Carbon isotope ratioss of coccolith-associated polysaccharides of Emiliania huxleyi as a function of growth rate and CO2 concentration
The calcite plates, or coccoliths, of haptophyte algae including Emiliania huxleyi are formed in intracellular vesicles in association with waterâsoluble acidic polysaccharides. These coccolithâassociated polysaccharides (CAPs) are involved in regulating coccolith formation and have been recovered from sediment samples dating back to âŒ180âŻMa. Paired measurements of the carbon isotopic compositions of CAPs and coccolith calcite have been proposed as a novel paleoâpCO2 barometer, but additional proxy validation and development are still required. Here we present culture results quantifying carbon isotopic offsets between CAPs and other cellular components: bulk organic biomass, alkenones, and calcite. E. huxleyi was grown in nitrateâlimited chemostat experiments at growth rates (”) of 0.20â0.62/d and carbon dioxide concentrations of 10.7â17.6âŻÂ”mol/kg. We find that CAPs are isotopically enriched by 4.5â10.1â° relative to bulk organic carbon, exhibiting smaller isotopic offsets at faster growth rates and lower CO2 concentrations. This variability suggests that CAPs record a complementary signature of past growth conditions with different sensitivity than alkenones or coccolith calcite. By measuring the isotopic compositions of all three molecules and minerals of self-consistent origin, the ratio ”/[CO2(aq)] may be reconstructed with fewer assumptions than current approaches
Coverage of obesity treatment: A state-by-state analysis of Medicaid and state insurance laws
Objectives. We determined whether state Medicaid programs cover recommended treatments for adult and pediatric obesity and to what extent states regulate the treatment and coverage of obesity by private insurers.
Methods. We conducted a state-by-state document review of Medicaid manuals and private insurance laws and regulations.
Results. Eight state Medicaid programs appear to cover all recommended obesity treatment modalities for adults. Only 10 states appear to reimburse for obesity-related treatment in children. In the small-group insurance market, 35 states expressly allow obesity to be used for rate adjustments, while 10 states do so in the individual market. Two states expressly allow obesity to be used in eligibility decisions in the individual market. Five states provide for coverage of one or more treatments for obesity in both small-group and individual markets.
Conclusions. Very few states ensure coverage of recommended treatments for adult and pediatric obesity through Medicaid or private insurance. Most states allow obesity to be used to adjust rates in the small-group and individual markets and to deny coverage in the individual market
miR-23~27~24 clusters control effector T cell differentiation and function
Coordinated repression of gene expression by evolutionarily conserved microRNA (miRNA) clusters and paralogs ensures that miRNAs efficiently exert their biological impact. Combining both loss- and gain-of-function genetic approaches, we show that the miR-23~27~24 clusters regulate multiple aspects of T cell biology, particularly helper T (Th) 2 immunity. Low expression of this miRNA family confers proper effector T cell function at both physiological and pathological settings. Further studies in T cells with exaggerated regulation by individual members of the miR-23~27~24 clusters revealed that miR-24 and miR-27 collaboratively limit Th2 responses through targeting IL-4 and GATA3 in both direct and indirect manners. Intriguingly, although overexpression of the entire miR-23 cluster also negatively impacts other Th lineages, enforced expression of miR-24, in contrast to miR-23 and miR-27, actually promotes the differentiation of Th1, Th17, and induced regulatory T cells, implying that under certain conditions, miRNA families can fine tune the biological effects of their regulation by having individual members antagonize rather than cooperate with each other. Together, our results identify a miRNA family with important immunological roles and suggest that tight regulation of miR-23~27~24 clusters in T cells is required to maintain optimal effector function and to prevent aberrant immune responses
Dilatonic global strings
We examine the field equations of a self-gravitating global string in low
energy superstring gravity, allowing for an arbitrary coupling of the global
string to the dilaton. Massive and massless dilatons are considered. For the
massive dilaton the spacetime is similar to the recently discovered
non-singular time-dependent Einstein self-gravitating global string, but the
massless dilaton generically gives a singular spacetime, even allowing for
time-dependence. We also demonstrate a time-dependent non-singular
string/anti-string configuration, in which the string pair causes a
compactification of two of the spatial dimensions, albeit on a very large

Global embeddings of scalar-tensor theories in (2+1)-dimensions
We obtain (3+3)- or (3+2)-dimensional global flat embeddings of four
uncharged and charged scalar-tensor theories with the parameters B or L in the
(2+1)-dimensions, which are the non-trivially modified versions of the
Banados-Teitelboim-Zanelli (BTZ) black holes. The limiting cases B=0 or L=0
exactly are reduced to the Global Embedding Minkowski Space (GEMS) solution of

Magnetic Field Amplification in Galaxy Clusters and its Simulation
We review the present theoretical and numerical understanding of magnetic
field amplification in cosmic large-scale structure, on length scales of galaxy
clusters and beyond. Structure formation drives compression and turbulence,
which amplify tiny magnetic seed fields to the microGauss values that are
observed in the intracluster medium. This process is intimately connected to
the properties of turbulence and the microphysics of the intra-cluster medium.
Additional roles are played by merger induced shocks that sweep through the
intra-cluster medium and motions induced by sloshing cool cores. The accurate
simulation of magnetic field amplification in clusters still poses a serious
challenge for simulations of cosmological structure formation. We review the
current literature on cosmological simulations that include magnetic fields and
outline theoretical as well as numerical challenges.Comment: 60 pages, 19 Figure
Epigenome-Wide Analysis of DNA Methylation and Optimism in Women and Men
Objective: Higher optimism is associated with reduced mortality and a lower risk of age-related chronic diseases. DNA methylation (DNAm) may provide insight into mechanisms underlying these relationships. We hypothesized that DNAm would differ among older individuals who are more versus less optimistic. Methods: Using cross-sectional data from two population-based cohorts of women with diverse races/ethnicities (n = 3816) and men (only White, n = 667), we investigated the associations of optimism with epigenome-wide leukocyte DNAm. Random-effects meta-analyses were subsequently used to pool the individual results. Significantly differentially methylated cytosine-phosphate-guanines (CpGs) were identified by the ânumber of independent degrees of freedomâ approach: effective degrees of freedom correction using the number of principal components (PCs), explaining >95% of the variation of the DNAm data (PC-correction). We performed regional analyses using comb-p and pathway analyses using the Ingenuity Pathway Analysis software. Results: We found that essentially all CpGs (total probe N = 359,862) were homogeneous across sex and race/ethnicity in the DNAm-optimism association. In the single CpG site analyses based on homogeneous CpGs, we identified 13 significantly differentially methylated probes using PC-correction. We found four significantly differentially methylated regions and two significantly differentially methylated pathways. The annotated genes from the single CpG site and regional analyses are involved in psychiatric disorders, cardiovascular disease, cognitive impairment, and cancer. Identified pathways were related to cancer, and neurodevelopmental and neurodegenerative disorders.
