interaction domains.</p

    Large branchiopod occurrence and community structure in relation to land-use types in temporary ponds of northern Tanzania

    This research article was published by Springer Nature Switzerland AG., 2023Large branchiopods are a key component of the fauna of temporary ponds and play an important role in the functioning of these vulnerable ecosystems. Owing to the establishment of new settlements and agricultural expansion, temporary ponds in Tanzania are disappearing at an alarming rate whilst little is known about their diversity and ecology. We contrasted temporary ponds from a protected area with those in communal lands to detect associations between land-use types and large branchiopod community structure. Six large branchiopod species were collected, five of which have been previously reported from Southern Africa, whilst one turned out to be new to science: Streptocephalus manyarensis n.sp. Kafula and Brendonck (2023). The clam shrimp Cyzicus sp., fairy shrimps Streptocephalus lamellifer Thiele (1900) and S. bourquinii Hamer and Appleton (1993) were the most abundant and widely occurring. Variation in large branchiopod community structure was explained by the presence of Nothobranchius killifish and orthophosphate concentration. The large branchiopod community structure was different in settlement and protected areas. Our study on the occurrence and structure of large branchiopod communities in relation to land-use types serves as a base for formulation of guidelines and management tools to regulate land-use practices adjacent to temporary pond ecosystems

    AURORA : bariatric surgery registration in women of reproductive age : a multicenter prospective cohort study

    Background: The expansion of the obesity epidemic is accompanied with an increase in bariatric procedures, in particular in women of reproductive age. The weight loss induced by the surgery is believed to reverse the negative impact of overweight and obesity on female reproduction, however, research is limited to in particular retrospective cohort studies and a growing number of small case-series and case-(control) studies. Methods/design: AURORA is a multicenter prospective cohort study. The main objective is to collect long-term data on reproductive outcomes before and after bariatric surgery and in a subsequent pregnancy. Women aged 18-45 years are invited to participate at 4 possible inclusion moments: 1) before surgery, 2) after surgery, 3) before 15 weeks of pregnancy and 4) in the immediate postpartum period (day 3-4). Depending on the time of inclusion, data are collected before surgery (T1), 3 weeks and 3, 6, 12 or x months after surgery (T2-T5) and during the first, second and third trimester of pregnancy (T6-T8), at delivery (T9) and 6 weeks and 6 months after delivery (T10-T11). Online questionnaires are send on the different measuring moments. Data are collected on contraception, menstrual cycle, sexuality, intention of becoming pregnant, diet, physical activity, lifestyle, psycho-social characteristics and dietary supplement intake. Fasting blood samples determine levels of vitamin A, D, E, K, B-1, B-12 and folate, albumin, total protein, coagulation parameters, magnesium, calcium, zinc and glucose. Participants are weighted every measuring moment. Fetal ultrasounds and pregnancy course and complications are reported every trimester of pregnancy. Breastfeeding is recorded and breast milk composition in the postpartum period is studied.
