    Disruption of Nrf2 Enhances the Upregulation of Nuclear Factor-kappaB Activity, Tumor Necrosis Factor-α, and Matrix Metalloproteinase-9 after Spinal Cord Injury in Mice

    Matrix metalloproteinase-9 (MMP-9) plays an important role in the acute periods of spinal cord injury (SCI), and its expression is related to the inflammation which could cause the disruption of the blood-spinal barrier (BBB). Nuclear factor erythroid 2-related factor 2 (Nrf2) is a key transcription factor that plays a crucial role in cytoprotection against inflammation. The present study investigated the role of Nrf2 in upregulating of nuclear factor kappa B (NF-κB) activity, tumor necrosis factor-α (TNF-α), and MMP-9 after SCI. Wild-type Nrf2 (+/+) and Nrf2-deficient (Nrf (−/−)) mice were subjected to an SCI model induced by the application of vascular clips (force of 10 g) to the dura after a three-level T8-T10 laminectomy. We detected the wet/dry weight ratio of impaired spinal cord tissue, the activation of NF-κB, the mRNA and protein levels of TNF-α and MMP-9, and the enzyme activity of MMP-9. Nrf2 (−/−) mice were demonstrated to have more spinal cord edema, NF-κB activation, TNF-α production, and MMP-9 expression after SCI compared with the wild-type controls. The results suggest that Nrf2 may play an important role in limiting the upregulation of NF-κB activity, TNF-α, and MMP-9 in spinal cord after SCI

    The Design of Reference Service System in Cordova-based Hybrid Frameworks

    With the rise of mobile technology, the library reference service has dramatically changed. Targeting the new requirements, this paper aims to design a new library reference service system in Cordova-based hybrid frameworks, which caters to the web service embedded in two major mobile platforms, iOS and Android, as well as the PC platform. The new system adopts the WebSocket based technology to realize the function of independent online reference, which improves the quality of the normal digital reference service. The newly designed system also applies the ECS cloud server technology, thereby significantly slashing the hardware setup cost, extending the basic reference service, and improving its fitness-for-use and convenience, and optimizing the allocation of local resources

    LibSignal: An Open Library for Traffic Signal Control

    This paper introduces a library for cross-simulator comparison of reinforcement learning models in traffic signal control tasks. This library is developed to implement recent state-of-the-art reinforcement learning models with extensible interfaces and unified cross-simulator evaluation metrics. It supports commonly-used simulators in traffic signal control tasks, including Simulation of Urban MObility(SUMO) and CityFlow, and multiple benchmark datasets for fair comparisons. We conducted experiments to validate our implementation of the models and to calibrate the simulators so that the experiments from one simulator could be referential to the other. Based on the validated models and calibrated environments, this paper compares and reports the performance of current state-of-the-art RL algorithms across different datasets and simulators. This is the first time that these methods have been compared fairly under the same datasets with different simulators.Comment: 11 pages + 6 pages appendix.     Acute Low Alcohol Disrupts Hippocampus-Striatum Neural Correlate of Learning Strategy by Inhibition of PKA/CREB Pathway in Rats

    The hippocampus and striatum guide place-strategy and response-strategy learning, respectively, and they have dissociable roles in memory systems, which could compensate in case of temporary or permanent damage. Although acute alcohol (AA) treatment had been shown to have adverse effects on hippocampal function, whether it causes the functional compensation and the underlying mechanisms is unknown. In this study, rats treated with a low dose of AA avoided a hippocampus-dependent spatial strategy, instead preferring a striatum-dependent response strategy. Consistently, the learning-induced increase in hippocampal, but not striatal, pCREB was rendered less pronounced due to diminished activity of pPKA, but not pERK or pCaMKII. As rats approached the turn-decision area, Sp-cAMP, a PKA activator, was found to mitigate the inhibitory effect of AA on intra- and cross-structure synchronized neuronal oscillations, and rescue response-strategy bias and spatial learning deficits. Our study provides strong evidence of the critical link between neural couplings and strategy selection. Moreover, the PKA/CREB-signaling pathway is involved in the suppressive effect of AA on neural correlates of place-learning strategy. The novel important evidence provided here shows the functional couplings between the hippocampus and striatum in spatial learning processing and suggests possible avenues for therapeutic intervention

    Sparse data-extended fusion method for sea surface temperature prediction on the East China Sea.

    The accurate temperature background field plays a vital role in the numerical prediction of sea surface temperature (SST). At present, the SST background field is mainly derived from multi-source data fusion, including satellite SST data and in situ data from marine stations, buoys, and voluntary observing ships. The characteristics of satellite SST data are wide coverage but low accuracy, whereas the in situ data have high accuracy but sparse distribution. For obtaining a more accurate temperature background field and realizing the fusion of measured data with satellite data as much as possible, we propose a sparse data-extended fusion method to predict SST in this paper. By using this method, the actual observed sites and buoys data in the East China Sea area are fused with Advanced Very High Resolution Radiometer (AVHRR) Pathfinder Version 5.0 SST data. Furthermore, the temperature field in the study area were predicted by using Long Short-Term Memory (LSTM) and Gate Recurrent Unit (GRU) deep learning methods, respectively. Finally, we obtained the results by traditional prediction methods to verify them. The experimental results show that the method we proposed in this paper can obtain more accurate prediction results, and effectively compensate for the uncertainty caused by the parameterization of ocean dynamic process, the discrete method, and the error of initial conditions

    Genotype-phenotype analysis of three Chinese families with Jervell and Lange-Nielsen syndrome

    Long QT syndrome (LQTS) is characterized by QT prolongation, syncope and sudden death. This study aims to explore the causes, clinical manifestations and therapeutic outcomes of Jervell and Lange-Nielsen syndrome (JLNS), a rare form of LQTS with congenital sensorineural deafness, in Chinese individuals.Three JLNS kindreds from the Chinese National LQTS Registry were investigated. Mutational screening of KCNQ1 and KCNE1 genes was performed by polymerase chain reaction and direct DNA sequence analysis. LQTS phenotype and therapeutic outcomes were evaluated for all probands and family members.We identified 7 KCNQ1 mutations. c.1032_1117dup (p.Ser373TrpfsX10) and c.1319delT (p.Val440AlafsX26) were novel, causing JLNS in a 16-year-old boy with a QTc (QT interval corrected for heart rate) of 620 ms and recurrent syncope. c.605-2A>G and c.815G>A (p.Gly272Asp) caused JLNS in a 12-year-old girl and her 5-year-old brother, showing QTc of 590 to 600 ms and recurrent syncope. The fourth JLNS case, a 46-year-old man carrying c.1032G>A (p.Ala344Alasp) and c.569G>A (p.Arg190Gln) and with QTc of 460 ms, has been syncope-free since age 30. His 16-year-old daughter carries novel missense mutation c.574C>T (p.Arg192Cys) and c.1032G>A(p.Ala344Alasp) and displayed a severe phenotype of Romano-Ward syndrome (RWS) characterized by a QTc of 530 ms and recurrent syncope with normal hearing. Both the father and daughter also carried c.253G>A (p.Asp85Asn; rs1805128), a rare single nucleotide polymorphism (SNP) on KCNE1. Bizarre T waves were seen in 3/4 JLNS patients. Symptoms were improved and T wave abnormalities became less abnormal after appropriate treatment.This study broadens the mutation and phenotype spectrums of JLNS.     Mechanotransduction regulates inflammation responses of epicardial adipocytes in cardiovascular diseases

    Adipose tissue is a crucial regulator in maintaining cardiovascular homeostasis by secreting various bioactive products to mediate the physiological function of the cardiovascular system. Accumulating evidence shows that adipose tissue disorders contribute to several kinds of cardiovascular disease (CVD). Furthermore, the adipose tissue would present various biological effects depending on its tissue localization and metabolic statuses, deciding the individual cardiometabolic risk. Crosstalk between adipose and myocardial tissue is involved in the pathophysiological process of arrhythmogenic right ventricular cardiomyopathy (ARVC), cardiac fibrosis, heart failure, and myocardial infarction/atherosclerosis. The abnormal distribution of adipose tissue in the heart might yield direct and/or indirect effects on cardiac function. Moreover, mechanical transduction is critical for adipocytes in differentiation, proliferation, functional maturity, and homeostasis maintenance. Therefore, understanding the features of mechanotransduction pathways in the cellular ontogeny of adipose tissue is vital for underlining the development of adipocytes involved in cardiovascular disorders, which would preliminarily contribute positive implications on a novel therapeutic invention for cardiovascular diseases.
