Bioorthogonal Double-Fluorogenic Siliconrhodamine Probes for Intracellular Superresolution Microscopy
A series of double-fluorogenic siliconrhodamine probes were
synthesized. These tetrazine-functionalized, membrane-permeable
labels allowed site-specific bioorthogonal tagging of genetically
manipulated intracellular proteins and subsequent imaging using
super-resolution microscopy
Quasispecies evolution in general mean-field landscapes
I consider a class of fitness landscapes, in which the fitness is a function
of a finite number of phenotypic "traits", which are themselves linear
functions of the genotype. I show that the stationary trait distribution in
such a landscape can be explicitly evaluated in a suitably defined
"thermodynamic limit", which is a combination of infinite-genome and strong
selection limits. These considerations can be applied in particular to identify
relevant features of the evolution of promoter binding sites, in spite of the

Stomatin-like Protein 2 Links Mitochondria to T-Cell Receptor Signalosomes at the Immunological Synapse and Enhances T-Cell Activation
T cell activation through the antigen receptor (TCR) requires sustained signalling from microclusters in the peripheral region of the immunological synapse (IS). The bioenergetics of such prolonged signaling have been linked to the redistribution of mitochondria to the IS. Here, we report that stomatin-like protein-2 (SLP-2) plays an important role in this process by bridging polarized mitochondria to these signaling TCR microclusters or signalosomes in the IS in a polymerized actin-dependent manner. In this way, SLP-2 helps to sustain TCR-dependent signalling and enhances T cell activation
Ordered phase in the two-dimensional randomly coupled ferromagnet
True ground states are evaluated for a 2d Ising model with random near
neighbor interactions and ferromagnetic second neighbor interactions (the
Randomly Coupled Ferromagnet). The spin glass stiffness exponent is positive
when the absolute value of the random interaction is weaker than the
ferromagnetic interaction. This result demonstrates that in this parameter
domain the spin glass like ordering temperature is non-zero for these systems,

Quantitative system drift compensates for altered maternal inputs to the gap gene network of the scuttle fly Megaselia abdita.

Available from eLife Sciences Publications via the DOI in this record.The segmentation gene network in insects can produce equivalent phenotypic outputs despite differences in upstream regulatory inputs between species. We investigate the mechanistic basis of this phenomenon through a systems-level analysis of the gap gene network in the scuttle fly Megaselia abdita (Phoridae). It combines quantification of gene expression at high spatio-temporal resolution with systematic knock-downs by RNA interference (RNAi). Initiation and dynamics of gap gene expression differ markedly between M. abdita and Drosophila melanogaster, while the output of the system converges to equivalent patterns at the end of the blastoderm stage. Although the qualitative structure of the gap gene network is conserved, there are differences in the strength of regulatory interactions between species. We term such network rewiring 'quantitative system drift'. It provides a mechanistic explanation for the developmental hourglass model in the dipteran lineage. Frequency evaluation of the doubly forbidden transition in bosonic Yb
We report an uncertainty evaluation of an optical lattice clock based on the
transition in the bosonic isotope Yb by use
of magnetically induced spectroscopy. The absolute frequency of the
transition has been determined through comparisons
with optical and microwave standards at NIST. The weighted mean of the
evaluations is (Yb)=518 294 025 309 217.8(0.9) Hz. The uncertainty
due to systematic effects has been reduced to less than 0.8 Hz, which

Boron Abundances in Main Sequence B-type Stars: A Test of Rotational Depletion during Main Sequence Evolution
Boron abundances have been derived for seven main sequence B-type stars from
HST STIS spectra around the B III 2066 A line. In two stars, boron appears to
be undepleted with respect to the presumed initial abundance. In one star,
boron is detectable but it is clearly depleted. In the other four stars, boron
is undetectable implying depletions of 1 to 2 dex. Three of these four stars
are nitrogen enriched, but the fourth shows no enrichment of nitrogen. Only
rotationally induced mixing predicts that boron depletions are unaccompanied by
nitrogen enrichments. The inferred rate of boron depletion from our
observations is in good agreement with these predictions. Other boron-depleted
nitrogen-normal stars are identified from the literature. Also, several
boron-depleted nitrogen-rich stars are identified, and while all fall on the
boron-nitrogen trend predicted by rotationally-induced mixing, a majority have
nitrogen enrichments that are not uniquely explained by rotation.
The spectra have also been used to determine iron-group (Cr, Mn, Fe, and Ni)
abundances. The seven B-type stars have near solar iron-group abundances, as
expected for young stars in the solar neighborhood. We have also analysed the
halo B-type star, PG0832+676. We find [Fe/H] = -0.88 +/- 0.10, and the absence
of the B III line gives the upper limit [B/H]<2.5. These and other published
