    Development of Refractory Silicate-YSZ Dual Layer TBCs

    Development of advanced thermal barrier coatings (TBCs) is the most promising approach for increasing the efficiency and performance of gas turbine engines by enhancing the temperature capability of hot section metallic components. Spallation of the yttria-stabilized zirconia (YSZ) top coat, induced by the oxidation of the bond coat coupled with the thermal expansion mismatch strain, is considered to be the ultimate failure mode for current state-of-the-art TBCS. Enhanced oxidation resistance of TBCs can be achieved by reducing the oxygen conductance of TBCs below that of thermally grown oxide (TGO) alumina scale. One approach is incorporating an oxygen barrier having an oxygen conductance lower than that of alumina scale. Mullite, rare earth silicates and glass ceramics have been selected as potential candidates for the oxygen barrier.     Germline breast cancer susceptibility genes, tumor characteristics, and survival.

    BACKGROUND: Mutations in certain genes are known to increase breast cancer risk. We study the relevance of rare protein-truncating variants (PTVs) that may result in loss-of-function in breast cancer susceptibility genes on tumor characteristics and survival in 8852 breast cancer patients of Asian descent. METHODS: Gene panel sequencing was performed for 34 known or suspected breast cancer predisposition genes, of which nine genes (ATM, BRCA1, BRCA2, CHEK2, PALB2, BARD1, RAD51C, RAD51D, and TP53) were associated with breast cancer risk. Associations between PTV carriership in one or more genes and tumor characteristics were examined using multinomial logistic regression. Ten-year overall survival was estimated using Cox regression models in 6477 breast cancer patients after excluding older patients (≥75years) and stage 0 and IV disease. RESULTS: PTV9genes carriership (n = 690) was significantly associated (p < 0.001) with more aggressive tumor characteristics including high grade (poorly vs well-differentiated, odds ratio [95% confidence interval] 3.48 [2.35-5.17], moderately vs well-differentiated 2.33 [1.56-3.49]), as well as luminal B [HER-] and triple-negative subtypes (vs luminal A 2.15 [1.58-2.92] and 2.85 [2.17-3.73], respectively), adjusted for age at diagnosis, study, and ethnicity. Associations with grade and luminal B [HER2-] subtype remained significant after excluding BRCA1/2 carriers. PTV25genes carriership (n = 289, excluding carriers of the nine genes associated with breast cancer) was not associated with tumor characteristics. However, PTV25genes carriership, but not PTV9genes carriership, was suggested to be associated with worse 10-year overall survival (hazard ratio [CI] 1.63 [1.16-2.28]). CONCLUSIONS: PTV9genes carriership is associated with more aggressive tumors. Variants in other genes might be associated with the survival of breast cancer patients. The finding that PTV carriership is not just associated with higher breast cancer risk, but also more severe and fatal forms of the disease, suggests that genetic testing has the potential to provide additional health information and help healthy individuals make screening decisions

    Expression and Function of Androgen Receptor Coactivator p44/Mep50/WDR77 in Ovarian Cancer

    Hormones, including estrogen and progesterone, and their receptors play an important role in the development and progression of ovarian carcinoma. Androgen, its receptor and coactivators have also been implicated in these processes. p44/Mep50/WDR77 was identified as a subunit of the methylosome complex and lately characterized as a steroid receptor coactivator that enhances androgen receptor as well as estrogen receptor-mediated transcriptional activity in a ligand-dependent manner. We previously described distinct expression and function of p44 in prostate, testis, and breast cancers. In this report, we examined the expression and function of p44 in ovarian cancer. In contrast to findings in prostate and testicular cancer and similar to breast cancer, p44 shows strong cytoplasmic localization in morphologically normal ovarian surface and fallopian tube epithelia, while nuclear p44 is observed in invasive ovarian carcinoma. We observed that p44 can serve as a coactivator of both androgen receptor (AR) and estrogen receptor (ER) in ovarian cells. Further, overexpression of nuclear-localized p44 stimulates proliferation and invasion in ovarian cancer cells in the presence of estrogen or androgen. These findings strongly suggest that p44 plays a role in mediating the effects of hormones during ovarian tumorigenesis

    The utility of the YLS/CMI-SV for assessing youth offenders in Singapore.

    The Youth Level of Service/Case Management Inventory–Screening Version (YLS/CMI-SV) is designed to provide a preliminary estimate of the level of risk for antisocial behaviors as well as an indication of areas for intervention in youth offenders. This study examined the predictive validity of the YLS/CMI-SV for violent, nonviolent, and general recidivism in a sample of 3,264 youth offenders within a Singaporean context (Mfollow-up = 1,764.5 days; SDfollow-up = 521.5). Cox regression and Receiver Operating Characteristic analyses revealed that the YLS/CMI-SV is significantly predictive of general, violent, and nonviolent recidivism for the male youth offenders, but there were mixed results for the female youth offenders. Overall, these results indicated that the YLS/CMI-SV is a useful measure for assessing the levels of risk for male youth offenders, and more investigation is needed to determine the suitability of the YLS/CMI-SV for the female youth offenders. Its implications for clinical practice and policy are discussed

    Ergonomic Risk Assessment of Aluminum Form Workers&rsquo; Musculoskeletal Disorder at Construction Workstations Using Simulation

    This study analyzes an existing scenario of musculoskeletal disorder (MSD) associated with the ergonomic hazard of the aluminum formwork workstation and its workers. Aluminum form-workers have increasing evidence of MSDs from repetitive tasks such as the adjustment, alignment of pins, pulling, pushing, and installation of panels, because of the cumulative exposure to ergonomic risks. Existing research indicates that this is due to insufficient expertise, form-worker awareness, and a complex construction plan. Using the Tecnomatix process simulate, this study aims to identify awkward postures during the process of lifting, assembling, and installing formwork to quantify MSDs and assess the ergonomic risk of aluminum form-workers and provide simple solutions. This simulation method makes use of input data from a random sample of 92 participants retrieved from four construction sites. The Rapid Upper Limb Assessment (RULA), Ovako Working Analysis System (OWAS) scores, and Energy Expenditure Rate (EER) for three identified awkward cases were determined to be unsatisfactory, unsafe, and acceptable with suggested alternatives. The ergonomic scores correspond to various bodily stresses, allowing workers to better understand which body parts experience major stress when performing manual jobs. The suggested integrated preventive ergonomics system reduces MSDs and improves how people interact with their surroundings

    Analysis of Musculoskeletal Disorders and Muscle Stresses on Construction Workers’ Awkward Postures Using Simulation

    The negligence involved in musculoskeletal disorder (MSD) at construction sites results in high rates of muscle injuries. This paper presents findings identified by the MSD for each part of a worker&rsquo;s body, categorizing the awkward postures of each body part, estimating muscle stresses, and establishing the benchmark using anthropometry and hand force data. MSDs and their corresponding frequencies were identified by administering the Nordic Musculoskeletal Questionnaire (NMQ) survey, which solicits responses regarding construction workers&rsquo; awkward postures. Musculoskeletal stresses were estimated using three-dimensional static strength prediction program (3D SSPP) biomechanical software. The new benchmarks were established for existing preventive measures using the anthropometry and hand force data. Workers suffering from different body muscle pains in awkward postures may be predicted using the compression forces magnitude, strength capability, and body balance. The model was verified by comparing its outputs with the survey analysis results. The study is of value to practitioners because it provided a means to understand the contemporary scenario of MSD and to establish a practical benchmark based on the physical capability of workers. It is relevant to researchers because it digitally predicts MSD and facilitates experimentation with different dimensions, thereby contributing to construction productivity improvement.     GA-Based Optimization Method for Mobile Crane Repositioning Route Planning

    Mobile cranes have been used extensively as essential equipment at construction sites. The productivity improvement of the mobile crane affects the overall productivity of the construction project. Hence, various studies have been conducted regarding mobile crane operation planning. However, studies on solving RCP (the repositioning mobile crane problem) are insufficient. This article presents a mobile crane reposition route planning optimization method (RPOS) that minimizes the total operating time of mobile crane. It converts the construction site into a mathematical model, determines feasible locations of the mobile crane, and identifies near-global optimal solution (s) (i.e., the placement point sequences of mobile crane) by implementing genetic algorithm and dijkstra’s algorithm. The study is of value to practitioners because RPOS provides an easy-to-use computerized tool that reduces the lengthy computations relative to data processing and Genetic Algorithms (GAs).     Neighbourhood level real-time forecasting of dengue cases in tropical urban Singapore

    Abstract Background Dengue, a vector-borne infectious disease caused by the dengue virus, has spread through tropical and subtropical regions of the world. All four serotypes of dengue viruses are endemic in the equatorial city state of Singapore, and frequent localised outbreaks occur, sometimes leading to national epidemics. Vector control remains the primary and most effective measure for dengue control and prevention. The objective of this study is to develop a novel framework for producing a spatio-temporal dengue forecast at a neighbourhood level spatial resolution that can be routinely used by Singapore’s government agencies for planning of vector control for best efficiency. Methods The forecasting algorithm uses a mixture of purely spatial, purely temporal and spatio-temporal data to derive dynamic risk maps for dengue transmission. LASSO-based regression was used for the prediction models and separate sub-models were constructed for each forecast window. Data were divided into training and testing sets for out-of-sample validation. Neighbourhoods were categorised as high or low risk based on the forecast number of cases within the cell. The predictive accuracy of the categorisation was measured. Results Close concordance between the projections and the eventual incidence of dengue were observed. The average Matthew’s correlation coefficient for a classification of the upper risk decile (operational capacity) is similar to the predictive performance at the optimal 30% cut-off. The quality of the spatial predictive algorithm as a classifier shows areas under the curve at all forecast windows being above 0.75 and above 0.80 within the next month. Conclusions Spatially resolved forecasts of geographically structured diseases like dengue can be obtained at a neighbourhood level in highly urban environments at a precision that is suitable for guiding control efforts. The same method can be adapted to other urban and even rural areas, with appropriate adjustment to the grid size and shape
