    The Effect of Site Trust on Trust in the Sources of Online Consumer Review and Trust in the Sources of Consumer Endorsement in Advertisement

    Consumer endorsements have along been used as an advertising strategy, and now, it is also easy to see consumer endorsements in online shopping sites. A positive Online Consumer Review (OCR) is a consumer endorsement in the web site. Although the sources of both OCR and consumer endorsement in advertisement (CEA) are typical consumers, trust in the source of OCR could be perceived differently from trust in the source of CEA. Trust in the information source ensures that consumers comfortably accept the endorsement. In e-commerce, how is a consumerā€™s judgment involving trust based on endorsements made by other consumers? This experimental study investigates whether trust in a web site is transferred to trust in the source of OCR and CEA. Moreover, it also tests which source credibility could be more influenced by site trust

    Information Overload and its Consequences in the Context of Online Consumer Reviews

    An online consumer review is the information including experiences, evaluations and opinions on products from the consumer perspective. An online consumer review plays two roles - informant and recommender. Considering two factors of review structure (the number of reviews and review type), this study analyzes the effect of online consumer reviews on consumersā€™ information processing depending on their levels of involvement. Generally, more positive reviews seem better from the perspective of online consumer reviews as recommenders. However, from the perspective of online consumer reviews as information providers, consumers may be confronted with too much information when a large number of reviews are offered, which results in information overload. We investigate when information overload occurs in the context of online consumer reviews, what strategies against the information overload consumers use depending on their levels of involvement, and how the product attitude and purchasing intention are changed. Our findings have implications for online sellers in terms of how to manage online consumer reviews contents

    From Marxan to management: ocean zoning with stakeholders for Tun Mustapha Park in Sabah, Malaysia

    Tun Mustapha Park, in Sabah, Malaysia, was gazetted in May 2016 and is the first multiple-use park in Malaysia where conservation, sustainable resource use and development co-occur within one management framework. We applied a systematic conservation planning tool, Marxan with Zones, and stakeholder consultation to design and revise the draft zoning plan. This process was facilitated by Sabah Parks, a government agency, and WWF-Malaysia, under the guidance of the Tun Mustapha Park steering committee and with support from the University of Queensland. Four conservation and fishing zones, including no-take areas, were developed, each with representation and replication targets for key marine habitats, and a range of socio-economic and community objectives. Here we report on how decision-support tools informed the reserve design process in three planning stages: prioritization, government review, and community consultation. Using marine habitat and species representation as a reporting metric, we describe how the zoning plan changed at each stage of the design process. We found that the changes made to the zoning plan by the government and stakeholders resulted in plans that compromised the achievement of conservation targets because no-take areas were moved away from villages and the coastline, where unique habitats are located. The design process highlights a number of lessons learned for future conservation zoning, which we believe will be useful as many other places embark on similar zoning processes on land and in the sea

    The trinity of ribosome-associated quality control and stress signaling for proteostasis and neuronal physiology

    Translating ribosomes accompany co-translational regulation of nascent polypeptide chains, including subcellular targeting, protein folding, and covalent modifications. Ribosome-associated quality control (RQC) is a co-translational surveillance mechanism triggered by ribosomal collisions, an indication of atypical translation. The ribosome-associated E3 ligase ZNF598 ubiquitinates small subunit proteins at the stalled ribosomes. A series of RQC factors are then recruited to dissociate and triage aberrant translation intermediates. Regulatory ribosomal stalling may occur on endogenous transcripts for quality gene expression, whereas ribosomal collisions are more globally induced by ribotoxic stressors such as translation inhibitors, ribotoxins, and UV radiation. The latter are sensed by ribosome-associated kinases GCN2 and ZAKa, activating integrated stress response (ISR) and ribotoxic stress response (RSR), respectively. Hierarchical crosstalks among RQC, ISR, and RSR pathways are readily detectable since the collided ribosome is their common substrate for activation. Given the strong implications of RQC factors in neuronal physiology and neurological disorders, the interplay between RQC and ribosome-associated stress signaling may sustain proteostasis, adaptively deterrnine cell fate, and contribute to neural pathogenesis. The elucidation of underlying molecular principles in relevant human diseases should thus provide unexplored therapeutic opportunities

    LSM12-EPAC1 defines a neuroprotective pathway that sustains the nucleocytoplasmic RAN gradient

    Nucleocytoplasmic transport (NCT) defects have been implicated in neurodegenerative diseases such as C9ORF72-associated amyotrophic lateral sclerosis and frontotemporal dementia (C9-ALS/FTD). Here, we identify a neuroprotective pathway of like-Sm protein 12 (LSM12) and exchange protein directly activated by cyclic AMP 1 (EPAC1) that sustains the nucleocytoplasmic RAN gradient and thereby suppresses NCT dysfunction by the C9ORF72-derived poly(glycine-arginine) protein. LSM12 depletion in human neuroblastoma cells aggravated poly(GR)-induced impairment of NCT and nuclear integrity while promoting the nuclear accumulation of poly(GR) granules. In fact, LSM12 posttranscriptionally up-regulated EPAC1 expression, whereas EPAC1 overexpression rescued the RAN gradient and NCT defects in LSM12-deleted cells. C9-ALS patient-derived neurons differentiated from induced pluripotent stem cells (C9-ALS iPSNs) displayed low expression of LSM12 and EPAC1. Lentiviral overexpression of LSM12 or EPAC1 indeed restored the RAN gradient, mitigated the pathogenic mislocalization of TDP-43, and suppressed caspase-3 activation for apoptosis in C9-ALS iPSNs. EPAC1 depletion biochemically dissociated RAN-importin beta 1 from the cytoplasmic nuclear pore complex, thereby dissipating the nucleocytoplasmic RAN gradient essential for NCT. These findings define the LSM12-EPAC1 pathway as an important suppressor of the NCT-related pathologies in C9-ALS/FTD

In this work, we describe the new features and major improvements in Membrane Builderthat allow users to robustly build realistic biological membrane systems, including (1) addition of new lipid types such as phosphoinositides, cardiolipin, sphingolipids, bacterial lipids, and ergosterol, yielding more than 180 lipid types, (2) enhanced building procedure for lipid packing around protein, (3) reliable algorithm to detect lipid tail penetration to ring structures and protein surface, (4) distance-based algorithm for faster initial ion displacement, (5) CHARMM inputs for P21 image transformation, and (6) NAMD equilibration and production inputs. The robustness of these new features is illustrated by building and simulating a membrane model of the polar and septal regions of E. coli membrane, which contains five lipid types: cardiolipin lipids with two types of acyl chains and phosphatidylethanolamine lipids with three types of acyl chains. It is our hope that CHARMM-GUI Membrane Builder becomes a useful tool for simulation studies to better understand the structure and dynamics of proteins and lipids in realistic biological membrane environments

    Dipterocarpus tuberculatus as a promising anti-obesity treatment in Lep knockout mice

    IntroductionThe therapeutic effects and mechanisms of Dipterocarpus tuberculatus (D. tuberculatus) extracts have been examined concerning inflammation, photoaging, and gastritis; however, their effect on obesity is still being investigated.MethodsWe administered a methanol extract of D. tuberculatus (MED) orally to Lep knockout (KO) mice for 4 weeks to investigate the therapeutic effects on obesity, weight gain, fat accumulation, lipid metabolism, inflammatory response, and Ī²-oxidation.ResultsIn Lep KO mice, MED significantly reduced weight gains, food intake, and total cholesterol and glyceride levels. Similar reductions in fat weights and adipocyte sizes were also observed. Furthermore, MED treatment reduced liver weight, lipid droplet numbers, the expressions of adipogenesis and lipogenesis-related genes, and the expressions of lipolysis regulators in liver tissues. Moreover, the iNOS-mediated COX-2 induction pathway, the inflammasome pathway, and inflammatory cytokine levels were reduced, but Ī²-oxidation was increased, in the livers of MED-treated Lep KO mice.ConclusionThe results of this study suggest that MED ameliorates obesity and has considerable potential as an anti-obesity treatment

    Stable carbon and oxygen isotope investigation of Pleistocene growing-season paleoclimate using tree-ring cellulose from the Missinaibi Formation, Adam Creek, James Bay Lowland, Canada

    The isotopic compositions of cellulose in tree rings provide a record of seasonal and annual climatic conditions such as precipitation and temperature. We analyzed stable isotopes of carbon (Ī“13Ccellulose) and oxygen (Ī“18Ocellulose) in early-wood and late-wood of five conifer sub-fossil wood samples from the Missinaibi Formation, which likely grew during the peak of Marine Isotope Substage (MIS) 5a (~82 kya) at Adam Creek, Ontario. There was no systematic difference between early-wood and late-wood for either Ī“13Ccellulose or Ī“18Ocellulose, suggesting similar climatic conditions throughout the growing season. A shift to lower Ī“13Ccellulose with growth age in most samples indicated rising water availability. The estimated value of Ī“18Oprecipitation was similar or slightly higher than modern Ī“18Oprecipitation. Hence, the area may have experienced similar to slightly warmer and/or less humid conditions than today, which is in agreement with the few other studies of MIS 5a climate in this region
