    Anti-Inflammatory Activity of the Methanol Extract of Moutan Cortex in LPS-Activated Raw264.7 Cells

    Moutan Cortex (MCE) has been used in traditional medicine to remove heat from the blood, promote blood circulation and alleviate blood stasis. This study was conducted to evaluate the effects of MCE on regulatory mechanisms of cytokines and nitric oxide (NO) involved in immunological activity of Raw264.7 cells. Cells were pretreated with methanolic extracts of MCE, and further cultured for an appropriate time after lipopolyssacharide (LPS) addition. During the entire experimental period, 0.1 and 0.3 mg ml−1 of MCE had no cytotoxicity. In these concentrations, MCE inhibited the production of NO and prostaglandin E2 (PGE2), the expression of inducible NO synthase (iNOS), cyclooxygenase-2 (COX-2) and phosphorylated inhibitor of κBα (p-IκBα), and the activation of nuclear factor κB (NF-κB). MCE also reduced the concentration of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) and interleukin-6 (IL-6) in the Raw264.7 cells that were activated by LPS. These results demonstrate that MCE has anti-inflammatory effects through the inhibition of iNOS and COX-2 expression by suppressing the phosphorylation of I-κBα and the activation of NF-κB

    Higher Alpha/Theta Ratio May Indicate Decreased Brain Function in Older Adults during CSPFP10 Compared to Young Adults

    Declined physical ability and brain function observed in elder can affect daily living activities and negatively influence the quality of life (QOL). While the extensive research has explored the changes in cortical activity related to motor control in elder, less is known about how the brain functions during a physical function test. PURPOSE: To examine the electrocortical activation in the older adults’ brain during the Continuous Scale Physical Function Performance-10 (CSPFP-10) compared to young adults. METHODS: Twenty five older adults (OLD; 75.40±7.32 yrs, 70.33±18.23 kg, 162.87±7.55 cm) and twenty five young (YOUNG; 19.88±1.72 yrs, 66.52±14.22 kg, 167.50±10.22 cm) completed CSPFP10 while frontal (F3, Fz, F4), central (C3, Cz, C4), temporal (T7, T8, P7, P8), parietal (P3, Pz P4), and occipital (O1, Oz, O2) regions’ electrocortical activation was measured using a mobile electroencephalograph (EEG). Spectral power values (μV2/Hz) for theta (θ: 4-8 Hz) and alpha (α: 8-13 Hz) band were averaged across the target electrodes. Then α/θ ratio was calculated. Data were analyzed using independent t-test. RESULTS: OLD exhibited significantly higher α/θ ratio in frontal (M=0.98 vs.0.95, pp=.02), temporal (M=1.02 vs. 0.98, p=.006) compared to YOUNG. The separated t-test showed YOUNG had higher θ and α in frontal (θ: M= 40.93 vs. 37.56, pp=.03), central (θ: M=39.68 vs. 36.43, pp=.038), and parietal θ (M=40.55 vs. 37.98, p=.001) compared to OLD. Pearson’s correlation analysis did not find any significance between brain function and CSPFP10 scores in both groups. CONCLUSION α/θ ratio in OLD showed that their α power was relatively higher than YOUNG. Increased α power indicates suppression/selective cortical processing. It might indicate that OLD had inhibitory cortical activation process during the CSPFP10 task. Decreased frontal and parietal θ in OLD may indicate diminished decision making speed and attention with aging, while lower θ power in a central region (motor cortex) implies disrupt motor control. Overall, our result may indicate that brain activation patterns become less active during aging. It’s important to enhance both physical and brain function to improve activities of daily living and QOL in elderly population

    Cytoprotective Activity of Glycyrrhizae radix Extract Against Arsenite-induced Cytotoxicity

    Licorice, Glycyrrhizae radix, is one of the herbal medicines in East Asia that has been commonly used for treating various diseases, including stomach disorders. This study investigated the effect of licorice on arsenite (As)-induced cytotoxicity in H4IIE cells, a rat hepatocyte-derived cell line. Cell viability was significantly diminished in As-treated H4IIE cells in a time and concentration-dependent manner. Furthermore, results from flow cytometric assay and DNA laddering in H4IIE cells showed that As treatment induced apoptotic cell death by activating caspase-3. Licorice (0.1 and 1.0 mg ml−1) treatment significantly inhibited cell death and the activity of caspase-3 in response to As exposure. These results demonstrate that licorice induced a cytoprotective effect against As-induced cell death by inhibition of caspase-3

    A standardized extract of Rhynchosia volubilis Lour. exerts a protective effect on benzalkonium chloride-induced mouse dry eye model

    ETHNOPHARMACOLOGICAL RELEVANCE In contrast to other leguminous plants generally used as food, Rhynchosia volubilis Loureiro, a small soybean with a black seed coat, has been used as a traditional oriental remedy for various human diseases in Eastern Asia. In this study, we demonstrated the protective effect of R. volubilis against dry eye disease. AIM OF THE STUDY We aimed to investigate whether a standardized ethanol extract of R. volubilis (EERV) can protect the cornea in a benzalkonium chloride (BAC)-induced mouse dry eye model. MATERIALS AND METHODS Experimental dry eye was induced by the instillation of 0.2% BAC on mouse cornea. A standardized ethanol extract of R. volubilis (EERV) was orally administered following BAC treatment. The positive control group was treated with commercial eye drops. Fluorescein staining, tear break-up time (BUT), and hematoxylin and eosin staining were evaluated on the ocular surface. Squamous metaplasia and apoptosis in the corneal epithelial layer were detected by immunostaining. Furthermore, the protein expression of cytochrome c, Bcl-2, and Bax was determined. RESULTS EERV treatment significantly improved fluorescein scoring, BUT, and smoothness in the cornea compared to the vehicle group. In addition, EERV inhibited squamous metaplasia and apoptosis in the cornea. The expression of cytochrome c and Bax was upregulated, while that of Bcl-2 was downregulated in the vehicle group compared with that in the control group. However, EERV treatment inhibited the expression of cytochrome c and Bax, while that of Bcl-2 was improved. CONCLUSION Standardized EERV could be a beneficial candidate for the treatment of dry eye disease

    Anti-Inflammatory Effects of Bangpungtongsung-San, a Traditional Herbal Prescription

    Bangpungtongsung-san (BPTS), a traditional oriental herbal prescription, is widely used for expelling wind, draining heat, and providing general improvement to the immune system. In this study, we investigated the effects of BPTS on induction of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), proinflammatory cytokines, nuclear factor-kappa B (NF-κB), and mitogen-activated protein kinases (MAPKs) in lipopolysaccharide- (LPS- ) stimulated Raw 264.7 cells, and on paw edema in rats. At concentrations of 0.5, 0.75, and 1 mg/mL, treatment with BPTS inhibited levels of expression of LPS-induced NF-κB and MAPKs (ERK, JNK, and p38) as well as production of proinflammatory mediators, such as nitric oxide (NO), prostaglandin E2 (PGE2), tumor necrosis factor-α (TNF-α), and interleukin-6 (IL-6) by LPS. These results suggest that BPTS may exert anti-inflammatory effects via reduction of proinflammatory mediators, including NO, PGE2, TNF-α, and IL-6 through suppression of the signaling pathways of NF-κB and MAPKs in LPS-induced macrophages. In addition, using the carrageenan-induced paw edema assay, an antiedema effect of BPTS was observed in rats. These findings may provide scientific evidence validating the use of BPTS in treatment of patients with heat syndrome in Korean oriental medicine

    Features of Facial Asymmetry Following Incomplete Recovery from Facial Paralysis

    PURPOSE: The purpose of this study is to investigate peculiar patterns of facial asymmetry following incomplete recovery from facial paralysis that require optimal physical therapy for effective facial rehabilitation, and to decrease the incidence of avoidable facial sequelae. MATERIALS AND METHODS: This study involved 41 patients who had facial sequelae following the treatment of various facial nerve diseases from March 2000 to March 2007. All patients with a followup of at least 1 year after the onset of facial paralysis or hyperactive function of the facial nerve were evaluated with the global and regional House-Brackmann (HB) grading systems. The mean global HB scores and regional HB scores with standard deviations were calculated. Other factors were also analyzed. RESULTS: Four patterns of facial asymmetry can be observed in patients with incomplete facial recovery. The most frequently deteriorated facial movement is frontal wrinkling, followed by an open mouth, smile, or lip pucker in patients with sequelae following facial nerve injury. The most common type of synkinesis was unintended eye closure with an effort to smile. CONCLUSION: We described common configurations of facial asymmetry seen in incomplete recovery following facial nerve injury in an attempt to develop an optimal strategy for physical therapy for complete and effective facial recovery, and to decrease the incidence of avoidable sequelae.ope
